While theres certainly no shortage of trend outlooks for manufacturing technology spending based on near-term plans, looking at where industrial companies have been investing offers interesting insights into where things could be headed.

Source: Bank of AmericaAccording to Andrew Obin, managing director of Bank of America Merrill Lynch Equity Research, one of the biggest trends hes seen is how spending on software has overtaken hardware spending by U.S. manufacturers. In his presentation at the virtual ARC Forum, Obin noted that, although software spending in 2020 was flat, the fact that spending remained in line with 2019despite the pandemicsupports the investment firms position about the continued strength of software versus hardware spending in industry.

Essentially, what researchers like Obin are seeing play out across industry underscores the trend seen in virtually every business sectorthat software has become the predominant technology. Automation World discussed this trend with Michael Risse of data analytics software supplier Seeq in late 2019 for the article Is Software Eating Industry Yet?

Andrew Obin, managing director of Bank of America Merrill Lynch Equity ResearchObin clarified that much of the manufacturing industries spending on software in 2020 focused on enterprise level software, such as enterprise resources planning (ERP), customer relationship management (CRM), and supply chain management (SCM) than on specific manufacturing floor software. However, he noted that industrys spending on software supports one of the most powerful stories in U.S. manufacturing over the past 20 yearsthe significant margin expansion [of the industry] despite big offshoring trends in the U.S. [during that period]. Even though we didn't have growth in the U.S. [manufacturing industries], the [industrys] software investment has enabled very significant improvements in the profitability of U.S. manufacturing operations over the past 20 years as focus shifted away from growth and more toward efficiency.

Diverging from the manufacturing industries overall spending on technology is the lack of spending on such technologies by the energy sector. Obins data shows that, in 2019, manufacturing as a whole spent $228 billion on plants and equipment, while oil and gas spent $174 billion. More striking was the disparity in software spending between manufacturing and the oil & gas sector. In 2019 the oil & gas sector spent $8 billion on software while the manufacturing industry spent $134 billion.

Source: Bank of AmericaThe oil and gas industry has sort of done the opposite of the manufacturing industry, said Obin, noting how oil and gas spending on technology declined even as oil prices recovered. It's remarkable just how little oil and gas industry is spending today, Obin said. [But] if the oil and gas industry focused more on efficiency and less on growth in the new post-pandemic worldwhere we have more focus on green [initiatives]it could present a revolutionary opportunity for the industry.

Formulation intopical product development

The level of risk that a company takes largely affects their development strategy. This can vary between the development of prototype formulation (higher risk) or a full market-ready, commercially viable product (lower risk) to be used in initial preclinical/clinical evaluation. Different sizes of companies will base their own level of risk around commercially viability, their target markets, and corporate culture.

Typically, smaller biotech companies only have a single drug candidate and are often funded by external investment and therefore find the development of a prototype formulation more favourable. These small companies tend to be more risk tolerant too, addressing problems as they arise, so the drug candidate can be evaluated in a clinical Proof of Concept (PoC) study as quickly as possible.

However, this prototype formulation approach presents its own challenges. If the PoC study achieves a positive outcome then further reformulation work would be required to achieve a more patient-friendly and usable product, leading to extensive bridging safety studies. In turn resulting in the formulation development needing to be re-done to proceed to final Marketing Authorisation Application (MAA) or New Drug Application (NDA). Thus, the time and money initially saved in the early stages is often lost and/or exceeded later in the project.

Its important to emphasise that a topical products ease-of-use, aesthetic and cosmetic properties are often as important as efficacy. The selection of such a formulation for topical application is influenced by the physicochemical properties of the drug and its potency, the disease to which it is utilised, and the patient who will receive it (Figure 1).

Therefore, a Quality Target Product Profile (QTPP) should be created to define the key requirements around the quality, safety, and efficacy of the drug product. This should be updated as the project progresses where any Critical Quality Attributes influenced by Critical Material Attributes (CMA) and Critical Process Parameters (CPP) are identified, monitored, and controlled.

