Celldex's Phase 1 study of CDX-1401 published in Science Translational Medicine

HAMPTON, NJ (April 16, 2014): Celldex Therapeutics, Inc. (NASDAQ: CLDX) announced today that final data from its Phase 1 study of CDX-1401 in solid tumors, including long-term patient follow-up, have been published in Science Translational Medicine (Vol 6 Issue 232). The data demonstrate robust antibody and T cell responses and evidence of clinical benefit in patients with very advanced cancers and suggest that CDX-1401 may predispose patients to better outcomes on subsequent therapy with checkpoint inhibitors. CDX-1401 is an off-the-shelf vaccine consisting of a fully human monoclonal antibody with specificity for the dendritic cell receptor DEC-205 linked to the NY-ESO-1 tumor antigen. The vaccine is designed to activate the patient's immune system against cancers that express the tumor marker NY-ESO-1. While the function of NY-ESO-1 continues to be explored, references in the literature suggest that its expression might reflect the acquisition of properties that cancers find useful, such as immortality, self-renewal, migratory ability and the capacity to invade.
The Phase 1 study of CDX-1401 is the first clinical study to demonstrate that an off-the-shelf vaccine that targets dendritic cells in vivo through DEC-205 can safely lead to robust humoral and cellular immunityovercoming a significant challenge in the development of protein based vaccines. Targeting protein antigens to the DEC-205 receptor on dendritic cells was pioneered by the late Ralph Steinman, MD, a member of Celldex's Scientific Advisory Board. Dr. Steinman received the 2011 Nobel Prize in Physiology or Medicine for his discovery of the dendritic cell and its role in adaptive immunity. This now-proven ability to target proteins, like NY-ESO-1, to dendritic cells to generate potent immune responses specific to these proteins represents a promising approach for the next generation of vaccines against pathogens and cancer.
"CDX-1401 offers a novel, well-tolerated and practical approach to generating protein specific immunity that can be readily combined with other treatment strategies to boost immunity against pathogens and tumors," said Dr. Madhav Dhodapkar, MBBS, Arthur H. and Isabel Bunker Professor of Medicine and Immunobiology, Chief of the Section of Hematology at the Department of Internal Medicine and Clinical Research Program Leader of the Hematology Program at Yale Cancer Center and lead author of the paper. "The preliminary findings in patients who received therapy with a checkpoint inhibitor following the vaccine provide further rationale for combination immunotherapy strategies, meriting further investigation."
Thomas Davis, MD, Senior Vice President and Chief Medical Officer of Celldex Therapeutics added, "CDX-1401 has overcome a significant historical challenge in the development of protein based vaccines by successfully targeting dendritic cells in vivo. It now sits at the forefront of a new generation of off-the-shelf dendritic cell targeted vaccines that we believe hold significant promise. Based on the results observed in this Phase 1 study, we expect CDX-1401 to enter at least two combination studies this year with both our own investigational therapies and external therapies in melanoma and other indications where we believe a dendritic cell vaccine regimen could play an important role." Initial results from the Phase 1 study of CDX-1401 were presented at the 2012 Society for Immunotherapy Annual Meeting. The manuscript published today expands upon this data and includes longer-term patient follow up.
CDX-1401 Phase 1 Study Overview and Results
The study was designed to assess the safety, immunogenicity and clinical activity of escalating doses of CDX-1401 with TLR agonists (resiquimod and/or Poly ICLC (HiltonolTM) in 45 patients with advanced malignancies refractory to all available therapies. CDX-1401 was well tolerated, with no dose limiting or grade 3 toxicities reported. The most frequently reported adverse events were administration site reaction, fatigue, nausea and chills. Treatment induced humoral and cellular immunity to NY-ESO-1 in patients with NY-ESO-1 expressing tumors across various dose levels and adjuvant combinations.
Significant anti-NY-ESO-1 titers occurred in 79% (33/42) of evaluable patients, with high titers (>1:10,000) in 52% and very high titers (>1:100,000) in 33% of patients. Similarly strong humoral immunity developed in each cohort and in patients with or without confirmed NY-ESO-1 expression in their tumor. Approximately 54% of patients with NY-ESO-1 positive tumors had anti-NY-ESO-1 titers at baseline and most increased after vaccination. NY-ESO-1-specific T cell responses were absent or low at baseline, but increased post-vaccination in 56% of evaluable patients, including both CD4 and/or CD8 T cell responses. Durability of the T cell response was demonstrated in two patients from whom samples from additional cycles of CDX-1401 treatment were available. In these patients, the induction of NY-ESO-1 specific T cells was maintained through three cycles (approximately seven months) of treatment.

