Olson said knowing which water molecules need to be displaced, and which do not, provides essential information to guide the development of drugs to treat COVID-19. The first step toward determining that is mapping where the water molecules are located on the proteins, he said.

We were seeing clusters on multiple continents and were unable to trace the sources of infection, he said. It was clear this was going to be an immediate and international public health issue. That compelled me to do something.

The team utilized GIST, the computational tool developed by Kurtzman.

Now that solvation maps of the COVID-19 virus are available, Olson and his team plan to use that information to search for new drugs, they said. Making the information public and free also will aid others in their search for new and effective pharmaceuticals to defeat the coronavirus.

According to Altmetrics Attention Score, which measures the online attention and activity of research papers, the work conducted by the Olson team ranks in the Top 25%. Given that ranking, based on more than 2,200 views and over 300 downloads of the paper, Olson is encouraged about the development of drugs to fight the coronavirus.

I have never seen the international scientific community come together the way it has now to find some answers, he said. There is international collaboration and the sharing of information that previously would not have been shared.

Olson, who teaches forensic science at CCM, joined the colleges faculty in 2018. At CCM, he and several other professors developed the colleges first virtual reality class to teach students forensic science skills by analyzing murder scenes. Along with his work on the coronavirus, he is researching the development of painkillers to replace opiates.

Olson, who earned his associate degree in science from the Borough of Manhattan Community College, a bachelors in biochemistry from Hunter College and a masters in biochemistry from the City University of New York Graduate Center, is now working on his doctorate in biochemistry from CUNY Graduate Center.

Olson said he is a strong advocate for community colleges.

Community colleges place a high value on quality teaching, he said. I am so fortunate. I have fun researching tiny molecules and, on top of that, I get paid to talk to students about narcotics, murders and other fascinating topics. I cant imagine having a better job.

To investigate the impact of 68 Gallium prostate-specific membrane antigen (68 Ga-PSMA) tracers on the management of prostate cancer patients with biochemical recurrence (BCR) by systematical review and meta-analysis.

We performed a literature search on PubMed, Embase, and Web of Science databases up to October 29, 2019. We included studies that reported the proportion of management change after 68 Ga-PSMA tracers in patients with BCR. We used the Quality Assessment of Diagnostic Accuracy Studies-2 tool to evaluate the quality of the included studies. The proportion of management changes were pooled using a random-effects model. Subgroup analyses and meta-regression analyses were performed to explore the source of heterogeneity. Sankey diagram was used for displaying the treatment change from the before to after 68 Ga-PSMA tracers.

We included 20 eligible studies (2,026 patients). The pooled of management change occurred in 53% (95% confidence interval (CI), 46-60%) of patients with BCR and 51% (95% CI, 34%-67%) in early BCR group (prostate-specific antigen (PSA) < 0.5 ng/mL). The pooled PET-positive in patients with BCR was 68% (95%CI, 59%-78%). Fourteen studies reported the management change, with most changes being intermodality (42% vs 17% in intramodality).

The 68 Ga-PSMA tracers can alter the management of more than half of prostate cancer patients with BCR, as well as in the early BCR patients. The 68 Ga-PSMA tracers might be used to guide the individualised treatment in patients with BCR, particularly in the early recurrence patients.

The consumption of raw carrots triggers allergic reactions in many people. Contrary to popular belief, cooked carrots can also have this effect. This was recently discovered by a research team at the University of Bayreuth. The carrot's allergen, Dau c 1, assumes a structure that is harmless to allergy sufferers when highly heated. However, as soon as the temperature drops, it largely regains its natural structure. The researchers present their study in the journal "Molecular Nutrition & Food Research".

"The results of our research clearly suggest that sufferers who are sensitive to the carrot allergen should generally avoid eating carrots. Heating carrots does not destroy or only incompletely destroys the protein structures that can cause allergic reactions ", says Prof. Dr. Birgitta Whrl from the Biochemistry IV research group at the University of Bayreuth. "The risk of allergy patients developing an allergic reaction arises not only when eating freshly cooked carrots or canned carrots. It also arises when carrot extract is added to food," adds Thessa Jacob M.Sc., first author of the study and PhD student at the Biochemistry IV research group.

