DNA is one of the nucleic acids, information-containing molecules in the cell (ribonucleic acid, or RNA, is the other nucleic acid). DNA is found in the nucleus of every human cell. (See the sidebar at the bottom of the page for more about RNA and different types of cells). The information in DNA:
The key to all of these functions is found in the molecular structure of DNA, as described by Watson and Crick.
Although it may look complicated, the DNA in a cell is really just a pattern made up of four different parts called nucleotides. Imagine a set of blocks that has only four shapes, or an alphabet that has only four letters. DNA is a long string of these blocks or letters. Each nucleotide consists of a sugar (deoxyribose) bound on one side to a phosphate group and bound on the other side to a nitrogenous base.
There are two classes of nitrogen bases called purines (double-ringed structures) and pyrimidines (single-ringed structures). The four bases in DNA's alphabet are:
Watson and Crick discovered that DNA had two sides, or strands, and that these strands were twisted together like a twisted ladder -- the double helix. The sides of the ladder comprise the sugar-phosphate portions of adjacent nucleotides bonded together. The phosphate of one nucleotide is covalently bound (a bond in which one or more pairs of electrons are shared by two atoms) to the sugar of the next nucleotide. The hydrogen bonds between phosphates cause the DNA strand to twist. The nitrogenous bases point inward on the ladder and form pairs with bases on the other side, like rungs. Each base pair is formed from two complementary nucleotides (purine with pyrimidine) bound together by hydrogen bonds. The base pairs in DNA are adenine with thymine and cytosine with guanine.
In the next section we'll find out how long DNA strands fit inside a tiny cell.

DNA Identifies Jack the Ripper

A self-described "armchair detective" and a Finnish molecular biologist have claimed to have discovered the identity of the serial killer known as Jack the Ripper, who brutally murdered at least five prostitutes in the East End of London in the autumn of 1888.
A report in the Mail on Sunday names the killer as Aaron Kosminski, a Jewish Polish-born immigrant who lived in the East End and would have been 23 years old at the time of the murders. He had arrived in England in 1881 with his family, who were fleeing persecution by Russian authorities. He was later committed to an insane asylum, where he died of complications from gangrene.
Molecular biologist Dr. Jari Louhelainen tells the Mail on Sunday that he used a technique called "vacuuming" to remove DNA from a stained shawl purportedly belonging to one of the victims, Catherine Eddowes. Businessman Russell Edwards had bought the shawl at an auction in 2007, and had asked Louhelainen to help him find any clues that may be connected with the Ripper case.
The scientist said that infrared imaging revealed that the stains on the shawl were blood stains and were consistent with arterial blood splatter caused by slashing. Louhelainen claimed that the DNA from the blood stains found on the shawl was a match for Eddowes.
Other stains found on the shawl were florescent and had the characteristics of semen. Louhelainen said that a second set of DNA tests were carried out on the stains after cells from the epithelium -- a tissue that lines cavities and organs -- were discovered in the stains. Two strands of DNA were tested against a descendent of Kosminski's sister. The first was a 99.2 percent match, while the other strand was a perfect match. Louhelainen wrote that the DNA extracted from the shawl enabled him to specify Eddowes's killer as being of Russian Jewish ancestry with dark hair.
"Im excited and proud of what weve achieved," Louhelainen told the Mail, "and satisfied that we have established, as far as we possibly can, that Aaron Kosminski is the culprit."
However, scientists and scholars of the Ripper case have already cast doubt on Edwards and Louhelainen's specific claims, alleging that the evidence on the shawl is so compromised that any positive identification may be impossible.
"The shawl has been openly handled by loads of people and been touched, breathed on, spat upon," Richard Cobb, who runs Jack the Ripper conventions and tours, told The Times of London. "My DNA is probably on there. Whats more, Kosminski is likely to have frequented prostitutes in the East End of London. If I examined that shawl, Id probably find links to 150 other men from the area."
Sir Alec Jeffreys, who invented the technique of DNA fingerprinting, called the new research "An interesting but remarkable claim that needs to be subjected to peer review, with detailed analysis of the provenance of the shawl and the nature of the claimed DNA match with the perpetrator's descendants and its power of discrimination; no actual evidence has yet been provided."
However, Jeffreys also said that the lack of convincing evidence at a distance of 125 years does not rule out Kosminski as the prime suspect. "If I remember correctly when I visited the Black Museum at New Scotland Yard," Jeffreys told the Independent, "Kosminski was long regarded as by far the most likely perpetrator."

