It will be remembered: Trump allies cross him in special election – POLITICO

Allies of Meadows and Paul insist they are not intending to undercut the ex-president by backing Edmonds and Hood and are simply voicing their support for candidates, rather than opposing a Trump-endorsed hopeful.

But the turn of events underscores the conundrum confronting Republicans. With Trump picking favorites in primaries and looking to shape the party ahead of a potential 2024 comeback bid, he is forcing other Republicans to decide whether its worth crossing him by endorsing rival candidates.

Organizations that endorse candidates against the presidents endorsement do so at their own peril and, like the Democrats, will fail, said former Trump campaign manager Corey Lewandowski, a prominent Carey supporter and an informal adviser. But it will be remembered.

Representatives for Trump and Paul declined to comment. Debbie Meadows, whose organization has backed conservative female candidates such as Reps. Lauren Boebert (R-Colo.) and Marjorie Taylor Greene (R-Ga.), said in a statement: Our job at Right Women PAC is to find the most conservative, pro-Trump women, even in fields with multiple pro-Trump candidates and that's what we did here. This is certainly not an 'us versus them.' I totally support President Trump and always have.

Those in the former presidents orbit have been surprised to see Meadows and Paul break for other candidates given their closeness to Trump. Trump lavished praise on Meadows during a June appearance in North Carolina, noting that during the 2016 presidential race she was a supporter of mine even before her husband had been. Trump earlier this year endorsed Pauls 2022 reelection bid, saying his golfing partner has done a fantastic job for our Country, and for the incredible people of Kentucky.

People familiar with Debbie Meadows group say it operates independently of Mark Meadows, who has yet to come out in support of any candidate in the race. They also point out that her endorsement of Edmonds came on June 4, four days before Trump announced his support for Carey.

Meadows Right Women PAC, however, has continued promoting Edmonds since Trump made his move, highlighting its support of her on social media platforms including Instagram and Telegram. Paul, meanwhile, announced his support for Hood in early July, several weeks after Trump came out for Carey.

It isnt the first time Paul has broken with the ex-president: During the 2020 election, he came out in support of Tennessee Republican Senate candidate Manny Sethi over the Trump-backed eventual winner, now-Sen. Bill Hagerty. The Paul-aligned Protect Freedom PAC also waged an unsuccessful effort last year to defeat Texas Rep. Kay Granger, who had Trumps support.

Some Trump aides have been particularly rankled by Meadows, complaining that she is lining up against the former president despite his past receptivity to candidates she has encouraged him to back. She recently helped arrange for him to meet with congressional candidate Heidi St. John of Washington State, even though some prominent Trump supporters have begun to gravitate toward a different Republican running against Rep. Jaime Herrera Beutler: Army veteran Joe Kent. (Herrera Beutler is one of the 10 House Republicans who voted to impeach Trump following the Jan. 6 Capitol riot.) The St. John meeting drew eyerolls from some in the ex-presidents inner circle.

Meadows was also instrumental in persuading Trump to endorse one of her personal friends, Lynda Bennett, in last years election to fill her husbands vacant North Carolina congressional seat. Bennett suffered a lopsided primary defeat to now-Rep. Madison Cawthorn, infuriating Trump, who until that point had frequently boasted about his perfect record endorsing winning primary candidates in 2020. The former president repeatedly griped at Mark Meadows over the loss, and he has continued to express regret in private for not endorsing Cawthorn, who has emerged as a staunch ally.

To some, Debbie Meadows support for Edmonds is only the latest sticking point.

I am surprised that the wife of Trumps former chief of staff is not supporting the Trump-endorsed candidate, Mike Carey. Loyalty seems like a very important character trait in the Trump orbit, said Brian Darling, a former official at the Heritage Foundation, a prominent conservative organization.

Darling, a former Paul aide, distinguished Pauls endorsement of Hood from Meadows support for Edmonds, noting that the Kentucky senator was narrowly focused on backing like-minded libertarian candidates.

Those close to Meadows, who has been a major force in her husbands political rise, insist that by encouraging Trump to support certain candidates, she isnt doing anything unusual. Trump is lobbied by an array of people of get behind Republican hopefuls, they point out.

