How Penn Medicine and people from Pakistan are helping fight heart disease – The Daily Pennsylvanian

Penn Medicines new Center for Digital Health will serve as a connection between technology, social media and healthcare.
Astudy led in part by Penn scientistsexamined the DNA of over10,500 Pakistanis in order to guide research on treatments for heart disease.
Many drugs developed to lower heart disease havetargeted a gene linked to arterial plaque, Science Magazine explained. But this study found that the absence of this gene may not greatly affect one's risk for heart disease.
Instead, the study found that the absence of a different gene called APOC3 might help.
Penn professor Danish Saleheen and other researchers searched Pakistan for individuals missing a functional copy ofthe gene. Many Pakistanis marry their first cousins, so parents more frequently pass on identical copies of a gene to their children meaning that they're also more likely to pass on two nonfunctional copies.
People with these deficient copies of agene are known as genetic knockouts, Philly.com explained. Scientists can deliberately breed animals to produce knockouts, but they must survey large numbers of peopleto find them in humans.
The researchers found a man,woman and their nine children in Pakistan who were all APOC3 knockouts.
When they consumed a high-fat meal, study participants without functioningAPOC3 genes did not experience elevated levels of plasma triglycerides, according toPenn Medicine. Higher levels of triglycerides are abiomarker of heart disease risk.
The family seemed healthy otherwise, which could suggest that missing the function of the APOC3 gene could help treat heart disease without other, harmful effects, Science Magazine noted.
Saleheen told Penn Medicine that these were the worlds first APOC3 knockouts found. Hes been working in Pakistan for over 10 years, amassing as many blood samples as possible, to identify different genetic knockouts so far hes found over 1,300 in over 70,000 participants, a figure he hopes will eventually surpass 200,000 as the work continues.
The study was led by Penn's Perelman School of Medicine, the Center For Non-Communicable Diseases in Pakistan, the Broad Institute of MIT and Harvard and the University of Cambridge.
Co-senior author of the studySekar Kathiresan told Science Magazine that he hopes this work will lead to a "Human Knockout Project," similar in caliber to the Human Genome Project, in order to aggregate data on knockouts.

WSU sleep researchers discover why some people may toss and turn more than others – The Spokesman-Review

A research team led by a Washington State University biochemist could help scientists shed light on why we need to sleep, and why some people have an easier time resting than others.
Jason Gerstner, a research assistant professor at WSUs Elson S. Floyd College of Medicine, found that mutations in a sleep gene in the brain can cause humans, mice and fruit flies to have less restful sleep.
The results of the study were published in a peer-reviewed article in the journal Science Advances earlier this month.
It might sound odd, but scientists still arent sure why sleep is necessary. One theory is that sleep helps with memory formation and the brains growth and change: what scientists call neuroplasticity. Other theories maintain that sleep is restorative for the body and that it lowers metabolism, helping to conserve energy.
We still dont fully understand what biological function sleep is serving, Gerstner said. One of the ways we can get at answering that question is through examining neurobiological pathways.
Much of Gerstners research has focused on a particular gene, FABP7, thats been linked to sleep function. In previous research, Gerstner saw the genes expression cycles naturally during the day in mice, mirroring sleep-wake cycles.
For this study, Gerstners team looked at a sleep study of Japanese men, some of whom had a naturally occurring mutation in their FABP7 gene. Men with the mutation slept about as long as men without it, but their sleep was more fitful, with more bouts of time spent awake during the night.
Men with the mutation also reported more symptoms indicating clinical depression on an assessment, though neither group scored high enough to meet the criteria for depression. Gerstner said that suggests either the gene mutation itself or sleep disturbance might be linked to depression in some way.
There were no significant differences in health, age or sleepiness between the two groups of men.
The study showed similar restlessness in rats that had their FABP7 genes knocked out and in genetically engineered fruit flies with the same gene mutation. Because the mutation works the same way across species, its a promising finding for future research, and even for treatment of sleep disorders.
The FABP7 mutation causes the gene to create a different protein sequence. That affects which other proteins in the brain the sequences bind to, which in turn can influence a broad range of functions, like gene expression, inflammation and other brain functions.
The researchers also found the specific part of the brain, a star-shaped cell called an astrocyte, where FABP7 plays a role in sleep.
Previously, those cells were thought to be support cells for neurons, Gerstner said. Now, scientists are learning theyre important in their own right.
This is some of the earliest evidence that astrocytes really play a role in sleep, said Isaac Perron, a doctoral student in neurobiology at the University of Pennsylvania who worked with mice in the experiment.
Perrons interest is in sleep and nutrition. Because the proteins coded by FABP7 bind with fatty acids like omega-3s, he thinks the gene might be a link in showing how the fatty acids we eat can influence brain functions, including sleep.
Jerry Yin, a professor of genetics at the University of Wisconsin-Madison, who worked on the fruit fly portion of the research, said finding a common pathway like FABP7 helps people looking at medications or gene therapies target their treatments.
Knowing FABP7 works in astrocytes helps researchers tailor their focus, since those cells are where youre likely to have an effect manipulating this gene, Yin said.
Testing a therapy or medication is also easier because the FABP7 impact on sleep works in fruit flies and mice, both of which are commonly used in research.
Since weve narrowed down particular protein expressed within astrocytes, it underscores the importance of these cells in regulating complex behavior across species, Gerstner said.

