Researchers at Harvard Medical School have successfully restored vision loss and reversed glaucoma-induced damage in mice.

In the mice, the retinal ganglion cells, a principal cell that enables vision, were restored to a youthful state in cases of glaucoma, as well as when the optic nerve, another key component of eyesight, had been damaged. Both were achieved through expressing certain transcription factorsproteins that turn genes on and off.

The study sheds light on the mechanisms of ageing, and identifies new potential therapeutic targets for age-related neuronal diseases such as glaucoma, reads a statement from researchers at Harvard Medical School.

The new study, published in Nature, was conducted by Dr. David Sinclair, of the worlds foremost experts on ageing-related research in mice.

Along with genetic research, Sinclair has also looked at how supplement-ready compounds like resveratrol and Metformin affect aging, and his book, Lifespan: Why We Age and Why We Dont Have To, is a New York Times bestseller.

The science behind Sinclairsnew paper involves the curious process of methylation. Governed by epigeneticsthat is, changes in the genetic expression of the cell over timethe researchers found that methylation in mammalian tissues prevents the cells from replicating proteins properly while simultaneously encoding a kind of genetic history.

One can imagine this as scratches on the bottom of a CD. If the scratches could be removed, the record of proper function is still there, and could still be read by the laser in a CD player.

In his book, Sinclair details the modern theory of aging, which is that changes in epigenetics and damage to cells and tissues prevent the body from properly reading protein-encoding genes, resulting in either faulty, less-functional, i.e. older genes being transcribed, or the proteins not being replaced at all.

Here the authors found that when the mouse neurons were recovering from damage related to glaucoma, the methyl groups which built up over time left, like the scratches being removed from a disk.

This resulted in a process called demethylation. Demethylation was associated with younger genetic expression, in other words, the mouses genes remembered how to be young again, only after demethylation had occurred.

These data indicate that mammalian tissues retain a record of youthful epigenetic informationencoded in part by DNA methylationthat can be accessed to improve tissue function and promote regeneration in vivo, write the authors in their summary.

It remains to be seen whether records of youthful genetic expression are contained within other mammalian tissues, the liver for a random example, through methylation, and whether or not they can be accessed through demethylation.

6 Alcohol-Free Ways to Unwind at the End of a Long Day – Everyday Health

If youve noticed youre reaching for an extra glass of wine or beer (or spirit) to unwind lately, you're not alone.

A study published earlier this year in JAMA Network Open compared how U.S. adults reported using alcohol before the start of the COVID-19 pandemic and at two time points during it. The results? Adults indeed reported drinking more alcohol and doing so more frequently since the start of the worldwide health crisis.

Understandably, people have been faced with the task of managing a lot of uncertainty and factors that are out of our control, and that can be provoking, says Aimee Chiligiris, PsyD, an assistant professor of medical psychology (in psychiatry) at the Columbia University Medical Center in New York City who specializes in substance abuse.

Alcohol, for some people, can temporarily relieve anxiety. It can also be a distraction something to fill time, says Dr. Chiligiris. But while reaching for a drink whenever you feel anxious can provide temporary relief, it can also trigger a cycle of increased use that feeds on itself.

Whats more, its probably not helping you address the stressors youre facing.

Even if youre a casual drinker, Chiligiris says alcohol can have a boomerang effect on anxiety. For starters, alcohol can interfere with your sleep cycle, making it tougher to get a full night of quality rest. And we know poor sleep contributes to anxiety, so reaching for alcohol to calm your nerves becomes a vicious circle, she explains.

If alcohol is your only means to relax, that's concerning, says Timothy Fong, MD, a professor of psychiatry and the director of the addiction psychiatry fellowship at the University of California in Los Angeles. Dr. Fong says hes concerned about the 10 to 19 percent of the population who find themselves drinking more heavily than usual because of pressures from the pandemic, according to the aforementioned JAMA study.

The real question is not how this increase in alcohol use is impacting us (heavier drinkers) now, but its effects in the next two to three to five years, he says, referring to how excessive drinking (defined as 8 to 15 drinks per week, depending on your gender) can increase your risk of heart disease, cancer, digestive issues, depression, and anxiety, according to the CDC.

Coronavirus anxiety aside, a growing pile of research tells us that regular alcohol consumption just isn't good for you. Increasingly, experts say probably no amount of alcohol is really okay for your health.

Though red wine has been touted as heart-protective thanks to antioxidants such as resveratrol, even low amounts of alcohol have been associated with atrial fibrillation, or a rapid, irregular heartbeat, according to a study published in 2016 in the Journal of the American Heart Association.