In the backyard of a cottage here overlooking the water, two poles with metal slats shaped like ribcages jut out from the ground. They look indistinguishable from heat lamps or fancy light fixtures.
These are satellite dishes, but they arent for TV. Theyre meant for dispatching mindfiles, the memories, thoughts and feelings of people who wish to create digital copies of themselves and fling them into space with the belief that theyll eventually reach some benevolent alien species.

The beach house and the backyard and the memory satellites are managed by 31-year-old Gabriel Rothblatt, a pastor of Terasem, a new sort of religion seeking answers to very old kinds of questions, all with an abiding faith in the transformative power of technology.
Technology does feel and smell and look and act like a God.Beneath the cottage is a basement office where the mindfile operation is headquartered. Next door is an ashram, an airy glass building with walls that slide away to reveal a backyard home to a telescope for stargazing and a space to practice yoga. Tucked behind a shroud of greenery, most neighbors dont even know this house of worship exists.
The name Terasem comes from the Greek word for Earthseed, which is also the name for the futuristic religion found in the Octavia Butler sci-fi novel Parable of the Sower that helped inspire Gabriels parents, Bina and Martine Rothblatt, to start their new faith. Martine founded the successful satellite radio company Sirius XM in 1990. (Martine was originally known as Martin. She had sex reassignment surgery 20 years ago.)
Organized around four core tenetslife is purposeful, death is optional, God is technological and love is essentialTerasem is a transreligion, meaning that you dont have to give up being Christian or Jewish or Muslim to join. In fact, many believers embrace traditional positions held by mainstream religionsincluding the omnipotence of God and the existence of an afterlifebut say these are made possible by increasing advancements in science and technology.
Einstein said science without religion is lame. Religion without science is blind, Martine Rothblatt tells TIME. Bina and I were inspired to find a way for people to believe in God consistent with science and technology so people would have faith in the future.
Sure, its easy to dismiss people who think they can somehow cheat death with a laptop. But Terasem is a potent symbol of a modern way of life where the digital world and the emotional one have become increasingly entwined. It is also a sign, if one from the fringe, of the always evolving relationship between technology and faith. Survey after survey has shown the number of Americans calling themselves religious has declined despite the fact that many still identify as spiritual. People are searching, and no longer do they look to technology to provide mere order for their lives. They also want meaning. Maybe, its time to hack our souls.