The natural carrot allergen Dau c 1 is actually a mixture of several, structurally very similar proteins. These so-called isoallergens were produced individually in the laboratory in bacteria. Both the protein mixture of the natural Dau c 1 and the individual isoallergens were examined at temperatures up to 95 degrees Celsius to see how their structures change upon increasing and decreasing the temperature. This analysis showed that the natural mixture and almost all isoallergens are still capable of causing allergies after cooling down to 25 degrees Celsius. Despite the previous heating, antibodies present in the organism of allergy patients will still trigger allergic reactions. Although in some cases this effect is less pronounced than before heating, it is generally maintained.

"This is the first time that the Dau c 1 isoallergens have been subjected to such an extensive series of tests. The separate examination of the structurally similar molecules was particularly important to us in order to determine which of the isoallergens trigger immune reactions under the tested conditions," says Thessa Jacob M.Sc.

The tests clearly showed that the structural stability of the carrot allergen does not depend on temperature alone. Acidity, as expressed by the pH value, is also important. Of particular interest is pH value 3, which typically prevails in the stomach after food intake. At this level of acidity and at normal room temperature, at least some epitopes can continue to exist despite previous heating. Epitopes are those molecular substructures by which the immune system of allergy sufferers recognises the respective allergen, allowing an allergic reaction to occur.

In their structural investigations of the Dau c 1 isoallergens, the researchers mainly used magnetic resonance spectroscopy (NMR) and CD spectroscopy in laboratories on the Bayreuth campus. For other analytical procedures, they received support from the Paul Ehrlich Institute in Langen and from Nano Temper Technologies in Munich.

The 8th edition of the American Joint Committee on Cancer (AJCC) TNM staging system subdivides prostatic pT3 tumors into pT3a, which includes cases with extraprostatic extension (EPE) and pT3b, which is defined by the presence of seminal vesicle invasion (SVI) with or without EPE. Yet, it is not established whether combined SVI and EPE impart a worse prognosis compared to SVI alone. We studied a cohort of 69 prostatectomy patients with SVI with or without EPE. Patient age at the time of radical prostatectomy was documented and Gleason score and presence or absence of EPE and/or SVI were determined. Biochemical recurrence (BCR) was defined as a PSA rise >0.2ng/mL. The frequency of BCR was 33.9% in cases with combined EPE and SVI versus 12.5% in cases with SVI alone (relative risk=2.71). An additional cohort of 88 patients also showed a higher frequency of lymph node metastasis of 29% in patients with combined SVI and EPE at the time of radical prostatectomy versus a 10% frequency of lymph node metastasis in patients with SVI alone (relative risk=2.9). Based on our data, we propose further subdividing pT3 prostate cancers into three groups: EPE alone (pT3a), SVI alone (pT3b), and combined EPE and SVI (pT3c). This classification system would more accurately identify patients with pT3 prostate cancer who are more likely to experience worse outcomes and provide clinicians with additional information to aid in follow-up and postoperative treatment decisions.

UK research getting to bottom of COVID clots – ABC 36 News – WTVQ

The research led by Jeremy Wood, Zach Porterfield and Jamie Sturgill in the Department of Internal Medicine; Beth Garvy in Microbiology, Immunology & Molecular Genetics; and Wally Whiteheart in Molecular & Cellular Biochemistry, suggests localized inflammation in the lungs caused by COVID-19 may be responsible for the increased presence of blood clots in patients.

The study also provides evidence suggesting the risk of thrombosis could persist after the infection clears.

The study examined the blood of 30 COVID-19 patients including 15 who were inpatients in the intensive care unit, and 15 who received care as outpatients at UKs Infectious Diseases Clinic, along with eight disease-free volunteers who acted as a control group.

Compared to baseline, the COVID-19 patients had elevated levels of tissue factor, a protein found in blood that initiates the clotting process. Patients also had reduced levels of protein S, an anticoagulant that helps prevent blood clotting.

The researchers concluded that lung inflammation caused by COVID-19 is what leads to a decrease in protein S. Thisinflammation also causes immune and possible endothelial cell activation, which leads to increased tissue factor protein.