Carey remains the strong favorite in the Ohio primary, those tracking the race say. A June poll conducted by Careys campaign found him far out ahead of the other 10 Republicans running. Carey has made Trumps endorsement the centerpiece of his campaign, running ads spotlighting the former president praising him as a wonderful man who hes known for a long time during a recent Ohio rally. Trump called the candidate onstage to speak during the event.

But Meadows and Pauls involvement could be a factor in the final stretch, those involved in the race say. According to media tracking figures, Carey has spent and reserved just $171,000 on the radio and TV airwaves, less than several of his opponents. By comparison, the Paul-aligned organization, Protect Freedom PAC, has reserved $216,000 in advertising time to boost Hood.

The Meadows-run group, Right Women PAC, has yet to invest in ads, though there is speculation that it may. One Republican in the state noted the super PAC recently received a $1 million-plus contribution from Ohio-based donor Brenda Frecka, a substantial gift that could conceivably be used to fund a last-minute, pro-Edmonds barrage. Frecka and her husband, film packaging company founder David Frecka, are among the largest donors to a super PAC aligned with the conservative House Freedom Caucus, which Mark Meadows chaired during his congressional tenure.

Neither Debbie Meadows nor David Frecka would comment when asked whether the contribution would be used to bolster Edmonds. Meadows also would not say what activities she was planning for the final weeks of the contest.

Carey is also confronting an avalanche of spending from former GOP Rep. Steve Stivers, who vacated the seat to take a job overseeing the Ohio Chamber of Commerce. Stivers has so far spent nearly $300,000 in remaining funds from his campaign account to buttress another candidate, state Rep. Jeff LaRe.

The pro-Trump cavalry, however, may be coming to Careys rescue. The Lewandowski-run Make America Great Again Action, the principal pro-Trump super PAC, is considering parachuting in to close the spending gap. It would represent the organizations first investment in an election.

Whoever comes out ahead in the Republican primary is expected to win the general election, given that Trump comfortably won the conservative-leaning district in 2020.

While Carey hasnt won the support of Meadows and Paul, he has earned endorsements from other staunch Trump allies, including New York Rep. Elise Stefanik and Citizens United, a group overseen by former Trump campaign adviser David Bossie. He also has the endorsement of a political action committee run by Ryan Zinke, who served as Trumps Interior secretary.

Carey brushed off the support Meadows and Paul were giving to his rivals by highlighting his backing from someone else.

I am proud, he said in a text message, to have President Trumps endorsement.

WASHINGTON, D.C. Today, Senators Rick Scott, Ron Johnson, Marsha Blackburn, Mike Lee, Ted Cruz, Rand Paul, Mike Braun, Cynthia Lummis and Bill Hagerty released a joint statement on the Democrats multi-trillion dollar partisan spending package:

Our nation is nearing $30 trillion in debt. There is a day of reckoning if we dont get our fiscal house in order. We appreciate our colleagues efforts to get a bipartisan deal done. But supporting this infrastructure deal enables the Democrats to pass their $3.5 trillion spending package. Joe Biden said it himself: These two issues are welded together.

Everyone agrees America needs to invest in infrastructure. But with our national debt growing out of control and Democrats dedicated to passing a 100% partisan blowout spending package totaling $3.5 trillion, we cannot support any additional deficit spending on Joe Bidens, Nancy Pelosis, Chuck Schumers and Bernie Sanders liberal wish list.

We can pay for needed infrastructure without incurring additional debt by instead repurposing previously appropriated but unspent funds. We urge our colleagues to support an alternative approach and recognize that supporting an infrastructure bill that authorizes new spending also enables the Democrats $3.5 trillion tax-and-spend budget. We must stop mortgaging our childrens and grandchildrens futures.

Sadly, the good people of Cuba were beaten down by the brutal hand of the Fidel Castro regime, which preached a myth of equality and prosperity under communism. Make no mistake: Equality and prosperity cannot thrive in the presence of communism.

Now, Ive been studying Cuba for a while.