Using the new gene-editing enzyme CRISPR-Cpf1, researchers at UT Southwestern Medical Center have successfully corrected Duchenne muscular dystrophy in human cells and mice in the lab.
The UT Southwestern group had previously used CRISPR-Cas9, the original gene-editing system, to correct the Duchenne defect in a mouse model of the disease and in human cells. In the current work, they used a new variation of the gene-editing system to repair the defect in both a mouse model and in human cells.
"We took patient-derived cells that had the most common mutation responsible for Duchenne muscular dystrophy and we corrected them in vitro to restore production of the missing dystrophin protein in the cells. This work provides us with a promising new tool in the CRISPR toolbox," said author Dr. Eric Olson, Chairman of Molecular Biology, Co-Director of the UT Southwestern Wellstone Muscular Dystrophy Cooperative Research Center, and Director of the Hamon Center for Regenerative Science and Medicine.
The research appears in the journal Science Advances.
CRISPR-Cpf1 differs from CRISPR-Cas9 in a number of key ways. Cpf1 is much smaller than the Cas9 enzyme, which makes it easier to package inside a virus and therefore easier to deliver to muscle cells.
It also recognizes a different sequence of DNA than Cas9 does, which provides greater flexibility in terms of use. "There will be some genes that may be difficult to edit with Cas9 but may be easier to modify with Cpf1, or vice versa. The two proteins have different biochemical properties and recognize different DNA sequences, so these properties create more options for gene-editing," said Dr. Olson, who holds the Pogue Distinguished Chair in Research on Cardiac Birth Defects, the Robert A. Welch Distinguished Chair in Science, and the Annie and Willie Nelson Professorship in Stem Cell Research.
"By either skipping a mutation region or precisely repairing a mutation in the gene, CRISPR-Cpf1-mediated genome editing not only corrects Duchenne muscular dystrophy mutations but also improves muscle contractility and strength," said co-author Dr. Rhonda Bassel-Duby, Professor of Molecular Biology and Associate Director of the Hamon Center for Regenerative Science and Medicine.
Duchenne muscular dystrophy is caused by a mutation to one of the longest genes in the body. When there is a DNA error in the dystrophin gene, the body doesn't make the protein dystrophin, which serves as a sort of shock absorber for the muscle fiber. Since there are numerous places in the dystrophin gene where a mutation can occur, flexibility for gene-editing treatment is crucial.
Duchenne occurs in about 1 in every 5,000 boys, according to the Centers for Disease Control and Prevention. Duchenne muscular dystrophy is a progressive disease affecting both muscle used for movement and heart muscle, with patients typically succumbing before age 30 due to heart failure.
"CRISPR-Cpf1 gene-editing can be applied to a vast number of mutations in the dystrophin gene. Our goal is to permanently correct the underlying genetic causes of this terrible disease, and this research brings us closer to realizing that end," Dr. Olson said.
"CRISPR-Cpf1 differs from CRISPR-Cas9 in a number of key ways, including being easier to deliver to muscle cells, said Yu Zhang, a graduate student in Dr. Olson's lab and the first author of this study.

