What Do Your Tears Say About Your Health? – Chromatography Today

Some are happy, most are sad and others are just a result of chopping onions. Yes, tears. Psychiatrists could probably spend hours telling you what your tears mean for your mental health. But the little watery droplets are usually seen as fruitless for physical information. However, a team of scientists at Michigan Technological University may have changed that. Read on to see how tears can be used for medical testing.

Despite the perception of tears as empty and invaluable, they might contain important nutritional information. At present, the only way to access patients nutritional information, like vitamin levels, is through invasive (and expensive) blood tests. And it was this that led the team at Michigan Tech to their main question are there links between tear content and nutritional values in blood?

"We hypothesized that nutrients are transferred to the living cells of your cornea through your tears," explains Michigan Tech research specialist Maryam Khaksari. "We would like to translate the information we have for blood to tears. The link is certainly possible, with previous studies finding people with vitamin deficiencies blink more than those without.

To test the link, they focussed on three vitamins A, B and E. After taking samples of the tears and blood from 15 babies aged four-months as well as their parents, they used liquid chromatography and mass spectrometry to assess the content of the samples. Improved analytical development for this method is explored in the article, Capillary Flow LC-MS Unites Sensitivity and Throughput.

Overall, the results showed that levels of fat-soluble vitamins were higher in parents, while water-soluble vitamins were seen more in babies. There was a connection between the vitamin levels in parents and babies, with a correlation between vitamins B and E also shown by the results.

However, whats more significant is the promise this research shows in using tears for vitamin research. In this paper, we did show that there are correlations between vitamin concentrations in tears and bloodso it's possible," says Khaksari. The next step is to develop a device capable of performing these tests.

This project was the first step that proved vitamins are detectable in tears, that they do correlate with blood levels. Next we want to engineer a portable, lab-on-a-chip device relying on a minimally invasive sample from tears to assess nutrition, said Adrienne Minerick, professor of chemical engineering at Michigan Tech.

Looks like our parents were wrong when they said crying wont help.

Vitamin Research – All the latest research on Vitamins A …

We have gathered hundreds of published research on vitamins and presented them to you in plain English. See what the latest clinical research says about your favorite vitamins and the positive impact that vitamins have on disease prevention.

Thousands of new discoveries are made every single day, and you had better believe that no doctor has the time to keep up with all of the latest research, especially if they are a general practitioner and need to know about all of the body's systems and keep up with what the latest drug reports are. Who has time to read up on vitamins? Also keep in mind - no drug rep will ever visit a doctor to sell them a vitamin. There just isnt any money in it. That doesn't make vitamins any less valuable though. If nothing else, vitamins are one of the most beneficial supplements you could take. Literally hundreds of thousands of clinical studies on vitamins have been published, and the benefits of vitamins should not be ignored simply because nobody is directly selling them to you.

While a great many medical professionals read this site for information, we have "translated" every single of the hundreds of medical studies you see here into plain english so that the general public can - for the first time - be introduced to the latest cutting edge research that our world has discovered. With the power of the Internet, and the work of scores of people, this information is available to you completely free of charge.

Instead of wading through extremely thick and almost unintelligible clinical studies, or waiting ten or more years for research findings to reach your doctors office, take a look at what medical science has discovered now about your favorite vitamins and supplements. Feel free to print these out and show them to your medical professional, and ask them to do their own research. They may even learn a thing or two they did not know before. Remember, doctors are generally taught how to treat diseases with drugs (disease --> treatment) and they know the basics about vitamins, but they may not be aware of the powerful preventative effects that vitamins have on the body. Why would they, when they barely have enough time to see their patients in between their family life and visits with drug reps?

Try this the next time you are with your doctor. Ask them if they are aware of a strong link between high doses of vitamin C and vitamin E and a highly decreased incidence (occurrence) of Alzheimer Disease? If yours does, then you should be thankful that your doctor is more on top of things than most. Many doctor's simply don't know. Don't let what they do not know hurt your body. Afterall, nobody else looks out for your well-being as much as you do.

Vitamin – Methods used in vitamin research | Britannica

Determination of vitamin requirements

If a specific factor in food is suspected of being essential for the growth of an organism (either by growth failure or some other clinical symptoms that are alleviated by adding a specific food to the diet) a systematic series of procedures is used to characterize the factor.

The active factor is isolated from specific foods and purified; then its chemical structure is determined, and it is synthesized in the laboratory. Structural determination and synthesis, which may be achieved only after long and intensive research, must be completed before the function and the quantitative requirements of the factor can be established accurately. Established organic and analytical chemical procedures are used to determine the structure of the factor and to synthesize it.

