Danielle Sheehan has suffered eczema since she was a baby It disappeared during her childhood but returned when she was about 20 When she was 20 the patches returned on around her eyes and on arms She tried everything doctors suggested including creams, tablets, lotions, other natural remedies and light therapy but nothing worked 18 months ago she had Botox for the first time for cosmetic reasons Around five days later she noticed her eczema had suddenly cleared up

Since she was a baby, Danielle Sheehan has suffered painful bouts of eczema.
As a child the debilitating condition subsided. But four years ago red patches returned on her arms and around her eyes.
The model and dancer's self-esteem crumbled. She stopped booking photo shoots at work and refused to see her boyfriend during flare ups.
The eczema around her eyes was aggravated by moisture, becoming particularly painful when she cried.

Celgene gets positive CHMP opinion for OTEZLA to treat psoriasis and psoriatic arthritis

Celgene International Srl, a wholly-owned subsidiary of Celgene Corporation, announced that the European Medicines Agency's (EMA) Committee for Medicinal Products for Human Use (CHMP) has adopted a positive opinion for OTEZLA (apremilast), the Company's oral selective inhibitor of phosphodiesterase 4 (PDE4), in two therapeutic indications:
Psoriasis is an immune mediated skin condition characterised by raised scaly lesions on the skin. It affects approximately 14 million people across Europe and about 125 million people worldwide.
Plaque psoriasis, also called psoriasis vulgaris, is the most common form of the disease, representing about 80 percent of cases. Up to 30 percent of people with psoriasis may develop psoriatic arthritis, which involves pain and swelling in jointsand other manifestations and may lead to significant disability.
"This CHMP positive opinion is an important step forward for people with psoriasis and psoriatic arthritis in Europe. These immune mediated diseases are frequently debilitating and cause severe physical and emotional pain to the individual," stated Tuomo Ptsi, President, Celgene Europe, the Middle East and Africa (EMEA).
"We are proud to have moved one step closer to offering patients OTEZLA, a new, oral treatment approach that could significantly help control their symptoms and make a considerable difference to their quality of life."
In the ESTEEM studies, which form the basis of CHMP's positive opinion for apremilast in psoriasis, treatment resulted in significant and clinically meaningful improvements in plaque psoriasis as measured by PASI-75 (a 75 percent improvement in the Psoriasis Area Severity Index) scores at week 16, the primary endpoint.
Patients on apremilast also benefited from significant improvements in difficult to treat areas, such as nail and scalp, and itch, known to have a marked impact on patients' quality of life and perception of disease severity.
In the PALACE program, which forms the basis for CHMP's positive opinion for apremilast in psoriatic arthritis, treatment resulted in significant and clinically meaningful improvements in the signs and symptoms of psoriatic arthritis, as measured by the modified ACR-20 (a 20 percent improvement in the American College of Rheumatology disease activity criteria) response at 16 weeks, the primary endpoint.
Patients on apremilast showed improvement across multiple disease manifestations specific to psoriatic arthritis, such as swollen and tender joints, as well as dactylitis, enthesitis and overall physical function.

Novartis' Psoriasis Drug Cosentyx Backed for EU Approval – Analyst Blog

Novartis AG ( NVS ) announced that the European Medicines Agency's Committee for Medicinal Products for Human Use (CHMP) has provided a positive opinion on the approval of Cosentyx (secukinumab, formerly known as AIN457). Cosentyx is the first IL-17A inhibitor to be recommended as a first-line treatment option for psoriasis patients who require systemic therapy in Europe.
Cosentyx works by inhibiting IL-17A, a protein that is found in high concentrations in skin affected by the disease.
The CHMP's favorable opinion comes shortly after Cosentyx received a unanimous recommendation from the FDA's Dermatologic and Ophthalmic Drugs Advisory Committee (DODAC) last month -- a final decision from the FDA should be out in early 2015. The favorable opinion of the CHMP will now be reviewed by the European Commission with a final decision expected in the next couple of months.
With the CHMP in favor of approving Cosentyx, we believe chances of approval are high. Once approved, Cosentyx will be entering an extremely crowded psoriasis market given the presence of products like Stelara, Enbrel and Otezla.
Moreover, new treatments are currently in development including Amgen ( AMGN )/AstraZeneca's ( AZN ) brodalumab.
However, with Cosentyx consistently demonstrating very high skin clearance, including superiority to Enbrel in the head-to-head FIXTURE study, and given the high rate of dissatisfaction among patients regarding existing treatment options, Cosentyx could rapidly gain share once approved.
Meanwhile, Novartis is evaluating Cosentyx for several other indications including ankylosing spondylitis and psoriatic arthritis among others.
Novartis currently carries a Zacks Rank #3 (Hold). A better-ranked stock in the health care sector is Allergan Inc. ( AGN ) carrying a Zacks Rank #1 (Strong Buy).