China’s WeChat Is A Censorship Juggernaut | Fortune.com – Fortune

Earlier this month the Chinese social media giant Tencent passed Wells Fargo as the world's tenth most valuable publicly traded company.
It would never have grown to that size were it not for the company's close relationship with China's government. The government requires a high level of cooperation from technology companies, which is one reason Google ( googl ) decided to leave the market seven years ago and why overseas social networks like Facebook ( fb ) , Twitter ( twtr ) , Pinterest, Line, Telegram, among others, remain blocked in China.
When it comes to censoring topics the government wants, Tencent's cooperation has gone to impressive new lengths, according to a new report released yesterday by The Citizen Lab at the University of Toronto.
The report details the ways Tencent ( tcehy ) censors keywords without users ever knowing; deletes images appearing on WeChat news feeds, which was previously undiscovered; and doesn't subject overseas users to the same onerous censorship as Chinese users.
Similar censorship exists on Sina Weibo, China's Twitter.
But WeChat, with 889 million monthly average users and a Facebook-like closed infrastructure that creates a sense of privacy, is becoming the more important platform for China's one-party government to monitor and shape.
Researchers tested censorship using terms related to the imprisonment of 250 human rights lawyers and workers starting in the summer of 2015. The episode earned the moniker "709 crackdown" for the first disappearances of two lawyers on July 9th, 2015. The topic has since gained attention inside and outside the country as the biggest crackdown on human rights workers since just after the Tiananmen Square massacre. In January, one lawyer's description of his treatment was made public. Im going to torture you until you go insane, human rights lawyer Xie Yang said one of his interrogators told him .
Citizen Lab found 41 keyword combinations related to the "709 crackdown" were censored on WeChat without users knowing. If they sent messages to friends containing the phrases, or posted them to their Moments "news feed," the messages appeared on their end but couldn't been seen by anyone else.
Previous research has found WeChat censors terms, such as those relating to the Hong Kong protests in 2014. A Tencent spokeswoman didn't respond to a request for comment Friday.
New in the latest report was that images are now being censored. In the past, images were a way to evade censors. Citizen Lab found infographics and profiles of the lawyers caught up in the 709 crackdown were censored in individual messages and Moments pages. "Our discovery of related blocked images on WeChat confirms the existence of image filtering and reveals the high level and extent of censorship enforced on this popular chat app," the researchers wrote.
More keywords and images were censored if they were sent to group chats containing up to 500 members than one-on-one exchanges, researchers said. But the same keywords were not censored on accounts registered to a phone number outside China.
The research shows how easily China's censors can co-opt the social network, as well as the consequences. "While there is tremendous effort and numerous global petitions to help Chinese rights defenders, many of these messages fail to reach domestic audiences in China due to information control practices, including search filtering and keyword and image censorship on chat apps," researchers wrote.

Exiled, Then Exalted: Agnieszka Holland on Communist Censorship, the Holocaust, House of Cards and Spoor – MovieMaker Magazine

She left Poland twice. First when she went to study at the Film and TV School of the Academy of Performing Arts in Prague (FAMU) on the verge of the so-called Prague Spring. Then when she fled to France before the martial law was imposed in 1981. She always came back.
Agnieszka Holland is the living legend of Polish cinema, one who worked with Krzysztof Zanussi, Andrzej Wajda or Krzysztof Kieslowski. Just like them she is known not to shy away from exploring political and moral issuesin her films shes tackled demons from the past such as the holocaust or communist oppression.
Outside her home country Hollands worked in France, Germany, Czechia or the U.S. where she directed episodes of The Killing or The Wire next to several feature films. After Oscar nominations for 1985s Angry Harvest and 1990s Europa, Europa she has been most recently awarded The Silver Bear Alfred Bauer Prize in Berlin for her latest film, an anarchistic-feminist crime story with elements of black comedy Spoor.
In March Agnieszka Holland came back to her alma mater in Prague where she received the honorary title of Doctor Honoris Causa and on the occasion held a master class.