Biological studies may be performed to determine functions, effects of deprivation, and quantitative requirements of the factor in various organisms. The development in an organism of a deficiency either by dietary deprivation of the vitamin or by administration of a specific antagonist or compound that prevents the normal function of the vitamin (antivitamin) often is the method used. The obvious effects (e.g., night blindness, anemia, dermatitis) of the deficiency are noted. Less obvious effects may be discovered after microscopic examination of tissue and bone structures. Changes in concentrations of metabolites or in enzymatic activity in tissues, blood, or excretory products are examined by numerous biochemical techniques. The response of an animal to a specific vitamin of which it has been deprived usually confirms the deficiency symptoms for that vitamin. Effects of deprivation of a vitamin sometimes indicate its general physiological function, as well as its function at the cellular level. Biochemical function often is studied by observing the response of tissue enzymes (removed from a deficient host animal) after a purified vitamin preparation is added. The functions of most of the known vitamins have been reasonably well defined; however, the mechanism of action has not yet been established for some.

The procedure for determining the amount of a vitamin required by an organism is less difficult for microorganisms than for higher forms; in microorganisms, the aim is to establish the smallest amount of a vitamin that produces maximal rate of multiplication of the organisms when it is added to the culture medium. Among vertebrates, particularly humans, a number of procedures are used together to provide estimates of the vitamin requirement. These procedures include determinations of: the amount of a vitamin required to cure a deficiency that has been developed under controlled, standard conditions; the smallest amount required to prevent the appearance of clinical or biochemical symptoms of the deficiency; the amount required to saturate body tissues (i.e., to cause spillover of the vitamin in the urine; valid only with the water-soluble vitamins); the amount necessary to produce maximum blood levels of the vitamin plus some tissue storage (applicable only to the fat-soluble vitamins, particularly vitamin A); the amount required to produce maximum activity of an enzyme system if the vitamin has a coenzyme function; the actual rate of utilization, and hence the requirement, in healthy individuals (as indicated by measuring the excreted breakdown products of radioisotope-labeled vitamins).

The above procedures are practical only with small groups of animals or human subjects and thus are not entirely representative of larger populations of a particular species. A less precise, but more representative, method used among human populations involves comparing levels of dietary intake of a vitamin in a population that shows no deficiency symptoms with levels of intake of the vitamin in a population that reveals clinical or biochemical symptoms. The data for dietary intakes and incidence of deficiency symptoms are obtained by surveys of representative segments of a population.

A quantitative analysis of the vitamin content of foodstuffs is important in order to identify dietary sources of specific vitamins (and other nutrients as well). Three methods commonly used to determine vitamin content are described below.

The amount of vitamin in a foodstuff can be established by studying the physical or chemical characteristics of the vitamine.g., a chemically reactive group on the vitamin molecule, fluorescence, absorption of light at a wavelength characteristic of the vitamin, or radioisotope dilution techniques. These methods are accurate and can detect very small amounts of the vitamin. Biologically inactive derivatives of several vitamins have been found, however, and may interfere with such determinations; in addition, these procedures also may not distinguish between bound (i.e., unavailable) and available forms of a vitamin in a food.

Defending Vitamins: What’s wrong with the science? – Nutritional Outlook

Lets face it. Vitamins have generally not fared well in recent years when it comes to high-profile randomized controlled trials (RCTs) or meta-analyses that statistically combine selected data from those studies.

There have been some notable exceptions. For example, the AREDS trials found a specific combination of vitamins reduced the risk of age-related macular degeneration, and the Physicians Health Study II suggested a modest benefit that significantly reduced the risk of total cancer in the study population.

There have also been secondary outcomes in clinical trials that have produced positive findings worth exploring further, as is the case in the VITAL trial that suggested reduction in the rate of cancer deaths for the study population taking vitamin D.

But by and large, RCTs hypothesizing that vitamins will prevent chronic disease have been a swing and a misssomething disappointing to everyone from study funders to researchers and from the supplement industry to consumers.

Does this signal the end in the vitamin research wars, with a white flag raised by the supplement industry? Not by a long shot. Instead, its time to refresh the arsenal.

Whats Null Got to Do With It

Consumers are sticking with vitamins. With supplement use at an all-time high according to the 2019 Council for Responsible Nutrition (CRN; Washington, DC) Consumer Survey on Dietary Supplements, vitamins/minerals continue to be the most commonly consumed supplement category, with 76% of Americans taking them annually. The multivitamin (58% percent of Americans take them) and vitamins D, C, B, and E are all firmly planted in the list of most popular supplements in CRNs survey.

When it comes to the science, preeminent nutrition researcher Jeffrey B. Blumberg, PhD, FASN, FACN, at the Friedman School of Nutrition Science and Policy, Tufts University (Boston, MA), has a lot to say on this topic.

