Introgression in the pig genome leads to their altitude adaptation

Posted: January 27, 2015 at 10:47 am

Jan 26, 2015, Shenzhen, China -Scientists from Jiangxi Agricultural University, BGI and University of California published their latest research on genetic mechanism of pig altitude-adaptations in Nature Genetics online. Their research underlined the importance of introgression for the first time as a potential reason for pig adaptations to cold and hot environments, which provided novel insights into the evolutionary history of pigs and the role of introgression in adaptation more generally.

Pigs, as one of the earliest domesticated animals, were domesticated largely in Near East and China approximately 10,000 years ago. Since then, under the combined effects of natural selection and human-driven artificial selection, pigs evolved phenotypic diversity in appearance, fertility, growth, palatability, and local fitness. As the genetic basis of those adaptive evolution remained largely unexplored, scientists of this article conducted whole-genome sequencing and selective sweep analysis trying to figure out the genetic basis contributed to the adaptation of domesticated Chinese pig breeds, especially related to thermostatic regulation for varying-latitude environments.

In the current study, the scientists selected 69 pig individuals represented 11 geographically diverse breeds and 3 populations of wild boar from cold and hot environments in China. This is one of the first population genomics analyses to use high-coverage whole-genome sequencing in pigs, each individual has been sequenced more than 20-fold depth with the genome coverage of 95%. Totally, 41 million variants have been identified, with 21 million variants were absent from the dbSNP database. Substantially, a nearly complete catalogue of the genetic variants has been compiled, which allowed the scientist to identify a genome-wide set of loci for local adaptation in Chinese pigs.

From the genome wide scan, a set of gene loci correspond to thermostatic regulation has been identified. Notably, an exceptionally large (14 Mb) and low-recombination region on the X chromosome appeared to have two distinct haplotypes in the high- and low-latitude populations, which may be possibly responsible for adaptations to cold and hot environments respectively. Another surprising finding of this study was that the adaptive haplotype in the high-latitude populations was likely introduced from another divergent Sus species. This is the first example of adaptive evolution triggered by interspecies introgression in domesticated animals, which gave us a clue that introgression event between divergent species may be an important resource for evolutionary adaptation and could largely facilitate this process.

Likai Mao, the project manager of BGI, said, "We found lots of loci could be related with environmental adaptation. When we noticed a big region with special pattern on the haplotype figure of chromosome X, we knew something must be there. "

Eczema support group set up in North Somerset

Posted: at 10:47 am

15:10 27 January 2015

Report by Tom Wright

Annie Westaway with her three-year-old daughter Chloe.

Archant

A YATTON mother has set up a parental support group to help people caring for children suffering from eczema.

Annie Westaways daughter, Chloe, suffers with the common skin condition but because of its variety in severity, it can make treating it a bit of a headache.

The lack of information inspired Annie to start the group, which will meet this week for the first time.

Annie, who lives in Wood Kiln, said: It sounds like nothing, but when you are a parent, it takes up a lot of your time.

Ive not met a child with so much bad eczema.

The more information you can get the easier it is to help your child.

Gene expression at play in age-dependent vaccine response

Posted: at 10:46 am

A new Yale School of Medicine study has answered the question of why younger bodies respond better to flu vaccines than older ones.

Researchers found that healthy older adults produced protective antibodies for the flu vaccine at a different rate than younger adults. Lower vaccine responsiveness in older adults is related to less effective immune responses. The results represent the first genome-wide analysis of age-dependent responses to the flu vaccine. The study was published in the journal Aging on Jan. 14.

The research was conducted with the hope that the genetic pathways that were identified could be future targets for therapies to improve responses for vaccination, particularly in older adults, said Albert Shaw, professor of medicine and senior author of the paper.

Even when the influenza vaccine is a good match for currently circulating strains of the virus, most vaccines are less effective for older adults, Shaw said. In fact, roughly 90 percent of deaths from influenza in the country occur in people older than 65, Shaw said.

Shaw said he and his team wanted to identify patterns of gene expression that were found in older and younger adults who showed excellent responses to vaccination and compare those patterns to those who had poor responses to vaccination.

They found specific patterns of gene expression that corresponded to antiviral responses, including those that reflected activities of cells that make protective antibodies against the influenza strain and vaccine, he said. Although antibodies were created about seven days after vaccination for young adults, for older adults, the timing varied, he added.

The studys findings were not news to representatives from leading vaccine development companies.

It is well known in our industry that the elderly mount less of an immune response to vaccination, said Charles Altman, head of medical affairs at BioCSL. If you look in any product label, you will find that.

Many manufacturers are trying to improve elderly response, but they face limited options for innovation due to regulations put forth by the World Health Organization, which makes a recommendation each year about which viral strains manufacturers can put in vaccines.

Because manufacturers are not allowed to modify which viral strains they use in their products, they can only vary a few factors, among them dosage and added adjuvants material added to vaccines to promote immune response Altman said. Therefore, the study has little practical relevancy for manufacturers like BioCSL.
