Biotech start-up Voyager Therapeutics uses new gene therapy to attack diseases – Boston.com

A Cambridge biotechnology company launching Wednesday is taking aim at Parkinsons disease and ALS with a new gene therapy that deliberately infects patients with a virus.

The firm, Voyager Therapeutics, plans to use a class of viruses known as adeno-associated viruses as carriers to deliver vital proteins to the brain. Intentional infection may be counterintuitive, but the viruses used in the therapy are harmless to humans, making them ideal vehicles for moving proteins throughout the body, without troublesome side effects.

Boston venture capital firm Third Rock Ventures considered Voyagers research so promising that it invested $45 million to get the company off the ground, an unusually big bet on such an early stage life sciences firm.

Were just convinced that these viruses are going to be incredibly important delivery vehicles to different parts of the body and make a big difference in a lot of very serious disorders, said Third Rock cofounder Mark Levin, who will serve as Voyagers interim chief executive during the companys start-up phase.

The investment in Voyager marks Third Rocks latest foray into genetic medicine and the treatment of rare diseases. Bluebird bio of Cambridge, another gene therapy company in its portfolio, raised more than $100 million in an initial public stock offering last June. Bluebird is working on a treatment to slow the progression of a genetic brain disorder called childhood cerebral adrenoleukodystrophy, or CCALD.

In November, Third Rock joined two other venture firms in putting a combined $43 million behind a Cambridge start-up called Editas Medicine, which is developing a technique to edit faulty genes, such as those that cause Huntingtons disease and sickle cell anemia.

The investments reflect a broader belief among the scientific community that gene therapy could be the key to effectively treating some of the worlds most challenging disorders. Gene therapy techniques typically involve replacing a mutated gene with a healthy version or turning off a gene that causes disease.

Voyager plans to use adeno-associated viruses as carriers for both techniques. To treat Parkinsons, for instance, Voyager will use viruses to deliver a missing protein. For ALS, the viruses will help shut down a harmful protein.

Expecting gene therapy to produce cures for rare diseases might be unrealistic, Levin said, but the idea is to make a dramatic difference in patients lives.

ND Ag Department ex-staffer: Exit interviews 'a sham'

BISMARCK A former staffer involved in a controversy over what state Agriculture Commissioner Doug Goehring has called politically incorrect comments he made in her presence says her exit interviews with Goehring and a department director were a sham.

Katie Pinke resigned last March as the departments marketing and information division director.

Goehring and Ken Junkert, the departments administrative services director, jointly conducted a face-to-face exit interview with Pinke on March 7 and filled out her exit interview questionnaire based on her answers though Pinke said the completed questionnaires, which are now public record and were obtained by Forum News Service, dont accurately reflect her answers.

I said a lot about the communication and the problems that the commissioner was having, and theres nothing negative about the commissioner in the exit interview, Pinke, now working as a marketing consultant, said in a phone interview Thursday.

The questionnaires do mention lack of communication, leadership and mentoring as being among her reasons for leaving the department, but they dont mention Goehring specifically.

Pinke said a human resources representative wasnt in the room which Goehring confirmed and she didnt sign off on what was written on the questionnaire.

As a division director, Pinke herself conducted three exit interviews during her 14 months in the job, records show. She said none involved a human resources representative.

So, to me, those arent official, she said. They can be filled out after the employee has left.

Deputy Commissioner Tom Bodine, who oversees the departments day-to-day operations, said the department changed its exit interview process this past week, recognizing the flaw of not having a third party conduct the interview. He said a human resources liaison will now do the exit interviews and share the results with the employees supervisor and Bodine.

I wish that was a policy before, he said Thursday. We just really carried on what other administrations had done.

The Pulse: Tiger Couple wading into risky waters

When the husband of the Tiger Mom says you've entered a subject area that's "about as politically incorrect as we can get," that's saying something. I elicited that response after asking Jed Rubenfeld to analyze the success and relative stability of Jews vs. the decline of WASPs. Rubenfeld is the less-known half of the married duo that just brought us The Triple Package: How Three Unlikely Traits Explain the Rise and Fall of Cultural Groups in America.

Shame on Rubenfeld and Amy Chua for not including my own triple threat - Montenegrins, Italians, and descendants of the Austro-Hungarian Empire. Instead they lauded the success of Asians, Cubans, Jews, Indians, Nigerians, Mormons, Iranians, and Lebanese. The Yale Law School professors assert those eight groups distinguish themselves by leading lives dictated by their superiority complex, insecurity, and impulse control. (Each trait, they say, runs contrary to a core tenet of contemporary thinking.) They believe that while any person from any background can embody those traits, their research shows it is instilled more frequently by the groups they chart.

How else to explain why, in 2010, Nigerians were 0.7 percent of the black American population, but (in 2013) accounted for between 20 to 25 percent of the 120 black students at Harvard Business School? Or that, in 2010, the Wharton School placed 31 of its graduates at Goldman Sachs - exactly the same number as the much lesser-known and Mormon-affiliated Brigham Young University's Marriott School of Management?

"There are literally dozens of groups outperforming the national average," Rubenfeld told me. "We focused on the groups you mentioned for a couple of reasons: One, because they're the largest of the successful groups, but two, because their success is the most striking and the most quantifiable, and they change over time. This is something we really want to stress, that is, the successful groups in America. It's a dynamic thing. The traits remain the same that we focused on, but if we had looked 20 years ago, we would have found different groups at the top of the income ladder, and if we look 20 years from now, we will find different groups."

So what happens in the future in a world that New York Times columnist Thomas Friedman describes as "flat"? When groups intermarry and there are fewer differences among countries and cultures, don't the cultures that have been prospering lose the traits that make up the triple package?

Rubenfeld thought the question valid. He told me that Asian American kids famously scoring better on tests is "verifiable fact, not stereotype," but not permanent.

"This success tends to dissipate after a couple of generations. . . . Somehow those first-generation, second-generation immigrants, they're communicating something to their kids, they're putting a charge in them. They are communicating, through the values and the way they raise them, some kind of special drive. And we're saying, What if we could bottle that?"

One of the groups that have had a decline of fortune is white Anglo-Saxon Protestants (WASPs), who Chua told me were the original triple-package culture.

"But this is a very fluid theory, and that's what I think is so interesting," Chua said. "Look at the second half of our title. It's about the rise and fall of cultural groups because success breeds less insecurity."