To begin with, Blumberg offers, No investigator I know wants to come out with a null outcome from their studies. Investigators would like to see something positive.

Its understandable that researchers want to swing for the fences, hoping to make their mark with that home-run clinical trial that can elevate their reputation and help secure future grants. But given the multifactorial nature of chronic diseases like cancer and heart disease, why would anyone expect that a simple vitamin pill or even a combination of vitamins would, on their own, prevent cancer?

No one disagrees that nutrient-deficient diets are a risk factor for chronic disease, but theyre just one risk factor, says Blumberg. Lets look at the normative state of Americans today66% are overweight or obese. Therefore, I could be taking 10 supplements a day, but if Im also obese with a BMI of 35 and smoke two packs of cigarettes daily, it should come as no surprise that those supplements wont work to prevent chronic disease.

Its asking way too much of a supplement, he concludes.

Does the Past Predict the Future?

So why conduct these trials? Why spend the money and the time and risk the disappointment?

Blumberg explains that clinical trials build on prior research. He says, In order to justify doing a clinical trial, youre saying there is evidence that this intervention could work based on earlier research, fromin vitrostudies, animal models, observational studies, and maybe some small clinical trials. In other words, theres been some hint of benefit that would warrant the use of sponsor funding and all the time and effort involved from a research team to take the next step to the large RCT. No one does an RCT without evidence that there could be a benefit, he points out.

However, if the RCT doesnt confirm that benefit, there shouldnt be a rush to deny the prior benefits discovered. Says Blumberg, Thats a pretty big condemnation of all research approaches except for one. Maybe theres something limited about that gold standard of an RCT in that its not truly able to measure the benefits that were found in all of the other studies that justified that RCT in the first place.

Vitamins Are Not Drugs

Blumberg continues, RCTs are a good model to test drugs because they can be done quickly, effectively, and efficiently because if your drug doesnt work in one or two years, youre not going to even market it.

He sees a potential disconnect in the financial viability and long-term researcher commitment that could result in some success for vitamins and chronic disease. He asserts that it takes 20-30 years for cancers and heart disease to develop and that nutrients work best over a lifetime. To my knowledge, there has never been a 20-year RCT on any dietary supplement. What investigator wants to put in 20 years or more before they get their final results? he asks.

To compound the issue of studying vitamins and chronic disease prevention and considering how any positive results might be communicated to consumers, there is also the matter of a mandated government disclaimer for product labels using certain categories of claims not pre-approved by FDA. Blumberg gets the irony. If supplement product labels need to state the product is not intended to diagnose, treat, cure, or prevent any disease, Blumberg wants to know why is all this research focused on disease?

The Wrong Question

According to Blumberg, A big part of the problem has been asking the wrong question. It sounds like a reasonable one: how do dietary supplements prevent non-communicable diseases; which nutrients prevent cancer, heart disease, osteoporosis, COPD, you name it, he says. But with the critical limitations of using RCTs to test nutrients, its probably not the right question, and its likely not even what consumers are looking for a vitamin to do.

That doesnt mean Blumberg suggests eliminating all RCTs for future nutrition research. From his perspective, its not appropriate to tell any researcher how or what research he or she should conduct. He does, however, have some forward-thinking research considerations for studying vitamins that link new technology with improving the quality of lifetwo areas he thinks will make a difference.

Resetting the Bar for Science

One of the areas to consider is how the changing scientific landscape will reset the stage of nutrition and vitamin research.

According to Thomas Li, business development manager for IFF Health (New York City), an ingredient supplier and the manufacturer of the Grow Nutrients line of vitamins and minerals, Science is key for us because it aligns with our message of being premium and high quality; we want to prove our ingredients work. In this age of transparency, having new data is crucial to staying relevant.

Still, he contends: While it is true that many well-known ingredients with long histories of traditional use usually have a lot of science behind them, many product claims still rest on outdated science. With the emergence of new technologies and personalized nutrition, the bar for scientific research should be set higher.

The growing influence of personalized nutrition could change the research in numerous ways. Blumberg is not only energized about what that science means for individuals; hes energized about how that discipline can translate into clinical research.

Blumberg is so energized, in fact, that he co-authored a paper on personalized nutrition for the American Nutrition Association (ANA; Hinsdale, IL), for which he serves as the Board Chair, published in December 2019 in the Journal of the American College of Nutrition. The paper proposed a definition of personalized nutrition as a field that leverages human individuality to drive nutrition strategies that prevent, manage, and treat disease and optimize health.

If our diets are poor (and they are), says Blumberg, will taking vitamins help improve structure or function? I think the evidence is there that it does.

The Future of Research

In addition to research evolving in these ways, changing attitudes could also help improve the way we study these nutrients.