LONDON - An amateur detective claims to have used pioneering DNA technology to prove serial murderer Jack the Ripper was a Polish immigrant. The killer's identity is still a mystery some 126 years after he murdered at least five women and left their mutilated bodies in alleyways across East London. But author and amateur sleuth Russell Edwards says he used DNA from a scarf found near one of the murder scenes to unmask the Ripper as Aaron Kosminski. "I am a hundred percent certain," he told NBC News' U.K. partner ITV News. "Definitive proof, conclusively proven, put the case to bed - we've done this." His theory is detailed in his book "Naming Jack the Ripper," published Tuesday

The Newest Treatments for Psoriasis

People who suffer from psoriasis know how aggravating it can be, but treatments for the persistent skin condition are improving.
As we better understand the disease, researchers know more about what specific factors to target in order to develop effective treatments, Melinda L. McCord, M.D., a dermatologist at the Food and Drug Administration, told the FDAs Consumer Updates page.
Psoriasis is a condition in which skin becomes red and irritated, often developing thick silver flakes called scales, according to the National Library of Medicine. An estimated 7.5 million people in the United States have psoriasis. There is no cure for psoriasis, although symptoms can be managed with the right therapy.
Traditionally, treatment has been gradual, the FDA says, with patients starting with from creams and ointments all the way to phototherapy and systemic medicines. But now, the FDA says, doctors often try to optimize treatment i.e. start at a higher level right from the beginning. And treatments will become more tailored to the needs of each patient.
Tomorrows treatments will become even more personalized because the drugs in development now are targeting different aspects of the immune system, McCord noted. Psoriasis is an autoimmune disorder, meaning that the immune system mistakenly believes it is being attacked and then damages or inflames tissue that is actually healthy.
Therapies for psoriasis include topical treatments (i.e. those applied to the skin; light treatment (phototherapy) and medicines taken orally or via injection.
In the past, the FDA says, doctors treated psoriasis using a step-wise approach. Patients who had only mild to moderate psoriasis would start with topical treatments and, if necessary, move on to phototherapy or systemic drugs. People who had moderate to severe psoriasis were immediately treated with phototherapy or systemic drugs before being offered biologic therapies, a treatment that works with your immune system. But that strategy is changing, the FDA says. Today, patients can choose a treatment based on a number of conditions include lifestyle considerations, risk factors, and other diseases.
Most recently, the FDA approved a biologic product called Stelara (ustekinumab). This contains an antibody thats designed to bind to a specific target in the immune system. When given to patients, this antibody blocks the action of two proteins (interleukin 12 and 23) that contribute to the inflammation and the overproduction of skin cells, McCord said. By targeting these proteins, ustekinumab can interrupt the inflammatory pathway.
Researchers are also exploring medicines that target another protein, interleukin 17, McCord said. Looking forward, she added, the drugs in development are targeting different pathways in the immune system.
For the best treatment, McCord recommends a team approach in treating psoriasis, saying that different health-care providers need to work with patients to address the other diseases that might occur at the same time as psoriasis. These conditions can include lymphoma, heart disease and/or depression. We do not completely understand the relationship of theseto psoriasis, but it is an area of active research, she said.

UNC researchers find new genetic target for a different kind of cancer drug

CHAPEL HILL, NC Researchers from the UNC School of Medicine have discovered that the protein RBM4, a molecule crucial to the process of gene splicing, is drastically decreased in multiple forms of human cancer, including lung and breast cancers. The finding, published today in the journal Cancer Cell, offers a new route toward therapies that can thwart the altered genetic pathways that allow cancer cells to proliferate and spread.
"Historically, scientists haven't targeted the proteins in cancer cells that are involved in gene splicing," said Zefeng Wang, PhD, associate professor in the department of pharmacology and senior author of the Cancer Cell paper. "This is a whole new ballgame in terms of gene regulation in cancer."
There are approximately 25,000 genes in the human genome the same amount as in a fruit fly. But in humans, these genes are spliced together in different ways to create various kinds of messenger RNA to produce the many different proteins humans require. It's like a filmmaker splicing together bits of movie scenes to create alternative cuts of a movie. In genetics, this process is called alternative splicing.
Wang's lab found that RBM4 is an important film editor.
Wang, a member of the UNC Lineberger Comprehensive Cancer Center, studies how alternative splicing happens in normal cells and in cancer cells. Through a series of biochemical experiments and high-throughput screening methods, his team identified about 20 proteins that are involved in regulating alternative splicing. Then his team conducted further experiments to pinpoint changes in the activity of these proteins in various kinds of human cancer cells and in mouse models. Such "misregulated" protein expression would provide evidence that the proteins are involved in cancer development or metastasis.
Wang found that the protein RBM4 was decreased, as compared with normal tissue. In lung and breast cancer patients, RBM4 was drastically "down regulated."
"In normal cells, RBM4 inhibits alternative splicing," Wang says. "It makes genes splice from a long form into a short form. For one of the genes we study, which is called Bcl-x, we want the short form because it has anti-cancer properties."