My interest in Cuba began in 2014, after former President Barack Obama met with Ral Castro in 2014 in an effort to begin the process of normalizing relations between the two countries.

This was intriguing to me because it represented the first glimpse of a potential end to the U.S. embargo against Cuba an embargo that has now lasted 63 years.

Originally designed to bring Cuba to its knees and break its communist rule, it was and continues to be a massive failure. Communist rule has actually thrived in Cuba since the embargo was initiated in 1958.

Sadly, though, many politicians continue to support the embargo as a way to scare voters into believing that if the embargo were lifted, wed be rewarding a murderous communist regime.

Of course, this is complete nonsense.

If we can do business with China, Russia, and Saudi Arabia, theres no logical reason we cant do business with Cuba.

Congressman Ron Paul opined on this many years ago, saying

We talked to the Soviets. We talk to the Chinese. And we opened up trade, and we're not killing each other now. We fought with the Vietnamese for a long time. We finally gave up, started talking to them, and now we trade with them. I don't know why the Cuban people should be so intimidating. I think we're living in the dark ages when we can't even talk to the Cuban people. I think it's not 1962 anymore. And we don't have to use force and intimidation and overthrow of governments.

I couldnt agree more.

Ending the U.S. embargo against Cuba should actually be something fairly easy for both sides of the aisle to agree upon. Economically, politically, and socially, this really is a no-brainer.

Of course, that doesnt mean itll happen. But if it does, make no mistake: We will be very eager to capitalize on such an opportunity that will not only benefit early investors but the good people of Cuba too.

Wild and the Expansion Draft: What you need to know – BlueJackets.com

While there weren't any surprises on the Wild's expansion protected list on Sunday, it certainly has a different look than it would have a week ago.

Like it did four years ago before the Vegas Golden Knights entered the NHL, Minnesota chose to go the 7-3-1 route for it's list, protecting seven forwards, three defensemen and one goaltender ahead of the 2021 Seattle Kraken Expansion Draft on Wednesday.

Teams could have also protected eight skaters of any position and one goalie ... and a few clubs did indeed choose that route.

But 7-3-1 was always the expectation for the Wild, especially after the club cut ties with Ryan Suter and Zach Parise in contract buyouts on Tuesday.

All players with no-movement clauses in their contracts are required to be protected (no-trade clauses are not).

Both Suter and Parise had no-moves in their deals, so when they were bought out, that opened up an additional forward spot and an additional spot for another defenseman to be kept in the mix.

Because Jared Spurgeon and Jonas Brodin each have no-move clauses, each was always going to be protected. But with more flexibility after Tuesday's move, the Wild accomplished one of GM Bill Guerin's primary goals of expansion, which was to keep Matt Dumba.

Dumba, who turns 27 next weekend, is coming off a six-goal, 21-point campaign in 2020-21, but is also a crucial cog for the Minnesota power play. Just two seasons removed from scoring 12 goals in just 32 games before a season-ending injury short-circuited what looked to be a breakout campaign, the Wild hopes Dumba can regain that form next season.

With Brodin, Spurgeon and Dumba protected, Carson Soucy is the most prominent blueliner available for the Kraken. Soucy has been outstanding the last two seasons for Minnesota, scoring a combined eight goals and 31 points in 105 games, but posting a plus-38 during that stretch.

It seems likely that with the current makeup of the Wild's roster, Soucy could be in line for a boost in minutes if the Kraken decide to go a different direction with Minnesota's roster.

Guerin could also offer Seattle GM Ron Francis additional compensation to steer the Kraken away from Soucy, or any other player Guerin is intent on keeping.

One of those guys could be goaltender Kaapo Kahkonen. With the emergence of veteran Cam Talbot in his first season with the Wild, Kahkonen was the goaltender left unprotected.

He could be a valuable asset too, just 24 years old, and coming off a rookie season in which Kahkonen went 16-8-0 with a 2.88 goals-against average and a .902 save percentage, numbers that are a little inflated in large part to two late-season games against the St. Louis Blues.

Without those two games, Kahkonen's goals-against would have been 2.41, a number that was much more indicative of how the former AHL Goaltender of the Year played for a bulk of his first full season in the League. Minnesota likes his future and liked the tandem he and Talbot formed last season, their first together.

