Scientists Tease out Genes That Signal Risk of Heart Failure

Scientists are unraveling a mystery behind a fairly common disease that leads to heart failure: Why do some people with a key mutated gene fall ill while others stay healthy?

Researchers tested more than 5,200 people to tease apart when mutations really are harmful or are just bystanders. The work could help in screening families prone to heart failure but also has broader implications as more people undergo genetic tests that can turn up unnecessarily worrying results.

Heart failure, when the heart cannot pump blood properly, is caused by a variety of conditions, including damage from heart attacks. But one trigger is dilated cardiomyopathy, a condition that makes the heart's muscle walls stretch out of shape, becoming progressively weaker. It can run in families, but often there's no obvious cause.

Titin is a protein that gives muscle tissue, including heart muscle, its elasticity. In 2012, researchers reported that gene mutations that make that protein shorter, or truncated, were a common cause of dilated cardiomyopathy, accounting for a quarter of cases. The problem: A lot of healthy people also harbor mutations in that stretchy muscle protein, yet some gene tests already look for the glitches.

So a British-led research team mapped the titin-producing gene in 5,267 people, ranging from the healthy to the seriously ill. Where the DNA glitches are located in this huge gene is key, the team reported Wednesday in the journal Science Translational Medicine.

Mutations that caused dilated cardiomyopathy are located at one end of the gene sequence, while mutations in healthy people occur in other spots apparently less important for heart muscle, they reported.

Another discovery: Titin-caused cardiomyopathy is more severe than other forms of the disease, said lead researcher Angharad Roberts of Imperial College London. Those patients are more likely to suffer life-threatening irregular heartbeats, suggesting doctors might use genetic testing to guide therapy including when to implant defibrillators.

Somehow, this truncated protein is poisoning heart muscle cells, the researchers concluded.

When cardiomyopathy runs in the family, close relatives get regular heart screening to see whether they're developing it, too. The researchers said finding which kind of mutation family members harbor could help narrow who's really at risk but it would take more in-depth genetic testing than is routine today.

"In an era where genetic testing and genome sequencing is increasingly available, more and more titin mutations will be identified, often as incidental findings," Roberts said. "Accurate interpretation of these results is vital to avoid unnecessary follow-up and anxiety."

1. IoT: The Insecurity of Things

The Internet of Things has become an inundation of things. Hundreds of innovative, connected devices have emerged to interact with, track, monitor, and simplify just about every area of our lives. Butthese technologies typically have access to sensitive, personal information, and they also introduce a wide variety of new security issues for attackers to exploit.

In previous years the Internet of Things was not a big deal," warns Robert Hansen, VP of WhiteHat Labs for WhiteHat Security, "but were seeing an increasing number of vulnerabilities in internet capable devices, like TVs, home security systems, automation.

2. Sophisticated DDoS Attacks

Denial-of-service attacks are more of an annoyance than anything else. They dont directly steal your information, or cause any overt harmthey just flood a site or service with so much traffic that it becomes overwhelmed and prevents legitimate users from connecting to it. As many Xbox and PlayStation gamers learned over the holidays, though, DDoS attacks are becoming more advanced, and have a very real impact.

In 2014, DDoS attacks became much more sophisticated. Though much of the reporting focused on the size of attacks, a more troubling trend was the advancement in attack techniques, stresses Barry Shteiman, director of security strategy for Imperva. He notes that attackers have evolved beyond simple flooding of traffic, and can now morph and adapt based on the defenses in place on the target network.

3. Social Media attacks

Mark Bermingham, director of global B2B marketing atKaspersky Lab, anticipates a rise in social media and waterholing attackscompromising a website or service commonly used by the target group in an effort to infect one or more of them, and allow the malware to spread from there.Attackers continue to develop new techniques to exploit social networks. As Bermingham puts it, Security measures cant overcome stolen credentials and click-throughs to dubious links.

Kevin Epstein, VP of advanced security and governance at Proofpoint, agrees that social media attacks are a serious concern for 2015. In a recent blog post, he notes, In 2015, Proofpoint expects inappropriate or malicious social media content to grow 400 percent as attackers target enterprise social media accounts to perpetrate confidence schemes, distribute malware, and steal customer data.Greater awareness and vigilance are the best defenses.

Censorship of information in the present age is an impossibility, Information and Broadcasting Minister Arun Jaitley said today but observed that aberrations like paid news are likely to creep in when news organisations do not have realistic financial models.

He said the definition of news and the behaviour of consumer have changed with technological advancement and something that camera cannot capture is hardly news these days.

Articulating his views on the media scene at a function here, Jaitley said, One of the worries is that the financial model for all (news) organisations must be a realistic model. If the financial model is not a realistic model, then imperfections will enter. And these imperfections will lead to aberrations. Paid news is one such aberration.

Paid news has been a concern about which even the Election Commission has been looking at ways to deal with.

At the same time, he made it clear that media censorship in the current age was not possible.

Fortunately, there are very few dictatorships in the world. But even if there were, censorship, because of technology itself would be an impossibility, he said.

Jaitley, who also holds Finance Ministry portfolio, said that in this age of competition and to gain more eyeballs, it may seem that quality is being compromised. However, he added that he had faith that in the long run, the best will succeed.

He said the rapid advances of technology in broadcasting sector have brought along their own challenges and it is difficult to predict the future horizons of this evolution.

With tools of information dissemination freely available, Jaitley said he had at times read his own speeches which he had never delivered.

Facebooks chief executive, Mark Zuckerberg, has defended his decision to condemn the recent terrorist attack on Charlie Hebdo magazine on free speech grounds, despite his companys willingness to censor some content.

Zuckerbergs status update on 9 January promising a service where you can speak freely without fear of violence sparked a debate about Facebooks own censorship, from breastfeeding photos to a post by Pakistani actor Hamza Ali Abbass that questioned the value of insulting speech.

Unsurprisingly, the subject of his Charlie Hebdo post came up during Zuckerbergs latest public question and answer session, held this week in Colombia where Facebook is launching its Internet.org initiative to get more people online.

He was asked why he had spoken out about the Charlie Hebdo attack, but not about other violent events around the world, including in Iraq and Palestine.

It wasnt just a terrorist attack about just trying to do some damage and make people afraid and hurt people. This was specifically about peoples freedom of expression and ability to say what they want, said Zuckerberg.

Although he took pains to stress that all terrorist incidents are really horrible, Zuckerberg said he spoke out about Charlie Hebdo because he saw it as an attack of freedom of expression, and so particularly relevant to Facebook.

That really gets to the core of what Facebook and the internet are, I think, and what were all here to do. We really stand up and try to make it so that everyone can have as much of a voice as possible, he said.

There are limits and restrictions on these things, but across the board we generally are always trying to fight to help as many people as possible share as much as they want. This event just seemed like an event where people needed to come together not only to fight back against terrorism... but also to stand up for giving everyone in the world a voice.

A follow-up question from a member of the audience at the Q&A asked whether Facebook would break the law in countries where free speech is restricted, to defend those principles.