For instance, improving the daily quality of life may or may not lead to a risk reduction for chronic disease, but if improving how we feel improves our health right now, that may be the right outcome.

Says Blumberg, Its an outcome we can measure. These kinds of studies are doable because I dont need to do a 10-year study to see if my immune responses, my cognitive performance, my muscle strength, or my vascular reactivity are improving. I dont even need 25,000 people to do it right. Im using the individual as his or her own control, as well as doing a group analysis. I can design a parallel design or a crossover RCT design looking at a physiological response or structure response.

He points to an observational study which he co-authored that published online in October 2018 inScientific Reports. In that study, the researchers analyzed data from a personalized nutrition company that provided results from 1032 individuals who had a personalized nutrition analysis and agreed to allow their anonymized data (baseline through follow-up tests after incorporating suggested nutrient interventions) to be analyzed. What we learned about them en masse is that their numbers changed, said Blumberg. We saw vitamin D levels go up, and we saw LDL cholesterol levels and glucose levels go down.

Using technologies that study the omics sciences (nutritional genomics, proteomics, metabolomics, etc.) allow for measuring thousands of metabolites at a time and is enormously powerful, he says. I can look at a nutrient with not just one outcome, but clusters of outcomes.

Hes realistic that while the possibilities for all these new technologies are endless, the costs may currently be a deterrent. But hes not personally deterred. Fifteen years ago, it cost about $1000 to test one genome; now its about $100.

More Sophistication in Science

Blumberg strongly believes that predictive analytics will also play a role. Now were using machine learning and artificial intelligence. I can measure 1000 things in your body through the use of omics to make predictions, but thats too expensive. Can I actually look at just 10 things and make predictions? Machines can take massive amounts of data from massive amounts of people and look for patterns. People are building these tools now and trying to understand how only a limited set of biomarkers are needed to tell you something about the state of your health and wellness to make an impactful prediction. This will work for vitamins and other nutrients.

Blumberg says that RCTs will be better designed when we have better biomarkers and when there are also novel statistical analyses to analyze observational data in a manner more like a clinical trial.

Our sophistication in looking at molecular and biochemical outcomes is getting much better in doing assessments and making predictions, but at the same time people doing the biostatistics are developing more sophisticated algorithms and approaches than we used to have before, he says.

Science Counts

Even as there is hope for future research, there is another dilemma the supplement industry faces: how best to get the research funded.

Nutrition research is not well-funded by the government, and academic institutions arent generally ponying up funds. Nor does there appear to be many (or any) Mike Bloomberg-esque scientists able to pull out personal checkbooks to fund their own studies. This is where industry can and should step in.

IFF Health prides itself on the clinical science for our branded ingredients to prove and support the claims made in marketing. We have a robust clinical pipeline to keep enhancing this. Not only do we want to make sure our science is updated to be relevant with what the industry is demanding, but also to find new unique features to pass on to our customers. We conduct our own research, but also partner with customers on clinical studies as well as a value added to our services.

Li advises IFF Health is always looking to support our customers in joint studies utilizing their finished products with our ingredients, as he sees a big gap in the lack of finished products being used in science.

Darned If You Do, Darned If You Dont

Herein lies the quandary: While companies that support nutrition research should be commended, considering the general lack of funding, that research or funding is often discounted by the medical journals and not always welcomed by the consumer press because it comes from the industry.

Taylor C. Wallace, PhD, CFS, FACN, at George Mason University, and editor-in-chief at the Journal of Dietary Supplements, worked in the supplements industry for a short time and now runs Think Healthy Group, a food and science nutrition consulting firm with expertise in developing and executing research to support product claims, policy decision-making, and educational campaigns. Wallace insists that transparency disclosures should be standard practices and are used by most researchers who are honest, credible, and transparent. He says that accusing a scientist of industry funding or industry bias implies that the scientist somehow deviated from standard procedure or either manipulated or fully fabricated data.

Wallace says he doesnt know any credentialed scientist in the field that would even vaguely consider manipulating or fabricating data to suit an industry sponsor, adding that act would be career-ending, extremely unethical, and embarrassing.

Blumberg, who was the director of the Antioxidants Research Laboratory at the Jean Mayer USDA Human Nutrition Research Center on Aging at Tufts University, has been an academic researcher for 44 years, beginning at Bostons Northeastern University and joining the Tufts University faculty in 1981. Hes received research grants from the food and dietary supplements industries, served on industry scientific boards, and accepted honorariums to speak at industry conferences. He is also highly regarded in the scientific research community. Yet, hes felt the discrimination that comes with working with industry and says hes not special in that regard.

You are tainted if you take money from a company or from a commodity board, even if its a non-branded thingsomehow its thought you cant be objective, says Blumberg. How can I do the research if I dont get the money to do it?

