Cancer-causing mutation discovered in 1982 finally target of clinical trials

In 1982, the gene TRK was shown to cause a small percentage of colon cancers. In 2013 and 2014, next-generation sequencing of tumor samples found fusions of the TRK family of genes in at least 11 tumor types, including lung, breast, melanoma and more. Now, a recent article in the journal Cancer Discovery describes clinical trials at the University of Colorado Cancer Center and elsewhere that match drugs to this long-overlooked oncogene, offering targeted treatment options for cancers that harbor these gene abnormalities (e.g. ClinicalTrials.gov #NCT02122913).
"We didn't initially discover the gene. But now technology lets us find the gene in actual patient samples and drugs are available to target these gene rearrangements, making it possible to treat TRK cancers in clinical trials in ways we only dreamed of thirty-two years ago," says Robert C. Doebele, MD, PhD, investigator at the CU Cancer Center and associate professor of Medical Oncology at the CU School of Medicine.
The TRK family of genes, including NTRK1, NTRK2 and NTRK3 are important in the developing nervous system. In the womb, these genes and the proteins they encode are essential for the growth and survival of new neurons. After birth, these genes are unneeded in many tissues and so are programmed to go dormant. Some cancers wake them up - when improperly fused with other nearby genes, genes in the TRK family can restart their ability to signal cells to grow and become immortal, which in adult tissues can cause cancer.
"What we're finding is that while TRK fusions may not be the major cause in any single, major cancer, it's the cause of small percentages of many cancer types," Doebele says.
For example, the recent article cites studies showing NTRK fusions in 3.3 percent of lung cancers, 1.5 percent of colorectal cancers, 12.3 percent of thyroid cancers, about 2 percent of glioblastomas, and 7.1 percent of pediatric gliomas (brain tumors).
"These numbers add up," Doebele says.
A decade ago, the NTRK fusion and other, related gene rearrangements were un-druggable. Now, an entire class of drugs has been developed to target this type of genetic abnormality, namely "tyrosine kinase inhibitors," which are able to precisely turn off these genes like NTRK that send dangerously misplaced cell-survival signals. For example, the FDA-approved drug crizotinib targets ALK and ROS1 fusion genes in lung cancer.
A host of new drugs in this class of tyrosine kinase inhibitors target TRK fusions, for example investigational anti-cancer agents RXDX-101, TSR-011, LOXO-101, PLX-7486 and more.
"A lot of doctors in academia or community hospitals are ordering next-generation sequencing panels for their patients. If it turns out that patients' tumors have TRK alterations, I want their doctors to know that there are treatment options available via clinical trials," Doebele says.

WHAT IS MODERN CENSORSHIP?
At Project Censored, we examine the coverage of news and information important to the maintenance of a healthy and functioning democracy. We define Modern Censorship as the subtle yet constant and sophisticated manipulation of reality in our mass media outlets. On a daily basis, censorship refers to the intentional non-inclusion of a news story or piece of a news story based on anything other than a desire to tell the truth. Such manipulation can take the form of political pressure (from government officials and powerful individuals), economic pressure (from advertisers and funders), and legal pressure (the threat of lawsuits from deep-pocket individuals, corporations, and institutions).
In our view, the only valid justification for declining a news story is that in a medium limited by time and space, another news story was simply more important to the people of the community, whether local, national or international. While admittedly a subjective process, it is nonetheless, a process to be undertaken by the news people themselves (the investigative journalists and editors), NOT by the managers and CEOs of their parent company. No professional journalist or researcher should ever have to face the destruction of his or her career (or life) simply because they wanted to tell the truth. While no two people will always agree on what story is more important than another, a system where the working reporters and editors run the newsroom would at least provide a fertile environment for debate, dissent and critical thinking.
The growth of independent media and journalism in recent years shows that people throughout the world yearn to hold not only their leaders accountable, but their media sources as well. For that reason, the Project Censored research program continues, in its small way, to support and highlight those who tell the truth about the powerful (no matter the consequences) and are relentless in their quest to hold Big Media accountable for their decisions.

