Gene Editing Tool was Modified to Improve Therapies for HIV – Science Times

The researchers from the City of Hope may have found a way to sharpen the cheapest, fastest, and most accurate gene-editing tool called the CRISPR-Cas9 so that it can successfully cut out undesirable genetic information. This cutting ability was improved so that one day, experts can fast-trackpotential therapiesfor sickle cell disease, HIV, and other immune conditions.

What is the CRISPR-Cas9 design

Tristan Scott, Ph.D., lead author of the study and a staff research scientist at City of Hope's Center for Gene Therapy, said that the CRISPR-Cas9 design might be the difference between trying to cut a rib eye steak with a butter knife versus cutting it with a steak knife. Other scientists have tried to improve CRISPR cutting through chemical modifications, but that is an expensive process, and it is like diamond-coating a blade. Scientists have designed a better pair of scissors that people can buy easily anywhere.

Thestudythat was done by Dr. Scott and his team was published in Scientific Reports, and it is the first time scientists have systematically gone through the guide RNA sequence to change it and to improve CRISPR-Cas9 technology. A patent application was filed at The Kevin Morris Lab at City of Hope, claiming this improved CRISPR-Cas9 design, which could result in a doubling of activity, but the exact amount was dependent on the target site.

Theeffects of this CRISPR-Cas9 designis downstream as it has more "clean" results in mouse model and cell experiments aimed at making new therapies because the target that was knocked out was more successfully removed. More results could quicken new therapies from the laboratory to patients' bedsides. The therapeutic product should have successful cuts, which could mean that an improved therapy is on the way, but further research is still needed. The exact ways and process of why this change to the CRISPR system improves gene editing still needs to be determined.

Experiments through nanotechnology

The researchers of the study experimented on cells by making certain changes to the tracrRNA or trans-activatingCRISPR RNAwhich they got from Streptococcuspyogenesbacteria, and it is a part of the components that are used to guide the genetic scissors, also known as Cas 9, to the right gene sequence. StreptococcuspyogenesCas9 is the most widely used genetic scissor. Dr. Scott and his team used an RNA protein system because it gives an increase of activity that disappears about 12 hours after being introduced into the cell, which means that there is a decreased chance of accidentally editing the human genelater after the fix has been made.

Scientists found that the modified tracrRNA improved the overall silencing of certain genes by increasing desirable mutations in the genetic material. In this study, the target was an essential component of HIV's lifecycle, the protein CCR5 on immune CD4+ T-cellsa current target in clinical trials seeking to re-create a person's immune system so they can be resistant to HIV. The modified tracrRNA had improved the overall cutting at this site and inactivation of CCR5, and hopefully, that will translate into better protection for the immune system.

The new tracrRNA design is better at improving the activity at the HBB gene and the BCL11A site. The HBB gene, which is responsible for producing beta-globin that is located inside red blood cells, and BCL11A, which may function as a leukemia disease gene, are targeted by the tracrRNA in order to make different therapies for HIV, which is known as a blood disease that does not have any known cure yet.

Dr. Scott stated that if the line of research remains consistent, and they can dependably sharpen the genetic scissor, the result could eventually be new or improved genetic therapies. He also said that his team is at the beginning of a long scientific process.

Nanotechnology could be used to screen for COVID-19: professor – Focus Taiwan News Channel

Taipei, June 30 (CNA) A form of nanotechnology developed by a professor at National Cheng Kung University (NCKU) could be used to screen patients for COVID-19 in a matter of minutes, the university said in a statement Monday.

Professor Liao Jiunn-der's () use of "nanocavities," which he describes as nanoscale traps that resemble upside-down pyramids, can be used to catch even a single virus, he said.

To screen patients, samples are obtained from their upper respiratory tract, purified, and placed on a testing slide that has the "nanocavities."

A laser is then used to detect if a virus has been trapped inside a cavity, which virus it is, and whether it is active, Liao said.

Liao began developing the nanocavity technology in 2009, and it has proven effective in screening for seven kinds of viruses, including the SARS coronavirus and H1N1, according to the statement.

His team obtained a patent for the technology in 2012, the NCKU statement said.

A variation of the technology developed by Liao's team, which uses the same kind of laser to detect whether viruses are present on a silicon testing slide, could also be applied to COVID-19 screening.