He gets the catch-22 of it allfor himself and for the supplement industry. You want to blame the supplement companies for not having evidence that their products work, but if they fund the studies to get the evidence, then you cant believe the evidence because the company funded it.

No one questions that drug companies shouldnt be funding their own clinical trials. Its a conundrum that food and vitamin companies are demonized for supporting research that would provide evidence for their products. I dont know how we ever get over the hump except to change attitudes, Blumberg says.

Can We Change Attitudes?

Only time will tell. Wallace thinks its time for nutrition scientists to break away from using protocols, tools, and research methods that were designed for studies of drugs, noting that drugs and nutrients (or other bioactive constituents in food) are not the same.Blumberg advises: What were really talking about is promotion of health. Theres a difference between not-sick and sick, and a difference between not-sick and well.

He explains that while someone may not be sick with a chronic disease, that doesnt mean theyre well. What hes interested inand what he believes consumers are interested inis the acute effect of vitamins in terms of physiology and structure or function.

I might not have cancer or heart disease or dementia, but lets say my immune system is sluggish, my energy levels are low, I dont sleep well, Im just not alert as I could bethese are things that are impacted by nutrient intake. These are measurable outcomes, and we know that nutrients have impacts on these outcomes, says Blumberg.

If we can learn to think about vitamins this way, the world would find a whole new value for vitamins and supplements.

Michael Mosley investigates the links between diets and disease – SBS

The you are what you eat connection between food consumption and health outcomes is so well-established its practically irrefutable. Search diet and disease on PubMed (the go-to database for scientific research) and youll get over 150,000 results and thats just one search term. Theres robust evidence that a diet rich in fruit, veg, wholegrains, fish and nuts can dial down your risk of heart disease, Type 2 diabetes, cancer and stroke, while dietary villains like soft drinks and processed meat do the opposite. You can drill down further, and link specific nutrients to health outcomes for instance, we know insufficient iron causes anaemia, zinc promotes immunity, while magnesium regulates blood pressure.

Still, our knowledge wasnt always at this level its taken some fearless scientists with a keen interest in self-experimentation to prove that what we ingest impacts our disease risk, as British doctor and science presenter Dr Michael Mosley reveals in episode three of Medical Mavericks.

Its hard to believe, but back in the 18th century, Mosley tells us, the human body was simply a machine which burnt fuel and it really didnt matter what the fuel was. Then, along came Dr William Stark, who in 1769, disproved this. To discover which foods were good and which were bad, he put himself on a diet of bread and water, adding in large amounts of unhealthy foods such as honey pudding, one by one. It was his decision to add Cheshire cheese next, instead of fruit, which ultimately led to his death from scurvy. He might have gone on to become one of the early pioneers of vitamin research, instead he became the first martyr of nutritional science, says Mosley.

Another case of self-experimentation that Mosley investigates in the episode is that of late scientist Dr Hugh Sinclair. Convinced that heart disease was caused by eating too little rather than too much fat, he put himself on an Inuit diet rich in oily fish and seal blubber. It was a perilous experiment: while fish oils produce less cholesterol than saturated fat, consuming them in excess can interfere with blood clotting. He would have had a risk of a severe gastric bleed on that diet and possibly having a stroke, says Professor Tom Sanders from King's College London, who appears in the episode.

It was his decision to add Cheshire cheese next, instead of fruit, which ultimately led to his death from scurvy.

After three months on the diet, Sinclair's blood had thinned significantly from the diet - from four minutes to a whopping 50. Hed finally proven the potency of essential fatty acids, Mosley tells us.

Calorie restrictors are other self-experimenters under Mosleys microscope in this episode. These sticklers for self-denial dramatically slash their energy intake, with the pay-off being reduced oxidative stress, which in turn protects against age-related disease.

According to a 2019 Lancet Diabetes & Endocrinology study, people who stuck to a calorie-restriction diet for two years had a persistent and significant reduction in cardiometabolic risk factors, plus improvements in C-reactive protein (a marker of inflammation), insulin sensitivity and metabolic syndrome scores.

There is a core of probably 50 people who are doing a real experiment on themselves because they are the people who are really convinced that calorie restriction will allow them to live longer, says Dr Luigi Fontana from the Italian National Institute of Health in Medical Mavericks. In the episode, Mosley meets calorie restrictor Dave Fisher, who has been living on less than two-thirds of the normal calorie intake for 17 years. Both men go through a batch of health tests (such as sight, hearing, lung capacity and reaction times, all of which deteriorate with age), with Fisher coming up trumps. Thankfully I did beat him with my lung function test, Mosley notes with relief.

