Mouse Model of Human Disease Still Good, but Significant Differences Exist

Newswise Scientists at Penn State College of Medicine, working alongside an international team of researchers, have produced the most complete encyclopedia of functional elements in the mouse genome to date and compared it to the human genome. The findings, published recently in Nature, uphold the mouse model of human disease, but pinpoint important differences in gene expression that will guide future health research.

Mice are the premier model organism for research into human health and disease because they share most of their protein-coding DNA with us. However, only a small fractionless than 2 percentof human and mouse DNA is used to code the building blocks of life. The regulatory elements in our DNA that control the expression of these genes are equally important, if not more so, to disease development and progression, and our understanding of it.

Using high-throughput DNA sequencing techniques, the research team looked at these functional elements in more than 1,000 data sets produced from over 100 mouse cell types and tissues. They were able to assign potential regulatory functions to 12.6 percent of the mouse genome. They then compared these elements and their functions to those of humans.

"This is the most comprehensive effort to do a genomic comparison between humans and mice at this level, including the regulatory elements and gene expression," said Feng Yue, an assistant professor in the department of biochemistry and molecular biology at Penn State College of Medicine. "We looked at regulations of genes, rather than genes themselves, and whether the gene is expressed in certain tissues or how much the gene is expressed there. We wanted to learn which gene expression patterns were conserved between humans and mice during mammalian evolution, and which gene expression patterns diverged."

Major similarities, but also important differences, emerged.

Although much similarity exists, mouse and human gene expression differs significantly in specific biological pathways.

"What we found is that the majority of the gene expression patterns are very similar in human and mouse," Yue said. "That shows that, indeed, the mouse is a good system to use in the study of human disease.

"But there are some genes whose patterns are not quite conserved between human and mouse," he continued. This divergence was most profound for areas of the mouse genome involved in the immune system and metabolic processes. "These genes may function in a different way between human and mouse."

Duplication of gene on X chromosome causes gigantism

BETHESDA, Md., Dec. 4 (UPI) -- Two is often better than one, but not when it comes to the DNA and RNA that make up the human chromosome. Researchers now believe gene duplication in a specific region, key in the regulation of childhood growth, is responsible for the rare disorder known as gigantism.

"Finding the gene responsible for childhood overgrowth would be very helpful, but the much wider question is what regulates growth," Constantine Stratakis, lead author of the new study, said in a press release.

Gigantism is a rare medical condition whereby children grow rapidly. Those affected typically grow to be abnormally tall; gigantism patients may also have delayed puberty, outsized hands and feet, and double vision.

To locate the offending genes, researchers at the National Institutes of Health used whole-genome analysis to study the DNA of 43 people with gigantism. The chromosomes of those afflicted with the rare disorder were then compared to family members who were born without the defect.

The analysis showed that gene duplication varied among the study participants, but researchers were able to narrow down the offending portion of the X chromosome to a stretch containing four duplicated genes. Scientists determined the gene most likely responsible for gigantism was GPR101.

"We believe GPR101 is a major regulator of growth," Stratakis said.

The gene duplication that causes gigantism results in a malfunctioning pituitary, the pea-sized gland that produces growth hormones. Researchers found activity of GPR101 was up to 1,000 times stronger than normal in children with enlarged and overactive pituitary glands.

The research was published this week in the New England Journal of Medicine.

Behind the boyish charm and impish innocence lies a ventriloquists doll with a dirty mind and a dirtier mouth. Jeff Dunham is the hilarious equal-opportunity offender of good taste. Hes so politically incorrect that anybody else would be arrested.

He takes on everybody from President Obama to star neighbor Britney Spears, from Mexican aliens to gays nobody is spared, not even the Popeyes and adjoining Chevron gas station restaurant manager from Mesquite or the retired print-paper salesman from Alabama, both of whom sat front row Wednesday night at Planet Hollywood.

Its savage and scything sarcasm and ridicule at Jeffs new show Not Playing With a Full Deck, and everybody wants to be the target of invective from Jeffs wacky collection of characters, including Tiny Jeff, who is an infomercial spawn of the comedian himself. The wrestling chase scene between the two of them is so realistic that I understand why Jeff himself gets grazed and banged up on the floor.

But its Peanut, the purple insult prince, and Achmed the Dead Terrorist who generate the most laughter, particularly with the question cards for Achmed from the audience. I will never know if Jeff walked into the trap from Phil McCracken or if he is the best actor at feigned surprise in the business.

Thats his charm. He or should I say his puppets? can say anything and do. Be it rude, vicious, comedic or vulgar, the jokes come at staccato speed, and your stomach is in pain from the laughter. Its a minefield of political sarcasm and religious risks that has audiences in hysterical delight while he softens the blows with intimate family photos that also get a lashing.

The figures, or the little people in the boxes, as he calls them, are the stars of the show to the point where the talented master manipulator behind them is almost overshadowed in the effortless back-and-forth alternating accents and body movements.

Iranian writers' group denounces continued censorship

On the occasion of the Day of Action against Censorship, Iran's Writers' Association announced that the Islamic Republic government continues to impose censorship in various and complex ways.

In a statement, the association writes that in the past year, despite President Rohani's election promises, censorship has continued to affect book publishing, the arts, the press and the internet, impeding freedom of speech.

They report that publishers are kept waiting for publishing permits, internet users are subjected to arrest and questioning, and many internet sites continue to be blocked.

The statement also refers to journalists being arrested, which make it a highly hazardous occupation for Iranians.

The Writers' Association cites the jamming of satellite programming as a serious form of government censorship imposed because it both violates the right of the public to access these programs and compromises their health.

Iran's Department of the Environment has called for an end to the use of interference waves to jam satellite programming, confirming that they can be linked to cancer.

China's influence threatens American universities, experts say

The Chinese government's influence over American universities is broad and deep, ranging from such subtle pressures as the denial of visas for vocal American scholars to more overt efforts, including opening Chinese cultural institutes on U.S. college campuses, experts on human rights and education told lawmakers Thursday.

That influence has become more pronounced as American universities open satellites campuses in the country and welcome Chinese government-funded programs on their stateside campuses, experts said.

"U.S. colleges and universities should not be outsourcing academic control, faculty and student oversight or curriculum to a foreign government," Rep. Christopher H. Smith (R-N.J.) said at a House Foreign Affairs Committee hearing. He said he would call for a Government Accountability Office study into academic agreements American universities have made with China.

Much of the debate centered on the Confucius Institutes, Chinese government-sponsored cultural centers at universities worldwide. Funded by the Chinese government, the institutes provide language instruction and sponsor cultural exchanges, but some experts worry they allow China to engage in "soft power" propaganda campaigns and encourage censorship over certain topics.

China says the institutes which experts estimate could number more than 400 worldwide are established only at schools that invite them and do not force their curriculum on any campus.

Experts suggested that the federal government could do more to finance Chinese-language programs within the United States, rather than relying on Chinese funds to do so.

"Why should we hand our young people over to an authoritarian government because they supply the funds?" asked Perry Link, a Chinese-language professor at UC Riverside. "We have enough funds for that."

Experts also called on lawmakers to withhold visas for Confucius Institute instructors as long as China continues to do the same for American scholars.

Mainly, they said, American universities must do more in their negotiations as they develop partnerships with China, pushing for broader discussion rather than quieting debates over controversial topics on their campuses.

"If you don't stake out the borderline, natural self-censorship will stick in," Link said.