And theres even more research to show that excessive alcohol consumption can be detrimental to immune function (per a 2015 Alcohol Research study), and increases the risk of cancer, cognitive problems, and stroke.

The bottom line: Reasons abound to find alternative ways to kick back and relax rather than reaching for the bottle.

If youve been drinking more than youd like, try replacing happy hour or whatever drinking ritual you have with a nondrinking activity that will activate the reward centers in your brain and curb your craving. Trying new things and experiencing pleasure without alcohol use can definitely provide the same sort of pleasure reinforcement that you may be looking for with alcohol, Chiligiris says.

You dont have to abandon your cocktails entirely or the rituals you share with friends and family, Fong says. But add other routines to your repertoire that bring you a sense of calm and joy. Add to your toolbox, he says.

Here are a few alternatives to try.

Taking time to connect with friends cheers you up it releases feel-good hormones like serotonin and oxytocin. Oxytocin calms the stress response, and there's evidence it curbs the urge to stress drink, according to a review published in 2014 in the Annals of the New York Academy of Sciences. Spending time with friends may even help you live longer, research shows. Even if you cant hang out like you used to, a long walk outside with a buddy or a video chat can have the same mood-boosting effect.

Fong says working out a bike ride, a walk, or cueing up a virtual HIIT class can alleviate the stress that drives us to drink. Plus, it will give you a shot of endorphins a chemical the body makes that triggers positive feelings.

If you dont want to bail on your weekly Zoom happy hour, there are plenty of non-alcoholic drinks that are just as tasty without the booze.

Instead of concocting your favorite cocktail, mix up a virgin version, or brew some soothing chamomile tea, which has its own calming properties, according to a study published this year in the International JournalofBiological Macromolecules.

Fong says meditation can help you relax as much as a vodka martini can. Alcohol chills us out by drowning out the noise and stress. Meditation, on the other hand, helps actually quiet the brain by relieving stress (rather than just covering it up), Fong explains.

Try using a meditation app to get you in the habit. Just remember to put your phone down for a while afterward doomscrolling or watching the news will just fire up your brain again.

Theres something very, very joyful about putting on your favorite music and dancing around the room by yourself, says Fong. Music releases dopamine, the same feel-good chemical that gets released in the brain when we drink alcohol, a study in the Proceedings of the National Academy of Sciences in 2019 showed.

Fong says tending to household projects youve long neglected can help distract you from the urge to drink as hard as they are to convince yourself to do. No one wants to clean out the closet, but when you actually do it, it brings an immense amount of joy, he says.

Dont be hard on yourself if your new happy hour habits take a few tries to catch on. Chiligiris says that once her patients make a change, they recognize that their overall quality of life has improved. They sleep better, are less anxious, and have more energy a virtuous cycle.

Breakthroughs in cures for COVID-19 – Guardian

Can a combination of chemical compounds in foods such as phycobilins and resveratrol, keto diet, novel cannabis plant extracts and steam inhalation therapy prevent and cure COVID-19?

According to a new study by plant biologists at North Carolina State University, United States, chemical compounds in foods or beverages like green tea, muscadine grapes and dark chocolate can bind to and block the function of a particular enzyme, or protease, in the Severe Acute Respiratory Syndrome Coronavirus type 2 (SARS-CoV-2) virus. SARS-CoV-2 causes COVID-19.

Proteases are important to the health and viability of cells and viruses, says De-Yu Xie, professor of plant and microbial biology at NC State and the corresponding author of the study. If proteases are inhibited, cells cannot perform many important functions like replication, for example.

One of our labs focuses is to find nutraceuticals in food or medicinal plants that inhibit either how a virus attaches to human cells or the propagation of a virus in human cells, Xie said.

In the study, the NC State researchers performed both computer simulations and lab studies showing how the so-called main protease (Mpro) in the SARS-CoV-2 virus reacted when confronted with a number of different plant chemical compounds already known for their potent anti-inflammatory and antioxidant properties.

Mpro in SARS-CoV-2 is required for the virus to replicate and assemble itself, Xie said. If we can inhibit or deactivate this protease, the virus will die.

Computer simulations showed that the studied chemical compounds from green tea, two varieties of muscadine grapes, cacao powder and dark chocolate were able to bind to different portions of Mpro.

Mpro has a portion that is like a pocket that was filled by the chemical compounds, Xie said. When this pocket was filled, the protease lost its important function.

