Psoriasis Medication May Be Effective in the Treatment of Alzheimer's

It is estimated that about 35 million people worldwide currently suffer from dementia and it is expected that the number will increase to 135 million by the year 2050. The disease is already one of the most common health problems in the elderly, which is why experts predict that the numbers of people affected will increase over time. Researchers at the Department of Psychiatry and Psychotherapy of the University Medical Center of Johannes Gutenberg University Mainz (JGU) have recently gained new insights into how it may in future be possible to treat patients with the currently most common form of dementia, Alzheimers disease. It seems that a drug that is actually approved for treatment of the dermal disorder psoriasis stimulates the activity of the enzyme ADAM10 in the brain of Alzheimers patients. There is already good evidence from basic research that this enzyme should be capable of suppressing Alzheimers disease-related effects such as impaired cerebral function and that it thus might improve learning and memory capacity in patients. The results of the related study have recently been published in the journalNeurology.

According to estimates of the German Alzheimers Association (DAlzG), approximately 1.5 million dementia patients currently live in Germany. Some 1 to 1.2 million of these suffer from Alzheimers. Medicine is currently only able to treat the symptoms of the disease and delay its progress and thus also the need for increased nursing care. No curative therapy has yet been developed. This means that Alzheimers disease remains one of the biggest challenges to modern medicine and is an important field for research.

There is still no consensus on what triggers the most common form of the disease, late-onset Alzheimers. However, it is generally accepted that the activity of certain enzymes called secretases plays a role here. These enzymes cleave proteins on cell membranes, releasing the products of this cleavage process into the extracellular space. What happens in Alzheimers is that there is increased cleavage of the amyloid precursor protein by beta-secretase, leading to the formation of amyloid-beta peptides. These peptides aggregate, damage nerve cells, and are the main component of the so-called Alzheimers plaques that accumulate in the brains of patients. The alpha-secretase ADAM10 is a competitor of beta-secretase. It cleaves the amyloid precursor protein in such a way that the synthesis of amyloid beta-peptides is prevented while the growth factor APPs-alpha, which protects nerve cells, is released.

Taking this information as their starting point, Dr. Kristina Endres and Professor Falk Fahrenholz of the Department of Psychiatry and Psychotherapy of the Mainz University Medical Center have decided to take a new approach to the treatment of Alzheimers. Working in collaboration with Professor Klaus Lieb and Professor Andreas Fellgiebel, both also working at the Department of Psychiatry and Psychotherapy, and with the cooperation of Professor Stefan Teipel and his team at the German Center for Neurodegenerative Diseases (DZNE) in Rostock, the researchers have demonstrated that oral administration of a psoriasis medication in a group of Alzheimers patients results in elevated levels of APPs-alpha in their spinal fluid. This is interpreted as a stimulation of the activity of the alpha-secretase ADAM10, which in turn would result in the reduced accumulation of Alzheimers plaques. In animal models of Alzheimers disease, it has also been shown that ADAM10 enhances learning and memory capacity. The medication was well-tolerated by the patients. In order to further investigate the effect of the test substance on cognitive performance and to establish whether it can be used as a long-term treatment for Alzheimers patients, larger clinical trials in which the substance is administered for longer periods will need to be undertaken.

Bluebird Gene Therapy Inducing Durable Cures in Blood Disorder Patients

SAN FRANCISCO (TheStreet) -- Bluebird Bio (BLUE) has now treated seven beta-thalassemia patients with its experimental, one-time gene therapy. Four of the patients -- all followed for longer than three months -- are producing enough oxygen-carrying hemoglobin on their own to eliminate the need for chronic blood transfusions.

Two of these super-responding beta-thalassemia patients -- followed for a year and nine months, respectively -- have hemoglobin levels of healthy adults. At this point, a single infusion of Bluebird's gene therapy has essentially cured them of this serious, inherited blood disease.

The remaining three beta-thalassemia patients were infused with Bluebird's gene therapy around one month ago so it's too early to assess their response. A single patient with sickle cell disease was also just treated within the past month.

It's still unreasonable to expect an equivalent strong response in every patient, but Bluebird is learning that as its therapy replaces the defective gene causing beta-thalassemia (or sickle cell disease) with a gene that is fully functional, the patient's ability to produce normal-functioning hemoglobin improves over time, said CEO Nick Leschly.

Stella Young spoke with 'searing honesty'

Stella Young was a diminutive comedian whose "searing honesty" made her a giant of disability advocacy.

Politicians, disability support groups and others paused on Monday to remember the feisty 32-year-old who labelled herself a "proud crip" and who used humour to challenge Australians to think differently.

Young passed away unexpectedly on Saturday, and a private funeral is being planned.

