Damage to DNA is an issue for all cells, particularly in cancer, where the mechanisms that repair damage typically fail. The same agents that damage DNA also damage its sister molecule messenger RNA (mRNA), which ferries transcripts of the genes to the tens of thousands of ribosomes in each cell. But little attention has been paid to this damage.

"Everybody thought, 'Why care about the messenger RNA? These molecules have high turnover rates and are quickly degraded, so what does it matter if one is damaged?'" said Hani Zaher, PhD, assistant professor of biology in Arts & Sciences at Washington University in St. Louis.

"In organisms like E. coli or yeast, that's probably true," Zaher said. "You don't have to worry about mRNA because it turns over really fast. But in neurons you can't use that argument because an mRNA can persist, in some cases for days. And if that mRNA is really damaged it can become a big problem."

"There may be cases where messenger RNA is just as important as DNA," said Carrie Simms, PhD, a postdoctoral associate in Zaher's lab. "Clearly oxidative damage to RNA is somehow involved in neurodegenerative diseases, such as Alzheimer's and ALS. It's not necessarily causing the disease; it may just be some sort of byproduct; but it's in the mix."

"Under normal conditions only about 1 percent of the cellular mRNAs are oxidized," Zaher said, "but if you have oxidative stress, for whatever reason, a higher percentage can be damaged.

One of the hallmarks of Alzheimer's is oxidative stress, and studies have shown that in people with advanced Alzheimer's, half of the RNA molecules in the neurons may be oxidized.

In the November 13 issue of Cell Reports, Zaher, Simms and their colleagues report that when they fed oxidized mRNA to ribosomes, the nanomachines that convert mRNA to protein, the ribosomes jammed and stopped.

A stuck ribosome could be rescued by factors that released it from the mRNA and chewed up the damaged transcipt. But if the factors involved in this quality-control system were absent, damaged mRNA accumulated in the cell, just as it does in Alzheimer's.

A surprise

The three cellular processes essential to life -- making copies of DNA, copying DNA into mRNA, and translating the mRNA into protein -- have been penalized for billions of years by evolution, are astonishingly accurate, because evolution has heavily penalized any sloppiness.

DNA-based memory can record multiple inputs from engineered gene circuits

A new DNA-based recorder allows bioengineers to create cell cultures that detect information in their environment and store it for later use. Such 'designer' cells might in the future be used to monitor water quality in a village, or measure the amount of sugar a person eats. The technique is described this week inScience.

Insynthetic biology, genes are engineered to regulate each other's expression in such a way that they can perform logic operations similar to those in computer circuits. Memory storage has long been considered one of the key components needed to fulfil the promise of this technology.

Building gene circuits requires not only computation and logic, but a way to store that information, says bioengineer Timothy Lu of the Massachusetts Institute of Technology in Cambridge. DNA provides a very stable form of memory and will allow us to do more complex computing tasks.

In previous synthetic-biology attempts, data storage has been laborious to create. It also recorded only the presence or absence of one particular sensory input, and could be used only for limited applications. In the latest paper, Lu and his colleague Fahim Farzadfard describe how they can record many types of data simultaneously, and can register the accumulation of the input over time, like a cars odometer counts kilometers. The stored information can then be read out by sequencing the DNA. They dub their method SCRIBE, for Synthetic Cellular Recorders Integrating Biological Events.

Its a nice addition to the toolbox, which could complement other memory-storage techniques, says Jrme Bonnet, a bioengineer at the Centre for Structural Biochemistry in Montpellier, France, who was not involved in the research. Theres room for different types of memory in synthetic biology as in computing you have the hard drive and the RAM.

Living memory The team's work on SCRIBE began three years ago as an attempt to improve gene editing, in which cells are coaxed to incorporate new information into their genomes. One seemingly straightforward approach involves using single-stranded DNA molecules.

Bacterial genomes, like human genomes, are made up of double-stranded DNA molecules. But when single-stranded DNA is floating around in the cell, it is possible to stimulate the bacterium to insert it into its genome, using an enzyme from a virus. However, most bacteria do not readily manufacture a significant number of single-stranded DNAs.

This, critics suggest, was the very same evidence that they had failed to produce in his first trial in 2007: DNA.

Critics, not least the late Margo Macdonald MSP, said prosecutors should have told the jury that Sinclair's DNA had been found on tights used to strangle Ms Eadie.

The Crown countered that the DNA was "low copy and low probability". Seven years on and it was DNA on ligatures used to kill the women, including their own underwear, that proved decisive in both securing the new trial and convicting Sinclair.

Forensic scientists, using state-of-the-art white-light technology developed since 2007, were able to identify spots of DNA on items of clothing stored for more than three decades. They included a full profile of Sinclair, trapped in the knots of one of the victim's bras used as a ligature.

In crucial evidence during the trial Geraldine Davidson, a forensic scientist, said the genetic material recovered was 20 million times more likely the DNA came from a combination of Sinclair and his brother-in-law Gordon Hamilton than from either of them acting alone or from neither of them.

Ms Davidson said there had been particular interest in the knotted areas of the tights and bra used as ligatures on Christine, as the knots would "preserve" the DNA present at the time they were tied. She added: "This fits with the DNA within the preserved areas having been present at the time the ligatures were tied."

In the case of Christine's bra, recovered with part of her tights from around her head and neck, Ms Davidson said specialist analysis had been carried out.

This confirmed Hamilton had been the major contributor of DNA within the knots.

But although Sinclair had been the minor contributor in terms of the amount of material left on the ligature, he was "fully represented". She added: "It's a full profile. Some of the components of his DNA are shared with the other three reference samples [Hamilton and both victims], but every one of the components of Angus Sinclair's profile are represented in the profile."
