Master Switch Found to Stop Tumor Cell Growth by Inducing Dormancy

Newswise (New York January 30, 2015) Two existing cancer drugs turn on a gene that tells tumor cells to remain inactive, according to a study led by researchers at the Icahn School of Medicine at Mount Sinai and published today in Nature Communications.
Researchers discovered that the gene NR2F1, when switched on, programs tumor cells to stay dormant. When the gene is switched off, tumor cells divide and multiply as part of abnormal growth, potentially allowing dormant cells to grow into tumors throughout the body (metastasis). Combining the anticancer drugs azacytidine and retinoic acid significantly increased the amount of active NR2F1 in tumor cells. These patterns were found in mouse models of several cancers, and confirmed in prostate cancer cells from human patients.
Results suggest that NR2F1 is a master regulator of tumor cell growth, influencing several genes that determine whether cells remain inactive, or quiescent in medical terms. According to the study, NR2F1 exerts control over long lasting programs in stem cells in the human embryo, where it directs cells to stop growing and become specialized cells (neurons) for life. This function suggests that NR2F1 may exert a long-lasting effect on tumor cells, keeping them dormant after they have broken off from an original tumor.
Our results explain why some tumor cells scattered through the body are committed to remaining harmless for years, while others cause active disease, said Julio A. Aguirre-Ghiso, PhD, Professor of Medicine, Hematology and Medical Oncology, and Otolaryngology at the Icahn School of Medicine. In finding this master switch we found a way to analyze tumor cells before treatment to determine the risk of a cancer recurrence or metastasis.
Azacytidine and retinoic acid, the latter a form of vitamin A, prevented tumor cells from rapidly multiplying, restored normal cell function, and activated several tumor suppressor genes that are often turned off in tumors, said study co-leader Maria Soledad Sosa, PhD, a postdoctoral fellow in Hematology at the Icahn School of Medicine. We now have strong evidence that combining these well-known drugs may have a profound, long-lasting therapeutic effect.
The current study builds on the research teams earlier finding that lowering amounts of tumor suppressor genes TGF2 and p38 awakened dormant tumor cells, fueling metastatic tumor growth. Azacytidine and retinoic acid restored TGF2 expression and p38 activation to drive tumor cell dormancy.

Iowa poll: Scott Walker leads GOP field

Wisconsin Gov. Scott Walker is the top choice for Iowa GOP voters ahead of the 2016 caucuses in the state according to a new poll. But Sen. Rand Paul (Ky.) is right behind.
Walker leads the field with 15 percent of voters, according to the poll from the Des Moines Register. His stock has been rising in conservative circles, especially in the Hawkeye State, after a strong showing at the Iowa Freedom Summit last week.
Paul is nipping at Walkers heels with 14 percent support. Iowa Republicans received the Paul family brand of libertarianism well in 2012, when Rand Pauls father, former Rep. Ron Paul (Texas), ran for president. The elder Paul initially came in third, and his campaign went on to secure the majority of the states delegates unbound by those results.
After that, support falls off. Sen. Ted Cruz (Texas) and former Sen. Rick Santorum (Pa.) come next at five and four percent respectively. And a mass of Republican contenders, including Sen. Marco Rubio (Fla.), former Texas Gov. Rick Perry (Texas), Louisiana Gov. Bobby Jindal and real estate magnate Donald Trump round out the group, with the lowest amount of support measured.
The Iowa caucuses are vital because they are the first contest in the presidential nominating process. But theres still a year left to go, and anything can happen.
Just months before the 2012 Iowa caucuses, former Rep. Michele Bachmann (Minn.) won among Republicans in the Ames straw poll, a popular pre-caucus poll. She won five percent of the popular vote and zero delegates in the actual caucuses, prompting her to drop out of the race.

How human testers are revealing our Facebook News Feed habits

It turns out that not everything you see in your Facebook News Feed has been picked by an all-seeing, all-knowing computer algorithm: A new report on the social networks internal testing system has revealed that human users are also heavily involved in providing feedback on what works and what doesnt.
Some of the secrets of the News Feed are revealed in a post on Mediums Backchannel, which explains the process that around 600 testers go through every weekday. These men and women are asked to give detailed feedback on what appears in their News Feed: What they liked, what they didnt and why. Its a more elaborate version of the feedback tools available to every Facebook user, and the company thinks its vital in keeping the relevant stories coming.

If you just watch people eat doughnuts, youre like, People love doughnuts, lets bring them more doughnuts,' product manager Greg Marra explained to Backchannel. But if you talk to people theyre like, No actually what I want is to eat fewer doughnuts and maybe eat a kale smoothie. Then we can give them some kale smoothies, too.
Facebooks human team of testers are paid for around four hours of work each day and will look at roughly 100 stories during that time. For each story, they must answer eight questions on how relevant it was and how well it connected them to the friend who posted it. Important posts from close friends and family are the ones that we most want to see, according to the testers, and it appears that clicking like doesnt necessarily mean we approve of whats been posted its just a way of us expressing a feeling of connection with the poster.
Facebook knows the quality of the News Feed is crucial in keeping users checking back in with the site. A few days ago it tweaked its internal code to cut down on the amount of hoax poststhat were appearing, and engineers say that minor changes to the News Feed algorithm are rolled out on a daily basis. When we asked what are the best stories the highest percentage of impact type is a strong emotional reaction, says News Feed product director Adam Mosseri. People really want to see stuff that drives a laugh or makes them feel happy, not necessarily information thats super valuable.