In vitro lab experiments completed by Yue Zhu, an NC State Ph.D. student in Xies lab, showed similar results. The chemical compounds in green tea and muscadine grapes were very successful at inhibiting Mpros function; chemical compounds in cacao powder and dark chocolate reduced Mpro activity by about half.

Green tea has five tested chemical compounds that bind to different sites in the pocket on Mpro, essentially overwhelming it to inhibit its function, Xie said. Muscadine grapes contain these inhibitory chemicals in their skins and seeds. Plants use these compounds to protect themselves, so it is not surprising that plant leaves and skins contain these beneficial compounds.

The paper was published in the journal Frontiers in Plant Science. Zhu is the papers first author. The research was supported by the U.S. Department of Agriculture.

Also, researchers from Tennesee State University and Meharry Medical College in the United States have released an interesting new study on the preprint bioRxiv server in November 2020 that describes the broad-spectrum of antiviral activity that phycobilins can have. Phycobilins, which are bioactive plant compounds, were found to inhibit two coronavirus enzymes.

The current study focuses on phytochemicals, which are less toxic than synthetic drugs in many cases, and have been reported to have antiviral activity against multiple viruses, including coronaviruses. Selecting 15 phytochemicals of various chemical classes based on existing knowledge of their antiviral activity, the researchers studied their binding interactions with the SARS-CoV-2 MPro and PLPro.

These compounds include flavonols like quercetin, flavins like riboflavins, isoflavones such as daidzein and genistein, phenolic ketones like gingerol, and phenolic alkaloids such as capsaicin, all of which are well-known plant compounds.

Their interactions with the viral enzymes were examined by docking studies. These showed that the phycobilins (PCB) docked with the best score or binding energy for MPro, followed by riboflavin, cyanidin, daidzein and genistein in a close cluster. There were 12 important residues at the active site involved in these interactions with the phytochemicals.

When other phycobilins, such as phycourobilin (PUB), Phycoerythrobilin (PEB) and Phycoviolobilin (PVB), were explored, they all bound strongly to the binding pockets of these enzymes at specific amino acids, with PCB having the highest or almost the highest docking score for both enzymes. It is notable that these compounds also have other powerful therapeutic properties such as scavenging oxidative radicals, inhibiting cancer cell division and platelet aggregation. They can also be used orally in the form of the phycobilin-protein complex, phycobiliprotein. Once ingested, it is digested in the human gut, and free PCB is released there, which may account for the therapeutic properties of this complex.

The researchers call for in-vivo studies on inhibition of CoVs infectivity using human cells and animal models. Moreover, they suggest that more phycobilin lead compounds could be developed by structure-guided development to rapidly lead to the discovery of a single agent with clinical potential against existing and possible future emerging CoV-associated diseases.

Also, researchers in Canada have conducted a study suggesting that novel Cannabis sativa extracts may decrease levels of the host cell receptor that SARS-CoV-2 uses to gain viral entry to target tissues.

The team from the University of Lethbridge and Pathway Rx Inc., Lethbridge developed hundreds of new C. sativa cultivars and tested 23 extracts in artificial 3D human models of the oral, airway and intestinal tissues.

As recently reported in the journal Aging, 13 of the extracts downregulated expression of the SARS-CoV-2 host cell receptor angiotensin-converting enzyme 2 (ACE2).

The observed down-regulation of ACE2 gene expression by several tested extracts of new C. sativa cultivars is a novel and crucial finding, say the researchers.

While our most effective extracts require further large-scale validation, our study is important for future analyses of the effects of medical cannabis on COVID-19, write Olga Kovalchuk and colleagues.

Numerous studies have also reported high levels of ACE2 expression in the lower respiratory tract of patients with chronic obstructive pulmonary disease (COPD). This patient group is associated with increased susceptibility to COVID-19 and more severe disease.

Cannabis sativa, particularly cultivars rich in cannabidiol (CBD), have previously been shown to alter gene expression and to possess anti-inflammatory and anti-cancer properties.

However, the effects of C. sativa on ACE2 expression are not known, says the team.

Using artificial 3D human models of oral, airway and intestinal tissues, we identified 13 high-CBD C. sativa extracts that decrease ACE2 protein levels, writes the team.

The researchers say the findings provide a foundation for further analyses of the effects C. sativa may have on the pathogenesis of COVID-19 and other viral diseases where the ACE2 receptor is used as a molecular gateway.

The extracts of our most successful novel high-CBD C. sativa lines, pending further investigation, may become a useful and safe addition to the prevention and treatment of COVID-19 as an adjunct therapy, concludes the team.