Her former employer, ABC Managing Director Mark Scott, said Young's advocacy liberated Australians to have a more frank and honest conversation about the issues people with disability faced, and the kind of support they needed.

"She spoke and wrote with a searing honesty and ... gave us insight into the lives of people with disabilities in a way that I don't think we'd really experienced before."

Young was the outspoken editor of the ABC's Ramp Up website, which recently ran out of funding.

"We set up that website with special government funding and that funding didn't continue ... but we kept an engagement with Stella because we knew what an important figure she had become," Mr Scott said.

She was due to record more ABC radio this week.

Political leaders joined in paying tribute to the comedian, who railed against the notion that disabled people were an inspiration.

"Stella dreamt of a society where people with a disability who studied, worked and achieved great things were conventional, even ordinary," Victorian Premier Daniel Andrews said.

In politically incorrect language, Jonathan Bernier may very well be the sports version of an idiot savant.

Brilliant at the thing he does but obtuse oblivious about stuff thats widely known to everybody else.

This is an affliction not uncommon among elite athletes, who grow up so intently focused on themselves the drive to succeed, where 99.9 per cent fail that the rest of the world slides right by. Its a kind of blinkered detachment. Many live in a bubble un-penetrated by matters of substance.

That said, Bernier is a nice guy, a kind person from what Ive seen in the dressing room, soft-spoken and still not entirely adept in English.

What the Toronto Raptors did to him over the weekend however inadvertently was just plain rotten.

His blunder on the red carpet last Friday was cringe-inducing, yet spontaneous.

Their role in the affair was careless.

The No. 1 Maple Leaf goalie has been absolutely roasted in recent days for obviously not knowing who Nelson Mandela was. Thats shocking, given that Madiba was arguably the most famous prisoner on the planet and global icon for human rights, a lifetime spent fighting against the apartheid horrors of South Africa before becoming its president, and a beloved statesman until his death a year ago.

The anniversary of Mandelas passing was beautifully marked by the Raptors in The Giant of Africa tribute incorporated into their game against the Cavaliers. Mandela was himself an athlete a boxer in his younger days who believed passionately in the power of sport to transform society. The event drew luminaries such as Magic Johnson and Charles Barkley to the Air Canada Centre to celebrate the man and his legacy.

Bernier and several teammates were among those in attendance. He was also, I dare say, the only person there who evidently had never heard of Mandela.

Vodafone blocks Chaos Computer Club site, fueling 'Net censorship concerns in UK

Vodafone UK isnt letting its customers access the website of the Chaos Computer Club (CCC), one of the oldest and largest associations of computer hackers in Europe.

Its not clear why the mobile operator has blocked the ccc.de website since late last week, but the CCC believes its because its site has been misclassified in the Internet filtering system used by Vodafone.

Since its founding in 1981 in Berlin, the CCC has highlighted security risks in technology affecting a large number of people, has exposed government surveillance and has advocated for privacy and freedom of information online. Every year the group organizes the Chaos Communication Congress, the largest hacker convention in Europe.

Vodafone UK did not immediately respond to a request for comment.

For years ISPs in the U.K. have blocked access to websites that host images of child sexual abuse based on a list maintained by an organization called the Internet Watch Foundation. However, in 2013 the U.K. government convinced ISPs to build their own Internet safety filters in order to block sites that host content unsuitable for minors, including pornography and drugs.

The initiative was criticized by human rights and Internet advocacy groups because it allows ISPs to judge what is right or wrong for young people and opens up the door to over-blocking, where users are prevented access to non-harmful and even helpful websites and information.

When these filters were introduced, their abuse was imminent, said CCC spokesman Dirk Engling in a blog post Friday. Today, we are shocked to learn that they not only block access to our site, but also to our conference. We see this as proof that censorship infrastructureno matter for which reasons it was set up, and no matter which country you are inwill always be abused for political reasons.

According to a check on http://www.blocked.org.uk, a website maintained by U.K.-based Open Rights Group (ORG), ccc.de was being blocked by both Vodafone and Three, another U.K. mobile operator, as of Sunday. The Chaos Communication Congress tickets site, tickets.events.ccc.de, was only being blocked by Vodafone.

Overblocking is a major issue when it comes to filters that are designed to stop children and young people from seeing adult content, said Pam Cowburn, ORGs communications director, Monday via email. When we tested the Alexa 100k websites, we found that almost 1 in 5 sites were blocked. Although many of these may have been deliberately blocked because, for example they are pornography, this high figure suggests that there is a real problem with over blocking.

Sites that have been incorrectly blocked in the past include political blogs, sites about womens rights and even a Porsche sales site, Cowburn said. A site that offers support to victims of sexual abuse and rape, aged 16 and higher, was also blocked and so were some sites that belong to charities, she said.