I believe that its the wrong choice not to get involved. Being passive means you are active against freedom. That said, artists role in society is to make art. And its up to everybody to decide what it means exactly. I dont think it should be defined and specified. An artist should make art. And if he feels like it, he can be civically or even politically engaged. But thats up to him or her. It cant be forced.
I dont care for stereotypes. What I am interested in are the people, their fate and the complexity. Some would say that Poles are like that and the Germans are like this To me the holocaust is not a matter of Jewish-Polish relations or any other relations; its a matter of humanitys experience. Its all about the moment when we were forced to look into the asshole of mankind and realized that the extent of evil in man is immense. And that everybody is capable of evil. I dont see the struggle as inter-nationalafter all the Poles for example played the roles of victims and murderers at the same time. It is definitely about the inner struggle of a man.
With holocaust films I realized people always expect their portrait to be flattering. When you take somebodys picture so that it captures what he really looked like at the moment, he wont be satisfied. Make some changes and suddenly its a good picture. But good isnt true. The highest-grossing holocaust films in Germany were always those with a good German herosuch as Schindlers List or The Pianist. They didnt want to watch films with only evil Germans in them. Its the same with every nation.
I usually talk a lot with the actorsnot just about the role, also about their life experience and all sorts of things really. Sometimes we rehearse a scene or try a little improvisation. But thats before shooting and then, if the casting is done right and the actors trust you and know what the character is about, theres no more talking. In my experience, at that point it would do no good and it would only disturb them. Of course there might be a problem with a scene or a dialog and then its my work to make things clear and better. But usually the actor understands the character more than the director or even the screenwriter so the only thing I have to worry about is not to interrupt the actors inner emotional process.
I dont think there are that many differences. Sure, the medium is different. But quality TV is pretty similar to film. You just have to construct the episodes so that the audience wants to tune in again next week. Ive always loved working for television, its not boring like theatre where you have to rehearse and rehearse forever.
Its interesting to see how popular quality TVs become and how restorative and innovative it is. It could be compared to what happened in the 19thcentury with epic novelsthey were aswell serialized and very popular, be it works of Balzac, Stendhal or Dostoyevsky. The success of quality TV shows us that two hours are not enough for epic understanding.
In the U.S. the real author of a series is always a showrunner who plans and supervises everything. Director is just a hired help, unless hes an executive producer too. So when I am working on a project like that I have to realize that my film-craft is in service of something thats not just mine and I have to forget the usual authorship. I try to deconstruct the overall style of a series and adapt. But of course its never without the ambition to try and do something different or better. When I worked on the House of Cards I always tried to do things differently, use a hand-held camera or a steadicam. I wasnt allowed! And no zoom either. The Wire was something elseI could do pretty much what I wanted and the showrunner David Simon appreciated it enough to ask me to work with him on the Treme series. That would be a nice exercise for film studentsto look for differences between episodes of a multi-director series. Because at first sight there are none, yet the styles differ.
When I came back to Poland from Czechoslovakia in 1971, it was definitely hard but it was also a time of great solidarity among filmmakers, foreshadowing the establishment of the Solidarity movement in 1980. Young filmmakers collaborated tightly with the older ones such as Krzysztof Zanussi or Andrzej Wajda. For example when the authorities were against me working on Wajdas Man of Marble as an assistant director he offered to adopt me. Even if there was censorship I remember those times as the best years of my filmmaking life. And censorship was in a way inspiring as we had to encrypt certain messages that couldnt be stated directly. In connection to that I think the Polish audience was way smarter under the Communist regime than it is nowit was capable of decrypting complex metaphors and symbols. Nowadays there are people who can react to a murder on screen as if it was sort of an instruction, something to imitate. Comfort leads to mediocrity and film is no exception.
We shot every movie knowing it can be banned or censored. There were documentary filmmakers who made five films and all of them were banned. However, I think the ban for A Lonely Woman made the film popular. Even though it was available only on illegal underground VHSes and in a terrible quality, it had a bigger audience than it would without the ban. And weve shown it at least 200 times at our apartment in Paris. Later, my brother-in-law was able to steal a film reel of A Lonely Woman from the corridors of the Polish television building and Huub Bals, the first director of the International Film Festival in Rotterdam took it to Netherlands with him. There he made a 16mm copy of the film which I could screen at festivals.
Emigration was a tragical experience for me. I didnt want it and I had to reconstruct myself. First country I went to was France. Thats where I also started shooting again and I had the opportunity to make a few feature films there. However, there still was this feeling that I am a foreigner. Like if I was invited inside but never got into the living room. My idea of storytelling was different from the French one, too pragmatic and accessible maybe, more Anglo-Saxon than Romance. I guess thats why Americans liked it and after Europa Europa I started getting offers from them. I picked a movie based on a childrens book The Secret Garden. The shooting took place in England and casting was mainly child actorsI thought that way I could work out of sight of Hollywood executives and without big stars and their big demands. Well, I was wrong. I had to fight hard to make a movie I wanted to make. And my experience with Communist censorship was very useful in dealing with studio executives. Anyway, Ive had great experience with American film crews. When shooting in Poland, Germany or France I always have to shout in order to get what I want. In America I dont even have to raise my voice.
Wajda was somebody with his antenna constantly in receiving mode. Meaning he was always looking for things to read or watch, for people to meet, for ideas. Excited about history as well as the future He took a lot from his surroundings and other people. Certain people might even call it stealing but I wouldnt go that far.
Wajda was on my mind when I went shooting winter scenes for Spoor. Everything was ready except it hadnt been snowing. And then, as we started shooting, snow started falling. Andrzej Wajda used to say that good directors have weather luck. So Ive realized Im a good director at last.MM