Also, a recent study published in the journal Phytotherapy Research reveals that resveratrol, a phenolic compound produced by spermatophytes, can inhibit the replication of SARS-CoV-2.

The scientists from Shenzhen Third Peoples Hospital, China, conducted the study to investigate how resveratrol, a phenolic plant compound, can modulate the replication of SARS-CoV-2 in Vero cells. They chose resveratrol as an experimental compound because of its proven efficacy in inhibiting replication of several viruses, including dengue virus, Zika virus, influenza virus, and more importantly, Middle East respiratory syndrome coronavirus (MERS-CoV), which is a betacoronavirus in the same family as SARS-CoV-2.

The current study identifies resveratrol as a potential antiviral candidate that is capable of inhibiting SARS-CoV-2 replication. Previously, it has been reported that resveratrol reduces cellular inflammation by decreasing the production of nitric oxide. Moreover, there is much evidence showing that resveratrol acts as a potent antioxidant. Another benefit of resveratrol is that it does not cause any serious adversity even when used at a very high concentration. Because oxidative stress and inflammation are common consequences of any viral infection, the scientists believe that resveratrol holds the potency to be used as an effective therapeutic agent to treat COVID-19 patients.

Meanwhile, Italian researchers at the Biochemistry and Pharmacology Laboratory, Meyer Childrens University Hospital, Florence, Italy, have published a study on the efficacy of steam inhalations as another possible treatment to help mitigate SARS-CoV-2 infection. Their study was published in the latest issue of the journal Life Sciences.

Early on in the viruss outbreak, it was found that soapy water could help break down the viral envelope and thus denature it. Researchers added that heat has also been seen to denature the proteins that lead to loss of infectivity of the SARS-CoV that caused the 2002 SARS outbreak. The researchers have shown that heat can also denature the SARS-CoV-2 virions proteins.

Steam inhalation cycles are thus considered to be useful in damaging the capsid of the SARS-CoV-2 envelope and prevent infection, write the researchers. They write that the European Pharmacopoeia VI edition has recommended steam inhalations as a procedure to treat respiratory diseases.

This study, although a small one, shows the beneficial effects of steam inhalation in reducing viral shedding from infected patients. The team writes that this could be an easily accessible, non-invasive and inexpensive procedure which has been proven to be effective. It should be subjected to larger clinical trials, the team wrote.

They concluded, Should our preliminary observations be confirmed, the protocol could be used against COVID19 or other viral infections using vapotherm masks, where temperature, time of exposure and size of steam particles can be set and monitored.

A study comparing severe COVID-19 incidence among obese patients on a KD versus those on an ordinary diet may throw more light on the validity of this hypothesis.

Everyone talks about masks. I like them and wear thembut that is external protection only. While important and useful, I will be totally honest with you about them. The way we respond from a pathogen has more to do with our internal protection, as in our immune system!

So ask yourself if its up to par. If its not, then you need to start fertilizing your garden of microflora (i.e. probiotics) and avoid or minimize anything that weakens your immune system. In no special order, here are the most common factors that suppress your immune system.

People think of this as good for the heart, but studies repeatedly show that free radical harm takes place in your liver and can lead to compromised liver enzymes as well as hepatotoxicity. If you cant filter out and neutralize poisons and pathogens, your immune system is weaker.

I can say everything here that I just said for alcohol. Additionally, smoking is known drug mugger of vitamin C which everyone knows helps reduce misery from the common cold and flu. If you smoke, you have lower levels of C and again, this weakens your ability to fight. Furthermore, smoking hurts your lungs and that is one target organ of COVID-19.

Need I say more than the word junk? If youre eating junk food, your body is still starving for nutrients at a cellular level. You may very well be at a healthy weight and BMI; however, your cells are dying for more nutrition and that means your immune system is weak.

There are some immune-suppressing medications that people take for autoimmune conditions to slow down the self-directed attack at their own tissue. For example, methotrexate, azathioprine, and etanercept are three that come to mind off the top of my head.

These are necessary, and Im not saying to discontinue them, however you should be extra vigilant about exposure, masks, social distancing and so forth. Since you shouldnt stop your medication, just be more careful. Another immune-suppressing category are the steroids such as inhalers for asthma and those used for allergies (prednisone, dexamethasone, hydrocortisone and so forth). Cyclosporine is another used frequently after organ transplants.