There has been much recent debate on the benefits and risks of screening for breast cancer using BRCA1 and BRCA2 mutations in the general adult population. With an estimated 235,000 new breast cancer diagnoses each year in the U.S. and more than 40,000 deaths, it is clearly important to be able to determine which women may be genetically predisposed to breast cancer.
Glenn E. Palomaki, PhD, associate director of the Division of Medical Screening and Special Testing in the Department of Pathology and Laboratory Medicine at Women & Infants Hospital of Rhode Island has recently published an invited commentary in the November issue of Genetics in Medicine. The commentary is entitled "Is it time for BRCA1/2 mutation screening in the general adult population? Impact of population characteristics."
A family history of breast or ovarian cancer or a personal history of early-onset cancer are strong risk factors for breast cancer. Systematic criteria when caring for a patient with a positive family history have been well established by such agencies as the U.S. Preventive Services Task Force and the National Comprehensive Cancer Network.
Dr. Palomaki said, "With the identification of the tumor suppressor genes BRCA1 and BRCA2 in the 1990s, the scientific community has extensively explored both the personal and population impact of carrying a deleterious mutation in these genes. Any new population-based screening test, such as testing for BRCA1 and BRCA2 mutations, requires consideration of key performance characteristics that evaluate both strengths and shortcomings before its introduction."
In his commentary, Dr. Palomaki cited two recent publications that present perspectives on routine, population-based screening for breast cancer using BRCA1/2 mutations in different populations.
"Together, these two publications offer an unusual opportunity to compare and contrast how distinct population differences, such as the mutations carrier rate, might influence the feasibility of population-based screening," said Dr. Palomaki. "Because founder mutations are more common in Ashkenazi Jewish women, are more easily identified and account for a higher proportion of all breast cancer cases, pilot trials in that population are indicated before launching widespread screening in Israel to identify and resolve implementation issues. Such screening in the United States, however, is more complicated, tilting the balance away from routine population screening, as least for the moment."

After mining the genetic records of thousands of breast cancer patients, researchers from the Johns Hopkins Kimmel Cancer Center have identified a gene whose presence may explain why some breast cancers are resistant to tamoxifen, a widely used hormone treatment generally used after surgery, radiation and other chemotherapy.
The gene, called MACROD2, might also be useful in screening for some aggressive forms of breast cancers, and, someday, offering a new target for therapy, says Ben Ho Park, M.D., Ph.D., an associate professor of oncology in the Kimmel Cancer Center's Breast Cancer Program and a member of the research team.
The drug tamoxifen is used to treat estrogen receptor-positive breast cancers. Cells in this type of breast cancer produce protein receptors in their nuclei which bind to and grow in response to the hormone estrogen. Tamoxifen generally blocks the binding process of the estrogen-receptor, but some estrogen receptor-positive cancers are resistant or become resistant to tamoxifen therapy, finding ways to elude its effects. MACROD2 appears to code for a biological path to tamoxifen resistance by diverting the drug from its customary blocking process to a different way of latching onto breast cancer cell receptors, causing cancer cell growth rather than suppression, according to a report by Park and his colleagues published online Nov. 24 in the Proceedings of the National Academy of Sciences.
Specifically, the team's experiments found that when the gene is overexpressed in breast cancer cells--producing more of its protein product than normal--the cells become resistant to tamoxifen.
One piece of evidence for the gene's impact was demonstrated when the Johns Hopkins scientists blocked MACROD2's impact in breast cancer cell cultures by using an RNA molecule that binds to the gene to "silence," or turn off, the gene's expression. But the technique only partially restored the cells' sensitivity to tamoxifen.
To conduct the study, the scientists examined two well-known databases of breast cancer patients' genetic information, The Cancer Genome Atlas and the Molecular Taxonomy of Breast Cancer International Consortium study. Patients who had MACROD2 overexpressed in primary breast cancers at the original breast cancer site had significantly worse survival rates than those who did not, according to an analysis of the patient databases.
With this in mind, the Johns Hopkins scientists suggest that clinicians may be able to look at MACROD2 activity to help them identify aggressive breast cancers at early stages of growth.