In an unusual broadcasting flub this week, Iran's official state medianetwork cut off the live video feed of a reporter in mid-sentence when she mentioned the name of presidential hopefulHamid Baghaei, one of the more controversial politicians who has filed to run for president next month.
Baghaei served as vice president to former president Mahmoud Ahmadinejad. Both represent a frequently contested hardline political position, and both men now have registered to run for president in Iran's May 2017 elections.
The broadcast, which aired on the Islamic Republic of Iran Broadcaster's Shabakeh Khabar channel, featuring a reporter identified as Ms. Nouri, was shot on location from the Ministry of Interior on Wednesday as prospective candidates filed their registration forms.
A video clip of the incident has gone viral on social media.The video plainly shows that within less than a second of the the reporter mentioning Hamid Baghaei, the broadcast feed abruptly cuts and goesback to the anchor in studio. As the anchor waits to resume the report, Nouri's voice can be heard asking her producers, I shouldn't mention Baghaei?

Candidate registration for the 12th Presidential elections of the Islamic Republic of Iran began on April 11 andwill last for five days, followed by a period when the registrants will be screened for their political and Islamic qualifications by the hardline and religious body tasked with vetting, the Guardian Council. The Council typically disqualifies the majority of registrants.
The Islamic Republic of Iran Broadcaster (IRIB) is known as a mouthpiece forIran's hardline conservatives, echoing the concerns and opinions of this establishment. The IRIB's director is a position directly appointed by the country's highest power, the Supreme Leader Ayatollah Khamenei.
When it comes to reporting on elections, the IRIB's decisions about who to mention and who to omit are often seen as areflection of internal sentiment aboutthe prospective candidates. The interruption of Nouri's report isa sign that Baghaei may be disqualified from the running.
It is unknown whether Nouri already knew not to report on Ahmadinejad's registration as well, or if she wasdeterred post-facto.