IN the post 9/11 scenario in countries coping with terrorism, the police can no longer remain restricted to traditional preventive and detection roles. They must also act as combatants and protectors of human rights. Every police service requires a counterterrorism and human rights apparatus. In the developing world, human rights and policing often find themselves in conflict but in the civilised world policing and human rights are two sides of the same coin.
Policing without observing human rights is incomplete policing. Good governance requires the transformation of a coercive model of policing into a humane service. All police actions are closely linked with basic human rights, and therefore the image of the police is directly proportionate to their respect for human rights.
Accountability ensures professional policing. In times of yore, Control Yuan in China, the Tribuni Plebis in Rome, the ombudsman in Sweden and parliamentary commissions in England monitored the police.
Apart from apolitical civilian oversight, accountability requires more transparency and openness in policing. Prior to the Police Order 2002, we were policing without an institutional public safety mechanism. The police order introduced public safety commissions and a complaint authority but these bodies have yet to flex their muscles.
Unfortunately, numerous amendments were made regarding the composition and functions of public safety commissions in Pakistan. If such bodies are to be filled with those who were responsible for the current policing model, inhuman though it is, then what is the need for cosmetic arrangements?
The autonomy of such bodies will protect human rights. Funding them requires special attention. The allocation of public safety funds will improve human rights indicators. These bodies must be vested with investigative powers. Further, before assuming the membership of civilian oversight, the capacity issue of the members must be catered for.
Owing to ineffective accountability mechanisms in the police, human rights violations persist. The internal accountability of the police enjoys low public trust. Article 114 of the Order requires that the IGP draft a code of conduct. Upon contravention, the police officer can be removed or suspended from service. But despite the lapse of a decade a code of conduct is yet to be drafted and implemented by all provincial police departments.
According to Article 3 of the Police Order, every police officer is bound to behave with members of the public with due decorum and courtesy. Respect for human rights enhances the effectiveness of the police. But this protective role can only be attained by improving the work station environment, training and accountability.
Last year, human rights officers were appointed in 13 police stations of Islamabad. Such initiatives always proved cosmetic as they had no dedicated individuals and were devoid of institutional and monitoring mechanisms. Such individual-driven initiatives are often closed down with the departure of its exponents. The protection mechanism provided for in the Police Order 2002 must be implemented in letter and spirit.
The police subculture often defeats accountability. Supervisory tiers need to understand that accountability is their prime responsibility .The majority of even good officers overlook this aspect as they do not want to lose popularity with their peers. Further reduction of the discretionary powers of police officers will also improve human rights indicators.

Australia Post chief executive Ahmed Fahour is on tenterhooks as the May 13 budget looms and the Abbott government prepares to release the Commission of Audit's report on how to make the government leaner and more efficient.
The report and the government's response to it could be a game-changer for the business that Fahour runs. What he has described as a ''win-win'' would see Australia Post's role as provider of government services expanded significantly, cutting government service delivery costs and giving Australia Post new income to offset growing losses on its mail delivery service. Australia Post lifted net profit by 11 per cent to $312 million and paid the Commonwealth government a dividend of $244 million in the year to June 2013, but the profits all came from an unregulated part of its business, express delivery services notably.
Its regulated mail business lost $218 million, and Fahour told a Senate estimates committee hearing in February that the decline in letter volumes was accelerating, and likely to push the group onto losses in the June half this year for the first time since its corporatisation in 1989.
Australia Post is already a major agent for private sector companies and government agencies, processing payments and documents for more than 750 entities, including passport applications, land title transactions and licence renewals.

In a submission to the Commission of Audit, the group has argued, however, that its coverage could be expanded to include other government services and payments, including ones currently delivered by the Department of Human Services and its main retail ''face,'' Centrelink.
It would be radical expansion and one that would not be without execution difficulties. In appearances before the Senate estimates committee, Fahour was grilled by Labor senator Doug Cameron about Australia Post's qualifications to expand into areas that involve case management, for example.
The commission's report and the government's initial response to it will be released before the budget. The commission was asked to find ways to cut service delivery costs and end duplication, and the private sector provides a template.
Many large companies have already consolidated service functions that existed inside separate divisions into single ''shared services'' operations that each division accesses. Similar changes inside the government would be sweeping, and Australia Post is a potential services supercentre for the government because it operates Australia's largest retail network, with more than 4400 outlets, including more than 2500 in rural areas.

A futurist world under oppressive rule will be the theme when Zacada Circus School presents its annual show.
Circus Revolution will combine local students with professional performers to provide an amazing experience for both aspiring students and audiences. Among the performers will be Grace Cumming of Dundas.
Grace will perform a duo silks act. She started attending the circus school in September of last year.
I learn a lot from my team and coaches, and I have a lot of fun. Its really exciting, Grace told Hamilton Community News before last years production.
I like all the tricks, the wraps and the drops.
Zacada Circus School directors Kosta Zakharenko and Christine Cadeau are former Cirque du Soleil coaches and performers. Seven years ago, they opened Zacada Circus School.
Other acts featured in Circus Revolution are synchronized trapeze, juggling, aerial hoop, aerial silks, triple trapeze, break dance, skipping, aerial cube and hacky sack.