New Zealand Security Intelligence Service director Rebecca Kitteridge says she thinks the public can "absolutely have faith" in the SIS being politically neutral.
"The people who work here are absolutely committed to compliance," Ms Kitteridge told ONE News.
Her comments come after her agency was slammed by Inspector-General of Intelligence and Security, Cheryl Gwyn, for its release of "incomplete, inaccurate and misleading" information to John Key's office about a 2011 meeting Mr Goff had with then SIS director Warren Tucker about alleged Israeli spying in Christchurch.
Ms Gwyn said she found "significant failures by the SIS to meet its obligations, both in the release of information and in upholding political neutrality".
"Overall there was a basic failure to provide a full and accurate picture," Ms Gwyn said.
Mr Tucker was found to have made a number of serious errors of judgement which were the principal reason for his staff providing incorrect and incomplete information.
The report goes on to say Dr Tucker also failed to maintain a relationship of trust and confidence with Mr Goff and failed to safeguard the political neutrality of the SIS.
Labour leader Andrew Little says what the report reveals is "a politicisation of one of our most important state agencies".
However, Ms Kitteridge said: "I don't see that as being a widespread or kind of systemic issue."

'Horrible Bosses 2' has horribly funny moments, but too few

In the sexually inappropriate and politically incorrect "Horrible Bosses 2," the bumbling workplace underdogs played by Jason Bateman, Jason Sudeikis and Charlie Day are about to try their hand at being in charge. And at times, they are horribly funny.
Being the boss makes for a lot of incredibly idiotic, highly amusing running amok, especially with a nasty new nemesis in Chris Pine, whose eyebrows and sense of entitlement are completely out of control.
Make no mistake, despite some well-earned laughs, "Horrible Bosses 2" is not what qualifies as a good movie or even a particularly good R-rated comedy. But there is more to laugh at in "2" than the first, so let's go with less horrible, shall we?
Directed by Sean Anders, who wrote the screenplay with creative partner John Morris, what "2" does better is play to its sweet spot not the bosses but the beaten-down schlubs beneath them. At least any offense coming from that quarter isn't intentional. They're not dumb as in "Dumb and Dumber," but they're not too bright either.
Nick (Bateman), Kurt (Sudeikis) and Dale (Day) are, however, a much more cohesive comic team this time. Their annoying silly sallies are much snappier. The chemistry between them is good enough to almost make them a modern-day Larry, Curly and Moe albeit more intelligent, more prurient and more attractive.
Kevin Spacey is back as horrible boss Dave Harken, best remembered for his scathing tirades, but he's behind bars and better now that his screaming comes in smaller doses.
Jennifer Aniston reprises Dr. Julia Harris, the R-rated dentist who specializes in sexual assaulting. There's still far too much of her porn dominatrix. Not a wise choice, since the naughty-doc bits never pay off. I'm guessing the implied hospital rape of a comatose male won't draw many laughs, even from the guys who come to see Aniston in a little leather and lace.
Completely inappropriate references are not limited to naughty doc. It must be in the "Horrible" articles of incorporation. As they did in the first, the filmmakers bet heavily on innuendo and politically incorrectness to carry the film when the plotting fails, starting with the opening scenes.
The guys are being featured on a local morning TV show for their new invention it involves a shower and shampoo. They kick things off with what sounds like a racial slur and move right into a demonstration that makes it look as if Dale is giving Kurt a "hand" in the shower.
The riffs are laughable, and I don't mean that as praise. They do, however, serve to lower expectations (assuming they weren't at rock bottom going in), which makes some of what follows seem almost brilliant in comparison.