The doctor takes on a fish-eating marathon. Source: Sharyn Cairns

As a self-professed human guinea pig, Mosley cant resist an extreme experiment of his own in this episode, putting himself on a two-month-long fish binge, involving bountiful oily fish and fish oil. Its claimed eating oily fish reduces the risk of blood clotting, declares Mosley. So at the start and end of my experiment, I'm going to measure my bleeding time.

His initial bleeding time clocks in at four minutes and 11 seconds. Skip to the end of the experiment and it nearly doubles, to eight minutes and 12 seconds. It means Im much less likely to develop blood clots which in turn means Im much less likely to have a heart attack, he says. Thankfully, theres no need for the rest of us to dabble in kooky experiments: a balanced, wholefoods-based diet is pretty hard to go wrong with.

Dr Michael Mosley explores the ways in which pioneering doctors laid the foundations of modern medicine by experimenting on themselves.Michael mosley: Medical Mavericks, Diets & Disease airs on Monday, March 16 at 9:40pm on SBS and then it will be available via SBS On Demand.

Vitamin Research – UNIV OF WISCONSIN

Progress 10/01/06 to 09/30/11

Outputs OUTPUTS: The overall focus of this work is the study of the fat-soluble vitamins A and D. Ongoing work in three project areas is described below. Project 1. Both nutritional and genetic approaches are being used to determine how vitamin A functions in developing embryos, neonates and adults. Our laboratory has developed animal models to study vitamin A function in both early and late embryonic processes, as well as during postnatal life. We have shown that vitamin A is required for the initiation of meiosis in both the female and male gonad. Additionally, we identified several retinoic acid responsive genes that are expressed in the nervous system of developing embryos and in the adult brain. One of these, neuron navigator 2 (Nav2 or retinoic acid-induced in neuroblastoma, RAINB1), plays an essential role in neurite outgrowth, and in the development of the cranial nerves, eye, and brain. Project 2. Cellular and in vivo approaches are being used to address how the 4-HPR analog, 4-hydroxybenzylretinone (4-HBR), exerts its antitumor effects. In collaboration with our colleagues at the OSU, we have shown that the carbon-linked 4-HPR analog, 4-hydroxybenzylretinone (4-HBR), does not need to be hydrolyzed back to atRA to function in culture or in vivo. Mechanistic microarray studies are underway to determine how these compounds work in cultured breast cancer cells and mammary tumors. Project 3. Additional studies in the lab are directed at developing improved vitamin analogs for the treatment of acne and other disease states. Using the Rhino mouse as a model, we have identified a unique subset of vitamin D analogs that induce comedolysis. Studies are underway to determine how vitamin D compounds act to produce these effects. PARTICIPANTS: Jamie Ahrens (Senior research specialist), Elizabeth McNeill (Research associate), Danielle Knutsion (Research associate), Allyson Anding (Research assistant), Melissa Rohrer (Research associate), Mark Marzinke (Research assistant), Nirca Nieves (Research associate), Jim James (Associate research specialist), Mariana Bormann (Associate research specialist) TARGET AUDIENCES: Not relevant to this project. PROJECT MODIFICATIONS: Not relevant to this project.

Impacts The reproductive efficiency and health of animals, including humans, is clearly influenced by nutritional status. Rather than striving only to prevent overt nutrient deficiency, we need to begin to develop an understanding of "optimal" levels of a given nutrient to promote health. A detailed understanding of the physiological and molecular processes for which vitamins are needed is essential not only for optimizing human health, but also for determining how vitamin derivatives might be used in the treatment of human disease. For example, vitamin A compounds have shown promise in the prevention and treatment of cancer, as well as in the treatment of acne. However, the toxicity of many retinoids limits their use. Similar potential exists for the use of vitamin D analogs in a number of diseases, but the side effect, hypercalcemia, has been a major limitation to the advancement of these efforts. Thus, studies of new vitamin A analogs and non-calcemic vitamin D analogs may offer new approaches to the treatment of several disease states, including acne and cancer. Numerous patents that have issued or have been filed through the Wisconsin Alumini Research Foundation have arisen from these studies.