What weve learned is that the clotting is not caused by anything systemic. Localized inflammation in the lungs is whats driving this whole process, Wood said. With an increase in tissue factor and a deficiency in protein S, COVID-19 patients get more blood clotting without the ability to shut it down or control it.

The study additionally showed that protein S levels remained low in some patients even after they tested negative for COVID-19, which suggests that blood clotting issues may persist after infection and long-term monitoring of thrombotic risk may be necessary.

Wood says this preliminary data could be a cause for concern. Certain viruses like HIV are linked to a long-term deficiency in protein S, which causes an ongoing risk of thrombosis in patients. It is not yet known if COVID-19 could cause a similar persisting protein S deficiency.

Tissue factor and protein S are good markers to monitor for long-term thrombosis risk and the data suggest that we need to be monitoring these patients because were not seeing these parameters corrected immediately, Wood said.

The research team recently received a grant from UKsCenter for Clinical and Translational Science(CCTS) to begin a longitudinal study to look at these levels in patients over the next year.

This will help answer the question: will this risk remain like it is in the HIV patients or will it go away?

The study was funded in part by anAlliance Grantthrough the College of Medicine as well as UKsCOVID-19 Unified Research Experts (CURE) Alliancethroughthe Vice President for Research and the College of Medicine and the CCTS. It was a product of collaboration between a number of different groups at UK that have been studying COVID-19.

As a part of her research at the Korb Lab, Erica Korb, an assistant professor of genetics in the Perelman School of Medicine strives to understand mechanisms of epigenetic regulation in the brain. Epigenetic regulationmeaning how the world around us impacts gene expressionis essential to the function of neurons in the brain and aides with the creation of new memories, and more.

Using methods such as microscopy, bioinformatics, biochemistry, behavioral testing, and more, Korb and her lab also study the role of histoneswhich are a group of basic proteins found in chromatinin neuronal function and in neurodevelopmental disabilities.

Our lab works at the intersection of neuroscience and epigenetics. Epigenetics, in its broadest sense, explores how our environment can change the expression of our genes, Korb says. Epigenetics is incredibly important in the brain and contributes to the creation of new memories, our ability to adapt to our environment, and to the development of neurological disorders.

Korbs work is also collaborative, joining new technologies and different labs to bring individuals with very different skills and expertise together.

Ive been really fortunate to work with a group of Penn faculty on a SARS-CoV-2 related project. Were examining a poorly understood protein encoded by SARS-CoV-2 and its role in disrupting host-cell epigenetic regulation, she says. Weve gotten to work with Susan Weiss, Shelley Berger, Ben Garcia, and Ed Morrissey, and learned an incredible amount in the process. Its been challenging to learn a new field since we are primarily a neuroscience lab but its also been incredibly rewarding to have the chance to work with experts and contribute to our understanding of this virus.

New crop of medical students are headed to the lab – Stanford Medical Center Report

Karen Malacons life plan is to open a neurology research lab perhaps to investigate prenatal brain development, or maybe cognitive decline in aging patients.

My ultimate career goal is to run my own lab, and use its discoveries to help patients, she said. I feel its very important to interact with patients to inform the questions Ill be asking in the lab.

Malacon is one of 10 students who are starting StanfordsMedical Science Training Program, a seven- to eight-year curriculum that awards both a medical degree and a doctorate. But many of the 80 other students entering medical school this year are also intent on research: Twenty-two, far more than the more typical three to five, have committed to spending at least one extra year of medical school in a laboratory.

The school intentionally pursued research-oriented students, saidPJ Utz, MD, associate dean for medical student research, because the number of physicians with research expertise has been dropping nationwide.

That trend has many atStanford Medicineand other academic medical centers concerned, as physicians who conduct research form a crucial link between laboratory exploration and patient care. They are especially suited to direct research toward treatments while also bringing laboratory findings into the clinic.

Some students want to focus on patients, and thats fine, said Utz, a professor of immunology and rheumatology. But we are seeing the extinction of the physician-scientist unless we do something. If anyones going to lead this effort its got to be Stanford.

As a biomedical science hub with laboratories a short walk from Stanford Hospital, Lucile Packard Childrens Hospital Stanford and the medical school, the university is well-positioned to educate physician-scientists.