The pre-formulation stage of a new drug candidate is the most critical step for topical programs and is best performed using automated and robotic systems. The lowest risk approach to any submission is to try and keep to excipients, packaging, processes, and parameters with which the regulatory authority is familiar. A series of formulations can be developed based on the QTPP once the formulatability of a drug is known. Ultimately, it is essential that the lead formulation is optimised and characterised not only to demonstrate that it will maintain its quality and performance during its entire shelf-life but also gives the formulation the best chance of success in the clinic.

Performance testing: Innovation, automation and high throughput screening

Performance testing tools and models can be divided into four main areas: 1) ex vivo pharmacodynamic or disease models 2) in vitro (drug) release testing (IVRT), 3) in vitro (drug) penetration and permeation (IVPT) and 4) product characterisation and stability testing. These models provide a way to evaluate new chemical entities (NCE) and drug products, drug delivery, product safety, efficacy, and quality of topical formulations to reduce their risk of clinical failure.

The development of pharmacological disease (PD) models using fresh human skin has experienced huge growth thanks to the increasing interest and understanding in the basic biology of inflammatory dermatosis. Skin research has a huge advantage thanks to its direct access to large sections of surgical tissue. Latest advances in tissue culture have allowed scientists to keep surgical skin alive in culture for much longer than previously, creating a living tissue explant. By combining an increased understanding of immunology, tissue culture, and pathway biology these ex vivo skin PD models have become critical in the development of NCEs and are helping bridge the gap for clinical translation along with de-risking the development process for topical products.

IVRT is routinely used throughout the development process from the early stages of formulation optimisation, through process development, and to scale-up. Additionally, regulatory bodies are increasingly requiring IVRT as a quality tool in release and stability specifications and for demonstrating generic bioequivalence. The majority of industry adopt an IVRT method based on an open chamber vertical diffusion cell (VDC) system (Figure 2a) fitted with a synthetic membrane as a support for semi-solid dosage forms in order to measure and optimise the drug release from the formulation over time (akin to tablet dissolution testing).

However, a fully automated VDC system (MedStat-HT) (Figure 2b) differs from traditional manual VDCs by providing significant improvements in sample collection, data variation, operator repeatability, and study robustness.

Most of the industry uses similar manual VDCs for IVPT studies where the drug is applied to the skin, mounted in the VDC onto which formulation is applied and the absorption of the drug into and across the tissue is quantified. However, these systems are known for variability and are challenging for modern lipophilic drugs, making data interpretation problematic. MedFlux-HT, a fully automated flow-through diffusion system for use in such IVPT studies dramatically reduces the variability and more closely mimics the clinical situation compared with the manual VDCs (4).

Using automated liquid handlers and robots reduces the risk of variability when compared to manual procedures. When assessing longer-term physical stability, automated instrumentation (LUMiSizer) provides a more accurate prediction of shelf-life compared to harsher centrifugation techniques. Assessment of product microstructure is quite often overlooked until much later in the clinical phases of development. However, it is not only key in the development of generic products to show uniformity, but also during early phase process development to show that the developed product is robust.

Final thought

Whether via the eye, skin, lungs, nail, or other mucosal membranes these alternative topical routes clearly offer key advantages despite presenting unique challenges. A formulation strategy that builds the formulation specific to the compound will generate a solid foundation that proactively focuses on a final product that will be physically and chemically stable and ready for the clinic. It will also de-risk potential product failure that may come from an inability to deliver the compound to the target site or elicit a biological response prior to the clinic.

The challenge of integrating automation on ships – Ship Technology

Automation has been gaining traction among shipowners as an enabler of greater efficiency and cost savings. However, the president of Hglund Marine Solutions Brge Nogva believes that the industry is still a long way from exploiting its great potential.

A Norwegian provider of advanced marine technology (specifically, integrated automation and energy solutions), Hglund has long been an advocate of automation in shipping in Europe and beyond.

The company recently launched a partnership with DNV GLs independent data platform provider Veracity to help shipowners accelerate their digital initiatives. As Nogva explains below, these alliances are becoming an increasingly important part of the sectors transition to automation, but this could be delayed by a lack of uniform standards and regulation.