The Kraken have a number of quality veteran options available to them in the goaltending category, including Montreal's Carey Price, Dallas' Ben Bishop, Los Angeles' Jonathan Quick, Ottawa's Matt Murray, Pittsburgh's Casey DeSmith, Vancouver's Braden Holtby and Washington's Vitek Vanecek.

Up front, the Wild had just one no-movement clause (Mats Zuccarello), so it was able to protect several of its younger, middle-six forwards.

Last season's leading scorer, Calder Trophy winner Kirill Kaprizov, is ineligible to be selected by Seattle and did not need to be protected.

After Zuccarello, the Wild chose to protect Joel Eriksson Ek, Kevin Fiala, Marcus Foligno, Jordan Greenway, Ryan Hartman and Nico Sturm.

Eriksson Ek and Sturm are quality young centers the Wild really coveted, while Foligno, Greenway and Hartman are each versatile forwards who can move up and down the lineup.

Fiala is one of the club's best players, so keeping him protected was a no-brainer.

Among Wild forwards left unprotected are centerman Victor Rask, who was drafted by Francis in the second round of the 2011 NHL Draft, when he held the same position with the Carolina Hurricanes, as well as the recently re-signed Nick Bjugstad, who inked a one-year extension with his hometown team on July 5.

The NHL's roster and trade freeze also went into effect on Saturday, putting a halt to any offseason deals Guerin may be working with other clubs, a stoppage that will last until Thursday.

Clubs can still make deals with the Kraken to protect players, but those won't be announced until Wednesday night's draft, which will be televised on ESPN beginning at 7 p.m. CT. The Golden Knights are exempt from the Expansion Draft and will not lose a player.

If the Facebook ads that incumbent Republican senators are running are any guide, next years midterm elections are set to be a showdown about Big Tech, former President Donald Trump and Dr. Anthony Fauci, director of the National Institute of Allergy and Infectious Diseases, as well as immigration.

Republicans up for reelection next year are already running digital ads to raise money and grow their mailing lists, and while there are unknown issues and political controversies that will yet shape the 2022 midterms, the topics incumbents chose to run on now act as a preview of the races to come. For now, its heavy on culture war issues, the border and fierce loyalty to Trump.

Big Tech has censored both President Trump and Ron Johnson for sharing their perspectives, one ad reads. Johnson has also run ads asking supporters what they think about the job Biden is doing.

Kennedys ads call for supporters to chip in to fight Democrats socialist agenda and protect gun rights. Hes also run ads criticizing Fauci. Faucis flip flopped like a banked catfish answering questions about COVID, one ad reads.

The unvaccinated are perpetuating pandemic

On July 15, the number of patients infected with COVID-19 at Sarasota Memorial Hospital was 25, up from 5 just one monthago.

The seven-day average of positive COVID tests at the hospitalwas 7.5%, up from 1.6% last month.

State rates of COVID have also risen dramatically, almost entirely among the unvaccinated. Continuing this pandemicrequires unvaccinated persons.

Anyone has the right to experience the infection themselves, but these unvaccinated persons infect others before they even know they are ill and thus perpetuate the pandemic.

It is time to stop being nice to those who choose to infect others. Theunvaccinated are the reason we all continue to experience the economic, social and physical consequences of COVID.

If you want to rid our society of all the negative consequences of the past year, get your shot and work on your friends and family who are reluctant to get theirs.

Womens health not up to Manatee County

In regard to the Herald-Tribune Reader Question:I am writing on behalf of women throughout Manatee County who stand against the abortion clinic ban under consideration in Manatee County, similar to the Heartbeat Act signed into law in Texas in May (Manatee County to explore local abortion regulations, June 9).

There are numerous reasons why this should never come to pass in Manatee County, nor should it be up to the county commissioners to decide womens health choices and, in some cases, their fate.

There are no health clinics in Manatee County that offer abortions, thus the commissioners are wasting time and money on this useless effort. They should be focused on helping the community deal with the hyperaggressive Delta variant of COVID-19, family planning services, foster care services and nonpartisan, nontheologically based reproductive health clinics.