Wednesday's registrations made headlines as former President Mahmoud Ahmadinejad, flanked by his close confidants formerVice President Baghaei andformer Chief of Staff Esfandiar Mashaei, stopped by the Ministry to announce that he and Baghaei hadregistered.
This is a change in course for Ahmadinejad, who inSeptember 2016 announced he would not join the race, after being warned by the Supreme Leader that his candidacy would cause polarization and harmful divisions in Iranian society. While also banned in 2013 from running for reelection, Ahmadinejad campaigned for the candidacyof Mashaei, whoultimately failed to win the Guardian Councils approval.
The IRIB is known for inserting bias an censorship into election reporting. During the 2013 presidential campaigns, several of the candidates remarked on the IRIB's unfair coverage and use of censorship. Rouhani accused the state broadcaster of defaming prominent figures during an interview on 27 May 2013, while the reformist candidate Mohamad Reza Aref's campaign accused the IRIB of cutting his campaign appearances in an inappropriate manner.
While the broadcaster is watched by millions of Iranians, it is notorious for not giving airtime to those who do not fall in linewith Khamenei and the clerical establishment, and for serving the interests of Iran's intelligence agencies, sometimes going so far as to air forced confessions of political prisoners under duress.

After Trump’s Syria Attack, What Comes Next? – Antiwar.com

Thursdays US missile attack on Syria must represent the quickest foreign policy U-turn in history. Less than a week after the White House gave Assad permission to stay on as president of his own country, President Trump decided that the US had to attack Syria and demand Assads ouster after a chemical attack earlier in the week. Trump blamed Assad for the attack, stated that somethings going to happen in retaliation, and less than two days later he launched a volley of 59 Tomahawk missiles (at a cost of $1.5 million each) onto a military airfield near where the chemical attack took place.
President Trump said it is in the vital national security interest of the United States to attack Syria over the use of poison gas. That is nonsense. Even if what Trump claims about the gas attack is true and weve seen no evidence that it is there is nothing about an isolated incident of inhuman cruelty thousands of miles from our borders that is in our vital national security interest. Even if Assad gassed his own people last week it hardly means he will launch chemical attacks on the United States even if he had the ability, which he does not.
From the moment the chemical attack was blamed on Assad, however, I expressed my doubts about the claims. It simply makes no sense for Assad to attack civilians with a chemical weapon just as he is winning his war against ISIS and al-Qaeda and has been told by the US that it no longer seeks regime change. On the verge of victory, he commits a suicidal act to no strategic or tactical military advantage? More likely the gas attack was a false flag by the rebels or perhaps even by our CIA as a last ditch effort to forestall a rebel defeat in the six year war.
Would the neocons and the mainstream media lie to us about what happened last week in Syria? Of course they would. They lied us into attacking Iraq, they lied us into attacking Gaddafi, they lied us into seeking regime change in Syria in the first place. We should always assume they are lying.
Who benefits from the US attack on Syria? ISIS, which immediately after the attack began a ground offensive. Does President Trump really want the US to act as ISISs air force?
The gas attack, which took some 70 civilian lives, was horrible and must be condemned. But we must also remember that US bombs in Syria have killed hundreds of civilians. Just recently, US bombs killed 300 Iraqi civilians in one strike! Does it really make a difference if you are killed by poison gas or by a US missile?
Whats next for President Trump in Syria? Russia has not backed down from its claim that the poison gas leaked as a result of a conventional Syrian bomb on an ISIS chemical weapons factory. Moscow claims it is determined to defend its ally, Syria. Will Trump unilaterally declare a no fly zone in parts of Syria and attempt to prevent Russian air traffic? Some suggest this is his next move. It is one that carries a great danger of igniting World War Three.
Donald Trumps attack on Syria was clearly illegal. However, Congress shows no interest in reining in this out-of-control president. We should fear any US escalation and must demand that our Representatives prohibit it. If there ever was a time to flood the Capitol Hill switchboard demanding an end to US military action in Syria, it is now!