Liao's team has yet to test the technology with the new coronavirus (SARS-CoV-2), but it will work with the university's hospital and College of Medicine to conduct tests over the next six months to determine how well the system works.

Should the technology pan out, the team's goal is to set up testing stations at ports and airports to facilitate COVID-19 testing, and create a mobile testing station that can provide services when needed, NCKU said.

The project is one of seven COVID-19 related studies that is being supported by the Ministry of Science and Technology's Department of Engineering and Technologies, according to NCKU.

How do you think the current pandemic could impact/change/shape the research community in the short/long term?

The COVID-19 pandemic has dramatically changed our life in many ways. One of these changes is the major disruption in research activities for the research community. We will have less lab-based research results and findings in the short term. However, going through this difficult time together, we will learn how to deal with crises like this and become more resilient to future ones. In the long term, it will give us an opportunity to rethink the way we work and communicate. I think we should use this time to rework the existing research process to achieve higher efficiency and productivity and identify new ways to strengthen the research community. Last but not the least, we should support each other and get through this pandemic together.

My research group uses food chemistry, food biophysics, material science and nanotechnology approaches to investigate structurefunction relationships of food proteins and polysaccharides with the ultimate goal of improving food safety and quality. We have made various nanostructures, including nanoparticles, nanoemulsions and nanolaminates, with applications including nutraceutical encapsulation and targeted delivery, antimicrobial packaging materials, food pathogen detection and edible coatings.

The most exciting aspect part of my work is my contribution to improving food safety and human health.

Keeping up with current research in my area worldwide and coming up with novel ideas are both very challenging.

As a professional scholar and educator, I wanted to contribute to the scientific field. With my expertise in food chemistry, food biopolymer biophysics and food nanotechnology, I hope to bring forward high-quality research in food science to the journal. I am very excited to join the RSC Advances team and am planning to give my time and effort to advance the journal.

By ensuring that submissions fit the journal, checking for flaws in experimental design and advancing our knowledge in chemistry, I'm hoping to bring novel, exciting and solid publications to the scientific community. Halifax, Nova Scotia--(Newsfile Corp. - May 26, 2020) - Sixth Wave Innovations Inc. (CSE:SIXW) (OTC Pink: ATURF) (FSE: AHUH) (the "Company" or "Sixth Wave") announces a strategic alignment with Natural Ascent Consulting (NAC) and the signing a hardware loan and production agreement which will allow NAC to field test and contribute to the commercialization process of the Company's Affinity cannabinoid purification system for the remediation of non-compliant hemp and cannabis extracts and standard extraction of cannabinoids from other crude extracts. The contract engages NAC in cannabinoid production using the Affinity System, and establishes a Southern California based production hub for Sixth Wave in one of the worlds largest cannabis markets. NAC will also use their lab capability with Sixth Wave to trial multiple production applications for the Affinity System including continued validation of the Systems capability to remediate failed extracts for heavy metals, pesticides, and possibly the chemicals associated with fire retardants that are found in both hemp and cannabis extracts due to their widespread use to combat forest fires.

As an established consulting firm supporting the cannabis and hemp industries, NAC brings an array of knowledge and resources to the venture. With experience in a wide variety of projects, NAC centers on processing CBD / CBG biomass to the highest quality downstream wholesale product lines at an industrial scale. NAC is currently focused on assimilating advanced technologies, such as THC remediation and cannabinoid conversion/isolation, into a contract farming business model for the hemp industry.

Focused on developing nanotechnology applications, Sixth Wave's Molecular Imprinted Polymers (MIPs) have also been applied in industries including homeland security, mining, and pathogen detection. Once completed, NAC will begin processing extracts using the pilot Affinity System in accordance with Sixth Wave's Total System Performance License Agreement which is still under negotiation. Further, Sixth Wave will replace the pilot system with a fully GMP compliant system to meet NAC's expected processing requirments. The NAC team will continue to support roll out and installation/support under contract to Sixth Wave when the Company begins fulfilling delivery of Affinity Systems to other customers.

The Company further advises that it has engaged Hybrid Financial to promote market awareness and further the Company's reach within the investment community. Hybrid will be engaged for a fee of $15,000.00 per month for an initial term of six months, with an automatic renewal for successive three month terms unless canceled by written notice 30 days prior to the end of the term.