If you have the measles, HIV, Lyme disease, bartonella, candida, mold exposure or mononucleosis for example, these types of chronic infections weaken your immunity. Your body is already fighting in these cases and dealing with an imbalanced set of pro-inflammatory cytokines so another infection would be harder to deal with because your immune reserves are spent! Supporting your body by eating healthy foods and taking antioxidants is key to keeping your immune system strong.

So, as we approach the colder months, consider both internal and external methods for immune support! This way you give yourself the best odds. Dietary supplements that may help improve internal protection include vitamin D, E, C and zinc. The herbal antioxidants that do this include skullcap, resveratrol, green tea extract, ginger, blueberry and Andrographis. There are many others.

If Im working with a patient or client who incorporates snacks into their daily fueling routine, one of the most frequent go-to snack choices I hear are peanuts or another member of the nut family, such as almonds or cashews. Strategic snacking between meals is one of the best ways to promote appetite management, improve daily nutrition intake and manage energy levels, which strongly correlate with blood sugar levels. Pairing certain foods and nutrients can help stabilize blood sugar, leading to steady energy levels, rather than distracting highs and lows. This is where peanuts come in.

Glycemic index is a value used to quantify the amount that a given food raises blood sugar levels. The values range from 0-100, with lower values indicating less robust blood sugar responses and higher values indicating a significant increase in blood sugar. One serving of a cereal such as corn flakes has a high GI of 81, whereas one serving of soy beans has a GI of approximately 16. Peanuts measure even lower at 14, which means they can help prevent spikes in blood sugar and contribute to a slower and steadier increase in energy. For Type 1 and insulin-dependent Type 2 diabetics, this translates to reduced insulin needs.

Recently, research has shown that the consumption of peanuts and peanut butter can be an effective strategy to stabilize blood sugar and in turn prevent the onset of Type 2 diabetes for those with prediabetes, as well as those with a heightened risk from factors including chronically high blood sugar values and thus drastic fluctuations in energy levels.

A study published in 2018 in the Journal of the American Medical Association found that women who consumed peanut butter five times a week reduced their risk of developing Type 2 diabetes by 21 percent. Additionally, in 2019, the Journal of American Nutrition published research exploring the effect of peanut butter on blood sugar when added to a meal with a high glycemic load. Results showed that the subsequent blood sugar response was significantly lower than when peanut butter was not included in the meal.

The magic lies in the nutrient composition of a peanut, which is low in carbohydrate and high in unsaturated fat. One handful of peanuts also delivers 7 grams of plant-based protein in conjunction with fiber. Thats more plant-based protein than any other nut, with more protein per serving than half a cup of chickpeas. When eating a meal or snack high in carbohydrate or with a high ability to raise your blood sugar, simultaneously consuming foods rich in protein or unsaturated fat adds balance to your blood sugar. Peanuts are an excellent choice, and for this reason some scientists call peanuts a superfood. In addition to protein and healthful fats, peanuts contain 19 micronutrients, including niacin, vitamin E and manganese, which function as antioxidants and contribute to overall health.

Speaking of antioxidants, they can improve vascular function and biomarkers such as blood lipids and cholesterol. Peanuts are a significant source of resveratrol, a nutrient that has recently received increased attention because of its antioxidant capacity. Resveratrol, which is found in grapes and red wine, can open blood vessels and improve blood flow. Peanut butter is very close to grape juice in terms of resveratrol content. If youre trying to up your antioxidant intake, opt for peanut butter, which has three times the reservatrol as roasted peanuts. A study examining bloodwork in response to an evening snack demonstrated that consuming 1 serving of peanuts before bed can reduce blood triglyceride levels. No wonder the World Health Organization and U.S. Department of Agriculture both recommend eating nuts daily.

Despite strong data to support the inclusion of peanuts and/or peanut butter as part of our daily nutrition intake, many shy away from nuts because of the perception that theyre fattening. Almost every day, a patient will tell me they eat peanuts for a snack, and then follow up with I know thats bad! or cringe with guilt. Though peanuts are high in fat, the type of fat found in nuts is the more healthful, protective, unsaturated kind that provides wonderful health benefits. Consumption in moderate amounts has been shown to have no effect on body weight and might even help the body burn more energy.

Other forms of nuts and nut butters provide generally similar nutrition benefits, with a few minute nuances. Peanuts and peanut butter can be a cost-friendly addition to your daily fueling routine. Peanuts can improve blood sugar management, energy levels, cardiovascular markers, stimulate satiety and help regulate appetite. Instead of avoiding this nutrient-dense food, pack on the peanuts!