Outputs OUTPUTS: The overall focus of this work is the study of the fat-soluble vitamins A and D. Ongoing work in three project areas is described below. Project 1. Both nutritional and genetic approaches are being used to determine how and when vitamin A is needed for developing embryos, neonates and adults. Our laboratory has developed methods that enable the study of vitamin A function in both early and late embryonic processes, and more recently during postnatal life. When limiting amounts of atRA are provided after embryonic day (E) 10.5 in the rat, a highly reproducible and penetrant state of late fetal vitamin A-deficiency (VAD) is produced in the organs of developing fetuses. Using this method, we have recently shown that vitamin A is required for the initiation of meiosis in the female gonad. In addition, we have published a detailed characterization of the molecular events that are perturbed in the developing eye when insufficient vitamin A is present. We are also studying the function of several novel atRA responsive genes, and have shown that one of these genes (RAINB1 also known as Nav2) plays an essential role in neurite outgrowth, and in the development of the cranial nerves, eye, and brain. Further, 4-HBR is less toxic than 4-HPR, and appears to act by inducing ER stress-mediated responses. Work to further elucidate the cellular pathways that are altered by 4-HBR is underway, as are studies of its effects in developing embryos. Project 3. Additional studies in the lab are directed at developing improved vitamin analogs for the treatment of acne. Using the Rhino mouse as a model, the ability of compounds to produce a reduction in comedone size is being studied. Whereas all vitamin D analogs appear to induce thickening of the epidermis, we have identified a unique subset of analogs that also induce comedolysis. Studies are underway to determine how vitamin D compounds act to produce these effects. Outputs OUTPUTS: The overall focus of this work is the study of the fat-soluble vitamins A and D. Ongoing work in three project areas are described below. Project 1. Both nutritional and genetic approaches are being used to determine how and when vitamin A is needed for developing embryos, neonates and adults. Our laboratory has developed methods that enable the study of vitamin A function in both early and late embryonic processes, and more recently during postnatal life. Our recently published work shows that when limiting amounts of atRA are provided after embryonic day (E) 10.5 in the rat, a highly reproducible and penetrant state of late fetal vitamin A-deficiency (VAD) is produced in the organs of developing fetuses. This work shows that a large number of organ systems are adversely affected, including the eye. The optic fissure does not close, and severe folding and collapse of the retina is observed at E18.5. In our most recent work, we show that coloboma and retinal folding represent distinct VAD-dependent effects. The downregulation of Pitx2 in VAD may be related to the failure of the optic fissure to close, whereas a reduction in cyclin D1 and the cell adhesion molecules, N-cadherin and B-catenin, may play a role in the retinal thinning, folding and collapse observed in late VAD fetuses. We are also studying the function of several novel atRA responsive genes, and have found that one of these genes (RAINB1) plays an essential role in neurite outgrowth, and in the development of the cranial nerves, eye, and brain. Project 2. Cellular and in vivo approaches are being used to address how the 4-HPR analog, 4-hydroxybenzylretinone (4-HBR), exerts its antitumor effects on cancer cells. In collaboration with our colleagues at the OSU, we have found that the carbon-linked 4-HPR analog, 4-hydroxybenzylretinone (4-HBR), does not need to be hydrolyzed back to atRA to function in culture or in vivo. Our studies also indicate that 4-HBR is less toxic than 4-HPR. Recently, we reported that 4-HBR rapidly induces ER stress-mediated responses and this may represent a mechanism whereby this compound produces its chemotherapeutic effects. Work to further elucidate the cellular pathways that are altered by 4-HBR is underway. Studies of additional nonhydrolyzable compounds, including glucuronide and glucose derivatives of 4-HBR are also ongoing. Project 3. Additional studies in the lab are directed at developing improved vitamin analogs for the treatment of acne. Using the Rhino mouse as a model, the ability of compounds to produce a reduction in comedone size is being studied. We have identified a specific subset of vitamin D analogs that are effective in this model. Studies are underway to determine how vitamin D compounds act to induce comedolysis. PROJECT MODIFICATIONS: Not relevant to this project.

Impacts The reproductive efficiency and health of animals, including humans, is clearly influenced by nutritional status. It is becoming abundantly clear that rather than striving only to prevent overt nutrient deficiency, that we need to begin to develop an understanding of "optimal" levels of a given nutrient to promote health. Thus studies of new vitamin A analogs and non-calcemic vitamin D analogs may offer new approaches to the treatment of several disease states, including acne and cancer. Numerous patents that have issued or have been filed through the Wisconsin Alumini Research Foundation have arisen from these studies.