The commissionersshould be looking after our poverty-level and homeless citizens.

There are so many ways the county commissioners need to spend their time, particularly since a ban on local abortion clinics would have to be approved by the state Legislature, in addition to the County Commission.

Do not turn back womens rights by decades and decades. Move forward together, as a whole, Manatee.

Responding to the Herald-Tribunes question about Manatee County considering an abortion ban: Many a day I drive by Planned Parenthood only to see some self-righteous, holier-than-thou, misguided protesters (many of them men) standing outside with signs, their presence there designed to intimidate and harass women entering the building.

My first question for these clueless zealots is, "Why dont you put yourself to good use and get involved with clubs, institutions and schools that get to the root of the problem, educating youths, promoting contraceptives and making them more accessible, and teaching personal responsibility, instead of trying to shame women for a deeply personal, difficult decision.

The men could mentor hormonally drivenboys and young men, who are the missing half of the sin."

My next question to these protesters is, How many of the unwanted children have youadopted and are you willing to pay more taxes and vote for more social programs and burgeoning foster care that subsidize kids?"

Women in every county should be free to seek medical care nearby andtomake decisions about their own bodies, decisions that should be made onlybetween the woman and her doctor, not in a back alley in Manatee County.

Mechanism of alcoholic osteonecrosis of the femoral head | IJGM – Dove Medical Press

Introduction

Osteonecrosis of the femoral head (ONFH) is a disease in which the local death of osteocytes and bone marrow components is caused by venous congestion or impaired arterial blood supply or the femoral head rupture.1,2 ONFH usually occurs between 30 and 50 years old.1 It is reported that the risk factors of ONFH included corticosteroid use, trauma, alcohol consumption, coagulation abnormalities, hyperlipidemia, and smoking.1 Numerous epidemiological and multicenter studies have shown that most ONFH patients have a history of alcohol abuse. It has been reported that alcohol can destroy bone homeostasis by directly inhibiting the proliferation and differentiation of bone marrow mesenchymal stem cells (BMSCs).3 In particularly, alcohol significantly inhibited the proliferation and DNA synthesis of osteoprogenitor cells.4 Therefore, alcohol is a factor that cannot be ignored to cause osteonecrosis of the femoral head.5 At present, there are two main types of osteonecrosis: traumatic and non-traumatic.2 Alcohol-induced ONFH belongs to non-traumatic osteonecrosis.6 Epidemiological studies have reported that 2045% of ONFH patients are associated with alcohol consumption.1 Excessive drinking may lead to dyslipidemia, abnormal differentiation of BMSCs differentiation, and abnormal bone metabolism. In addition, alcohol has a significant dose effect on bone homeostasis.7 However, each individual have different susceptibility, which may be related to genetic predispositions.8 Some studied showed that MMP-8 and MMP-3 gene polymorphisms were related to the risk of alcohol-induced ONFH.9

RAB40C, also known as RARL/RASL8C, is located in 16p13.3. It is a member of the RAS oncogene family and encodes the protein RAB40C. The RAB family of small GTPases was regards as the cellular regulators of vesicular transport,10 and RAB proteins were the key regulators of eukaryotic biofilm transport in all eukaryotes.11 Studies have found that RAB40C was directly regulated by let-7a and played an important regulatory role in the biological role of gastric tumorigenesis.12 Aberrant methylation in RAB40C may be related to the pathogenesis of prostate cancer.13 Our previous work has proved that RAB40C is a novel lipid droplet-related RAB protein,11 which is one of the RAB proteins regulating lipid droplets (LDs) homeostasis.14 Lipid metabolism disorder is considered as the main factor of the pathogenesis of alcohol-induced ONFH.15 Thus, the occurrence of alcohol-induced ONFH may be relevant to RAB40C gene.

In order to verify this hypothesis, this study aimed to search for the association between RAB40C single nucleotide polymorphisms (SNPs) and alcohol-induced ONFH susceptibility, so as to provide guidance for the potential treatment and prevention of the disease.