According to a report on ScienceDaily, novel nanoelectronic device research has enabled brain neurons and artificial neurons to communicate with each other over the internet in a new study that has been published in the journal Nature Scientific Reports.

Brain function occurs due to circuits of spiking neurons which are connected together by highly complex microscopic synapses. Scientists based at the Universities of Padova and Zurich and ETH reated a hybrid neural network in which biological and artificial neurons in different parts of the world were able to communicate with each other over the internet through a hub of artificial synapses made using nanotechnology.

Rat neurons were cultivated while the artificial neurons were created on Silicon microchips, and the virtual lab was brought together via setup controlling nanoelectronic synapses developed at the University of Southampton; these synaptic devices are called memristors.

Spiking events were captured being sent over the internet from the biological neurons in Italy to Southampton and then distributed over the memristive synapses; responses were sent onward to the artificial neurons in Zurich in the form of spiking activity. The process also worked simultaneously in reverse as well from Zurich to Padova thus the artificial and biological neurons were able to communicate bidirectionally and in real time.

"One of the biggest challenges in conducting research of this kind and at this level has been integrating such distinct cutting edge technologies and specialist expertise that are not typically found under one roof. By creating a virtual lab we have been able to achieve this, said Themis Prodromakis, Professor of Nanotechnology and Director of the Centre for Electronics Frontiers at the University of Southampton.

"We are very excited with this new development. On one side it sets the basis for a novel scenario that was never encountered during natural evolution, where biological and artificial neurons are linked together and communicate across global networks; laying the foundations for the Internet of Neuro-electronics. On the other hand, it brings new prospects to neuroprosthetic technologies, paving the way towards research into replacing dysfunctional parts of the brain with AI chips, added Prof Prodromakis.

This research is expected to inspire an interest in a variety of scientific disciplines to accelerate the pace of innovation and advancement in the field of neural interface research, particularly the ability to connect disparate technologies across the globe and move towards the democratisation of these technologies and remove a barrier to collaboration.

Blueberry Therapeutics has received an investigational new drug (IND) clearance from the Food and Drug Administration (FDA) to proceed with the clinical development of BB2603

Blueberry Therapeutics has received an investigational new drug (IND) clearance from the Food and Drug Administration (FDA) to proceed with the clinical development of BB2603 for the treatment of onychomycosis.

The IND is a crucial milestone in the continued development of BB2603, a nanoformulated, topical form of terbinafine for the treatment of onychomycosis (fungal toenail). With the IND in place the company has initiated a Phase IIb international, multicentre double-blinded study.

The primary outcome of this study is to look at the efficacy BB2603 in onychomycosis, with a secondary dose finding outcome also being investigated.

John Ridden, CEO said: Having received clearance from the FDA we are looking forward to the opportunity to work with over 20 international sites on the Phase IIb trial of BB2603 with the first patient expected to be recruited in March 2020.This is a key milestone in our path to commercialisation of our nanotechnology product portfolio.

With BB2603, Blueberry Therapeutics aim to maintain the efficacy of the oral gold standard treatment, but at a lower dose. Delivering the active pharmaceutical ingredient using nanotechnology ensures it penetrates across the nail plate and reaches the target site of action in a controlled manner, meaning lower doses can be used and systemic side effects are minimised.

FG to harness benefits of nanotechnology – Daily Trust

The Federal Government will soon produce a national nanotechnology policy to guide the operation of nanotechnology in the country, the Permanent Secretary of the Federal Ministry of Science and Technology, Mr. Bitrus Bako Nabasu has said.

When it is eventually produced, the policy will spearhead the use of nanotechnology in the country and would make products to be lighter, smaller, stronger, cleaner and precise, Mr Nabasu said.

The Permanent Secretary said this when he Inaugurated the Committee on the First Validation Exercise on the Draft National NanoTechnology Policy Document in Abuja on Friday.

He mandated the Committee to produce a National Nanotechnology policy to guide the operation of Nanotechnology in the country, assume responsibility, and come up with a draft National Nanotechnology policy document.

He said Nanotechnology is an emerging technology set to revolutionise the manufacturing technology process by making most products lighter, smaller, stronger, cleaner and very precise.

Mr. Nabasu said that application and utilization of this technology is being made in various fields such as agriculture, health, medicine, and chemical industries to enhance the quality of lives of citizens.