Outputs OUTPUTS: The overall focus of this work is the study of the fat-soluble vitamins A and D. Ongoing work in three project areas is described below. Project 1. Both nutritional and genetic approaches are being used to determine how and when vitamin A is needed for developing embryos. Our laboratory has developed methods that enable the study of vitamin A function in both early and late embryonic processes. Using this model, we have shown that vitamin A plays a critical early role in patterning the heart, hindbrain and axial skeleton. Our recently published work shows that when limiting amounts of atRA are provided after embryonic day 10.5 in the rat, a highly reproducible and penetrant state of late fetal vitamin A-deficiency (VAD) is produced in the organs of developing fetuses. These fetuses also display anteriorization of cervical regions of the skeleton and novel posteriorization events at the thoracic and sacral levels, and also show sternal and pelvic malformations not previously observed in genetic models. We are now using these nutritional models to explore the molecular mechanisms that are disrupted by embryonic vitamin A-deficiency. We are also studying the distribution and function of novel atRA responsive genes, and have found that one of these genes (RAINB1) plays an essential role in neurite outgrowth. Project 2. Cellular and in vivo approaches are being used to address how the 4-HPR analog, 4-hydroxybenzylretinone (4-HBR), exerts its antitumor effects on cancer cells. In collaboration with our colleagues at the OSU, we have found that the carbon-linked 4-HPR analog (4-HBR) does not need to be hydrolyzed back to atRA to function in culture or in vivo. Studies of the efficacy 4-HBR in chemotherapy of DMBA-induced mammary tumors in the rat show that the non-hydrolysable analog is equally effective in reducing tumor size and number, and does so without reducing blood retinol levels, an undesirable side affect associated with the use of the parent compound, 4-HPR. Although 4-HBR can bind and transactivate RARs, this is not required for it to induce cell death. Recently, we reported that 4-HBR rapidly induces ER stress-mediated responses and may represent a mechanism whereby this compound produces its chemotherapeutic effects. Project 3. Additional studies in the lab are directed at developing improved vitamin analogs for the treatment of disease. One example is the use of retinoids for the treatment of acne. A major limitation to current oral therapy is teratogenic liability. As a potential improvement, we are studying sterically hindered vitamin A analogs. Using the Rhino mouse as a model, the ability of compounds to produce a reduction in comedone size is being studied. We have also found that some vitamin D analogs are effective in this model. Studies are underway to determine whether these approaches may offer any advantage to existing therapies. PARTICIPANTS: J. Ahrens, Research Specialist M. Vitamin Research - UNIV OF WISCONSIN

Carson City nutrition factory to collaborate education with products – Nevada Appeal

Maria Watson wants Carson City to know she's back in business and is determined to bring the community quality health education, along with the relaunch of her company.

Formerly known as Vitamin Research Products, the CEO and president is reintroducing her natural vitamin supplement line as American Nutritional Products after a two year hiatus.

From stress relievers to belly busters, Watson's multivitamin products are manufactured with formulas containing plant-based ingredients, FDA approved.

Now that she's returned, she hopes to connect with the local community such as schools, churches, and other organizationsto teach the benefits of a healthy lifestyle and how to achieve it, while promoting her product.

Specifically, she hopes to target families and single parents, as nearly a third of the country spends dollars on fast-food and restaurant services, according to USDA.

Whether seminars take place at community colleges, health centers, or even at her facility, Watson hopes to fulfill one's health goals and produce new jobs in the area.

"It's been a painful and long journey," she said. "But this community helped us the first time and I want to educate as a way to give back."

When her company used to be Vitamin Research, Watson and her husband, Ron, supervised the facility on Highway 50 East in 2012, before constructing a larger building at 4610 Arrowhead Drive.

Back then, the company employed 127 people and packaged at least 3,000 products per day.

However, regulatory climate became a concern, to a point where the private equity group supporting Watson's company announced closure in July 2015. That closure led to the closure of Vitamin Research.

Now with 23 returning employees, Watson is preparing to get her manufacturing business up and running under the new name.

"My dream came true, but the rooms are empty." she said.

As for Watson's longtime customers, they're elated over the news of the original founder's return.

Jana Kay of Reno said she appreciates the natural ingredients the company uses in the multivitamins, as she couldn't find desired quality products in grocery stores or pharmacies.

"She's on a mission and wants to educate peoplewe need to educate people. That's why I was inspired to study nutrition and psychology in college, and learn about eating disorders within mental health."

Although Watson is a nutritional guru, what truly inspired her to get into the industry was her son, Josh, whom died at the age of 3 in 1987.

"He died in a car accident caused by a drunk driver, his babysitter," she said. "When he died, I gained 199 pounds and went through depression. But I knew I couldn't be like this foreverI believed I could change the world with my passion, and I still feel that way."

Watson is developing a plan for international collaboration. She connected with Deborah Torres Patel, a high prolific communication coach based out of Singapore.

Patel is known for mentoring best selling authors, entrepreneurs and leaders of Fortune 500 companies, and numerous celebrities in 79 countries.

Patel has never been to Carson City until last month when she met with Maria to help her develop a draft on how to combine health and education within the company.

One of Watson's ideas is to teach ways to relieve stress and anxietywith public speaking being a start.

"84 percent of first-time impressions is based on your tone of voice," Patel said. "It also affects how you feel and that's why I created my system to find your voice; you only get one chance to make a first impression."

Patel and Watson also discussed ways to collaborate education with new products, such as consumables or water with a natural ingredients to support energy boosts.

"It's about essential integrity and work," Watson said. "Why not amplify that in Carson City? People that live here are active. I want to educate those that will listen."

Watson advises the public to watch for future job announcements as the company grows.