The ethical approval of this study is in line with the ethical principles of the Helsinki declaration on human medical research. Our study has been approved by the ethics committee of Hong Hui hospital in Xi an, China, and all participants have signed informed consent before participating in the study.

All subjects were the Chinese Han population and included 201 patients and 201 controls. Participants underwent routine physical examinations including internal medicine, surgery, and specialized facial examinations. The diagnostic criteria for alcohol-induced ONFH are based on the clinical manifestations of hip, lumbar, and knee pain and mobility limitations. We further diagnosed the disease by MRI analysis and X-ray examination, such as deformities of the femoral head, hip stenosis, protuberances, or collapsed cartilage fractures.16 The patients were diagnosed with ONFH after using plain radiographs in stage II, III, and IV of the Ficat Classification systems. Stage I is characterized by no radiological abnormalities. Only some joints are stiff and painful, usually with limited joint movement. The symptoms were relieved after rest, and no positive results were found on X-ray films. Occasionally, uniform or spotty osteoporotic areas could be seen. The second stage is characterized by bone reconstruction on X-ray film, with sparse bone and diffuse bone, but no change in the shape of femoral head or joint space. A plain or CT scan of the femoral head shows osteosclerosis, focal osteoporosis, or cystic changes. Stage III is characterized by continuous fracture of subchondral trabeculae with obvious cystic changes and sclerotic margin around it. The femoral head is flattened due to subchondral fracture, mainly in the load-bearing area. Stage IV is characterized by progressive enlargement of subchondral osteonecrosis, further compression and destruction of the femoral head and acetabulum, with narrowing of joint space and typical changes of osteoarthritis.16 The enrolled patients had a history of drinking pure alcohol > 400 mL (320g/week, any alcoholic beverage) per week for six months or more. The inclusion criteria of the control group were as follows: (1) healthy, excluding asymptomatic ONFH (stage I) subjects; (2) age and BMI-matched Han Population in the control group; (3) no recent infection; (4) no other medical history; (5) no history of alcohol abuse.

In 1000 genome project (http://www.internationalgenome.org/), we selected RAB40C candidate SNP sites with allele frequency (MAF) over 5%. Three SNPs (rs4984677, rs62030917 and rs2269556) in RAB40C were finally identified in the case - control study. We isolated the genomic DNA from the whole blood sample using the Goldmag-mini Purification Kit (GoldMag Co. Ltd, Xian, China), and DNA concentration was measured using the NanoDrop 2000 (Thermo Scientific, Waltham, MA). Multiplexed SNP Mass EXTEND assay was designed by Agena MassARRAY Assay Design 4.0 software, and SNP genotyping was performed by Agena MassARRAY RS1000 (Agena, San Diego, CA, USA) according to the standard scheme.17 We also used Agena Typer 4.0 software to analyze and manage our data.18

We used Microsoft Excel (Microsoft, Redmond, WA) and SPSS Statistics (version 17.0, SPSS, Chicago, IL) to analyze the collected data. All the pvalues in the study were two - tailed, and p < 0.05 was statistically significant. The chi-square test was used to evaluate the deviation of Hardy-Weinberg equilibrium (HWE). Pearson Chi - square test or Fishers accurate test were also used to compare the allele frequency and genotype frequency of alcohol-induced ONFH patients with that of the control group. The association between polymorphisms in the RAB40C gene and the risk of alcohol-induced ONFH was calculated on the basis of logistic regression analysis. Four models (co-dominant, dominant, recessive, and log-additive) were established using PLINK version 1.07 software to assess the association between each sites and the alcohol-induced ONFH risk. The pairwise linkage disequilibrium (LD), haplotype construction, and genetic association of polymorphism loci were assessed using the Haploview software package (version 4.2).

This study contained 201 alcohol-induced ONFH patients with an average age of 42.68 12.875 years old and 201 healthy controls with an average age of 42.87 13.270 years old. The independent sample t test showed that no significant difference was between the case group and the control group. In the case group, there were 54 cases in stage I and II, 147 cases in stage III and IV. Moreover, 44 cases with unilateral lesions and 157 cases with bilateral lesions. The information of BMI, alcohol and tobacco use in both cases and controls were shown in the Table 1. Supplementary Table S1 listed the plasma lipoprotein and lipid levels between patients and controls, and the results of the table indicated that there was a significant difference in PLT levels between the two groups.