Maritime stakeholders have been preparing to adopt automation for some time, yet according to Nogva, their approach has so far been too slow and ineffective.

At the core of the issue is a string of inefficiencies that are affecting providers like Hglund, as well as shipowners, charterers and cargo owners. We provide quite traditional automation systems for traditional ships, he says. This is for us, in many ways, a dilemma, as we want to present better automation solutions, but the requests we get for our systems from the market arent really asking for much more than what has been done for years.

The slow shift towards digitalisation adds to several other issues, such as a decrease in the number of ships being built every year and different demands for automation from shipyards. The standard projects that come from shipyards in China and elsewhere tend to follow a very traditional specification that hardly says anything about automation, he explains.

On the other hand, there are more specialised projects in Europe where the requirement is to have a higher level of integration and a ship-to-shore solution, so that you can actually have data from the ship available from one shore-based office.

Inevitably, Covid-19 has helped decrease the number of similar projects taking place in Europe and bolstered a decline in ship orders that had already been hitting the market for the past five years.

Beyond their geographical variations, shipyard specifications are also posing technical challenges for system providers like Hglund. Whats frustrating is that we work a lot with shipowners and operators and tell them how they should write the specifications to bring about the kind of insight into the ships that they would like to add, but they dont get because of the low level of automation, says Nogva.

So, they will have to make a much better effort at developing the shipbuilding specification so that it actually describes in detail that there must be an onboard data server.

Owners now want to collect all this data and get access to it.

Having a centralised data server on a ship is an essential step, as this collects information from the different vessel systems and communicates them to the shore-based owner. However, because there are several systems on board and all of them speak different data languages and have different interfaces there needs to be an automated server that would work as, in Nogvas words, the nervous system of the ship.

He claims that retrofitting vessels with a data server is becoming an increasingly costly and tough operation. Owners now want to collect all this data and get access to it and those who will be able to fix it will be automation, engineers, and companies like us, who can do this because we are experts in dealing with interfaces so we can talk all these different languages, which is essential for actually making it happen, he adds.

Hglunds recent partner DNV GL is making significant steps forward in this context. They have now made new classification rules for shipowners who want to bring their ships to a more advanced level when it comes to not just automation but digital platforms, he comments, mentioning the companys cloud service Veracity and how it helps keep data stored in one place.

From this, if you manage to sort out all this data in a clever way, you can then create new services that shipowners will have the benefit of in the future, Nogva claims.

As markets start picking up after a disastrous 2020, a lot more will be expected in terms of compliance from shipyards.

There are many requirements that ship owners today have to comply with and these will develop further because of the International Maritime Organization (IMO) and the European Union making new rules, so owners will have to continue installing new systems or modify the ones they have on board, says Nogva.

It will be interesting to see if the owners are bringing new items into the ships specifications.

We are quite used to things going very slowly, though considering everything that is happening now, the specifications in the future should soon require a completely new level of automation system.

This transition will have to be facilitated and supported by regulators like the IMO and the EU. I am quite confident that if it is left to the shipowners and operators, they will not make many changes to this because they have to fight in a very hard market, says the Hglund president.

If were going to have a quick change, the requirements must come from the IMO and the EU who have to put legislation in place for us to comply.

The coronavirus pandemic has had devastating impacts on the global maritime industry, but one silver lining is that it may speed up the shift towards digitalisation.

The pandemic has shown the importance of making the most out of todays digital possibilities.

Hglunds expertise in providing remote access and modifications to ships has proved fundamental over the past year. This has shown several owners and shipyards of the possibility of reducing their costs, comments Nogva. Shipowners might well be eager now, but over the next few years, they will need more than mere interest to embrace automation. Charterers, cargo owners and the supply chain that serves the shipping sector will play a critical role, imposing their standards and demands to those they collaborate with.

[In the future] a stricter attitude from the cargo owners, charterers and authorities such as the IMO and EU will have to continue and rules will need to be more stringent, clearer and obliging to put systems in place that can actually monitor progress, he concludes. For now, this continues to go painfully slow in the maritime business when they actually need to shift from one technology to the other, and they need a proper push.