Chelsea Handler, Miley Cyrus fight Instagram's censorship of topless photos

LOS ANGELES, Dec. 29 (UPI) -- Chelsea Handler and Miley Cyrus have reignited a crusade against Instagram's nudity policy after topless photos from the entertainers were deleted from their accounts last week.
While Handler has famously protested the site's nudity policies in the past, her new quest was prompted by the removal of a topless photo of her in the snow. The comedian shared a topless image of herself looking for her missing German Shepard Chunk while skiing on Dec. 26, but the photo was taken off of her account for not adhering to the image sharing site's polices.
Handler later posted the same image on Twitter, under the caption: "Finding chunk. Instagram had him!"
Cyrus, on the other hand, stood up for the #FreeTheNipple cause on Saturday by posting a black and white photo of herself that showed her nipples, captioned: "Some lame ass deff gonna [flag] that but f--kkkkkkk it." Like she predicted, the picture was soon deleted from her account.
Since then, the 22-year-old singer has been posting Photoshopped pictures featuring photos of herself when she was young on the bodies of naked women with blurred nipples.
"Free em bb ( . ) ( . )" Cyrus wrote in one of the posts.
Cyrus and Handler join the likes of Rihanna and Scout Willis, the daughter of Bruce Willis and Demi Moore, in their protest against Instagram's policies when it comes to nipples.
While defending the campaign earlier this year, Willis said the protest had "nothing to do" with posting nudes on social media.
"It's about the basic hypocrisy of what is and isn't allowed," she said in May. "Violence, drugs, and the general degradation of women is ok but areola is 100 percent taboo. That's why I am mad."

Google Inc's Gmail was blocked in China after months of disruptions to the world's biggest email service, with an anti-censorship advocate suggesting the Great Firewall was to blame.
Large numbers of Gmail web addresses were cut off in China on Friday, said GreatFire.org, a China-based freedom of speech advocacy group. Users said the service was still down on Monday.
"I think the government is just trying to further eliminate Google's presence in China and even weaken its market overseas," said a member of GreatFire.org, who uses a pseudonym.
"Imagine if Gmail users might not get through to Chinese clients. Many people outside China might be forced to switch away from Gmail."
Google's own Transparency Report, which shows real-time traffic to Google services, displayed a sharp drop-off in traffic to Gmail from China on Friday.
"We've checked and there's nothing wrong on our end," a Singapore-based spokesman for Google said in an email.
Almost all of Google's services have been heavily disrupted in China since June this year, but until last week Gmail users could still access emails downloaded via protocols like IMAP, SMTP and POP3. These had let people communicate using Gmail on apps like the Apple iPhone's Mail and Microsoft Outlook.
China maintains tight control over the internet, nipping in the bud any signs of dissent or challenges to the ruling Communist Party's leadership.
The country is host to the world's most sophisticated internet censorship mechanism, known as the Great Firewall of China. Critics say China has stepped up its disruption of foreign online services like Google over the past year to create an internet cut off from the rest of the world.
The Google disruption began in the run-up to the 25th anniversary of the government's bloody crackdown on pro-democracy demonstrators around Beijing's Tiananmen Square on June 4, 1989.

The Wall Street Journal: Gmail totally blocked in China after censors close loophole

China has upped the ante in its longstanding censorship of all things Google. In the six months since Googles mail service Gmail was blocked in mainland China, users had been able to access it using third-party email applications such as Microsoft Outlook or Apple Mail.
Beijing now appears to have closed the loophole, completely shutting down access to Gmail behind the so-called Great Firewall. Google GOOG, -0.69% data showed Gmail appeared to have been walled off starting Friday. Google spokesman Taj Meadows acknowledged the drop in traffic and said Monday that theres nothing wrong on our end.
Chinas State Internet Information Office did not immediately respond to a request for comment Monday. At a daily press briefing, Foreign Ministry spokeswoman Hua Chunying said she wasnt aware of the matter. She added that the government always welcomes foreign businesses to carry out relevant work in China.
Beijing doesnt always publicize its reasons for shutting down websites, permanently or temporarily. In the past, such censorship has tended to coincide with periods of politically sensitive anniversaries or big political conferences in the capital like its annual legislature.
The last time Google came under such attack was in June, ahead of the 25 anniversary of Beijings deadly suppression of the Tiananmen Square pro-democracy protests. During that round, Googles suite of services including Google+, Gchat and Drive were all shut down. It was the first time that Beijing had blocked Google services to such a significant extent ahead of a Tiananmen anniversary.