Supplementary Table S2 showed the primers were used for this study. We have successfully genotyped three SNPs of RAB40C gene, and the genotype frequency distribution of all SNPs in the control groups did not deviate from the HWE (p > 0.05). Table 2 showed the basic information of chromosome position, role, MAF (Minor allele frequency) of cases and controls and HWE p-value of the three SNPs located in RAB40C gene. In these three SNPs, the minor allele G of rs62030917 was significantly associated with an increased alcohol-induced ONFH risk (OR = 1.47, 95% CI = 1.072.02, p = 0.017).

Then, we used four genetic models (co-dominant, dominant, recessive, and log-additive models) to analyze the relationship between three SNPs and alcohol-induced ONFH risk (Table 3). The result indicated that carriers with G/A-G/G genotype in rs62030917 were more likely to have alcohol-induced ONFH risk compared with AA homozygous carriers (adjusted OR = 1.52, 95% CI=1.022.26, p = 0.039) in the dominant model. In the log-additive model, rs62030917 also increased the risk of alcohol-induced ONFH (adjusted OR = 1.42, 95% CI=1.051.93, p = 0.025).

In addition, we also analyzed the association between these loci and the risk of alcoholic osteonecrosis by age stratification and hip lesions stratification (Table 4). Our results suggested that rs62030917 increased the risk of alcohol-induced ONFH among people 42 years old shown in the allele model (OR = 1.62, 95% CI = 1.042.54, p = 0.033), the co-dominant model (OR = 2.99, 95% CI = 1.088.30, p = 0.035), the recessive model (OR = 2.77, 95% CI = 1.037.47, p = 0.044) and the log-additive model (OR = 1.53, 95% CI = 1.002.33, p = 0.048). In patients with unilateral lesions vs controls, rs62030917 conveyed a increasing risk of alcohol-induced ONFH in the allele model (OR = 2.01, 95% CI = 1.233.29, p = 0.005), the co-dominant model (OR = 2.20, 95% CI = 1.084.45, p = 0.036; OR = 3.20, 95% CI = 1.089.48, p = 0.029), the dominant model (OR = 2.37, 95% CI = 1.224.61, p = 0.011) and the log-additive model (OR = 1.90, 95% CI = 1.183.08, p = 0.009). Likewise, rs2269556 also increased the risk of alcohol-induced ONFH (patients with unilateral lesions vs controls) in the allele model (OR = 1.70, 95% CI = 1.072.71, p = 0.023), the co-dominant model (OR = 2.83, 95% CI = 1.137.10, p = 0.027) and the log-additive model (OR = 1.68, 95% CI = 1.062.67, p = 0.028).

Linkage disequilibrium (LD) blocks composed of rs62030917 and rs2269556 were found in unilateral lesions (Figure 1) and older than 42 years old groups (Figure 2), respectively. However, there are no statistically significant correlation between the risk of alcohol-induced ONFH and haplotype. Comparative analysis of plasma lipoprotein and lipid levels (TC, TG, HDL-C, LDL-C and PLT) in carrier with different genotypes of rs4984677, rs62030917 and rs2269556 was shown in Table 5. The results showed that carriers of AA, GA and GG genotypes in rs2269556 had LDL-C levels of 2.55 0.71 mmol/L, 2.78 0.83 mmol/L and 2.95 1.08 mmol/L, respectively, which were significantly different (p = 0.047). Among them, carriers of GG genotype had the highest LDL-C levels.

The pathogenesis of non-traumatic osteonecrosis is vascular injury, osteocyte death, or defective bone repair.19,20 Long term excessive drinking can lead to dyslipidemia and then induce ONFH.21,22 In recent years, people began to pay attention to the relationship between hereditary susceptibility and alcohol-induced ONFH.2325 Our study on the relationship RAB40C gene polymorphisms and alcohol-induced ONFH is the novel study.

The coding region of RAB40C contains 281 amino acids, which is longer than most small GTPases because it contains a unique SOCS box domain between the conserved GTPase domain and the pre-acylated C-terminal. Special SOCS box domain interacted with the elongated protein B/C and Cul5 modules, and binded with the ring finger proteins to form active ubiquitin ligases, which mediated a series of cellular processes.2628 Our previous work has proved that RAB40C is a novel LDs-related RAB protein.11 This is because there is a unique SOCS box domain of RAB40C, which is necessary for LDs cluster.14 As the proadipocytes differentiate into adipocytes, the expression of RAB40C increased. During the formation and maturation of LDs in adipocytes, RAB40C gradually accumulated to the surface of LDs. RAB40C knockout moderately reduced the size of LDs, suggesting that RAB40C is involved in the biological genetic process of LDs.29

Alcohol induces cell differentiation into adipocytes.30 With the increase of alcohol exposure time and concentration, the number of adipocytes was increased.31 In the alcohol treatment group, intracellular lipid deposition also occurred, which eventually leaded to the death of osteocytes.31,33 These effective findings suggested that alcohol can directly induce adipogenesis, reduce bone marrow mesenchymal osteogenesis, and produce intracellular lipid deposition leading to osteocyte death, which may be related to the occurrence of alcohol-induced ONFH.31 In view of the research, rs62030917 of RAB40C significantly increased the risk of alcohol-induced ONFH. In subjects with unilateral lesions, rs62030917 and rs2269556 increased the alcohol-induced ONFH risk. Rs62030917 increased the risk of alcohol-induced ONFH in people 42 years old.

To sum up, the expression of RAB40C gene increases the risk of alcohol-induced ONFH, which indicates that the gene polymorphisms of RAB40C may increase its risk of disease. There are limitations in the scope and quantity of sample selection in this study. In this study, patients and people from Northwest China were selected, which may be biased. In addition, the sample size of this study is small, and further verification of our results needs larger samples to support. Finally, some polymorphic loci with alcohol-induced ONFH have been screened out at the DNA level, and the relationship between polymorphisms and gene expression level has not yet been evaluated. Therefore, the relationship between polymorphisms and gene expression level needs to be evaluated at the RNA level and protein level. The study only provides a direction for alcohol-induced ONFH research.

Our study indicated that RAB40C gene polymorphism rs62030917 significantly increased the risk of alcohol-induced ONFH and it may be a risk locus. Our study provides a direction for the research on the mechanism of alcoholic osteonecrosis, but it still needs intensive study.

Casio Is Re-issuing the Futuristic Digital Watch Ripley Wore In Alien – Gizmodo

When people think of iconic timepieces used as movie props, the Seiko Giugiaro 7A28-7000 worn by Sigourney Weavers Ellen Ripley in James Camerons Aliens always comes to mind. But the character wore another vintage timepiece in Ridley Scotts original Alien too, which Casio is now re-issuing just like Seiko did.

It might not be immediately obvious to anyone watching Alien because the watch was doubled up to create the futuristic-looking timepieces worn by Ripley, Dallas, and other crew members aboard the Nostromo, but the films prop makers used a vintage chronograph called the Casio F-100 (although it wasnt vintage at the time of filming) which was the companys first watch to feature a plastic casea common feature of cheap but durable digital watches today.

Aside from the resin case, the watch also included advanced features for the late 70s including a highly accurate stopwatch and a light for the screen. But what makes it stand out are the four function buttons located on the face below the display which today would be typically located on the sides of the watch. It makes the F-100 look more advanced than it really was, and also gives the watch the unique charm of vintage electronics, which is probably why Casio is bringing it back given collectors ravenous appetites for everything retro.

Renamed the A100 series, the watch has been updated with a metallic case and strap and includes a gold option and one with an inverted LCD display. The front-facing buttons are still here, and perform the same functions including changing modes, starting, stopping, and resetting the chronograph, and activating the screen lighting. The Casio A100 also now includes an LED that flashes along with alarms or when a countdown timer completes.

Available next month for $90, the A100 will also come in a version featuring a limited edition gold Pac-Man motif, with a matching etched stainless steel back and patterned strap reminiscent of the retro games maze levels.
