New Drugs That Could Help Us Live Longer – Healthline

Were living longer these days, but that hasnt stopped newer branches of medicine from trying to enhance and further extend our lifespans.

Researchers in longevity medicine and biogerontology are studying drugs and compounds that can prevent and reverse aging on the cellular level.

Over the past few years, public interest in longevity issues has grown, but that interest is mostly in health-extending therapies, not simply life-extending therapies. Most people are interested in living longer, but only if they can also be healthier longer, Sonia Arrison, author of 100 Plus: How the Coming Age of Longevity Will Change Everything, From Careers and Relationships to Family and Faith, and founder of Singularity University in California, told Healthline.

While many people are focused on lifestyle changes, the addition of drugs rapamycin and metformin are two that Arrison mentioned gives humans more options to extend their lives as well as the quality of their lives.

Compared with preventative drugs such as statins to avoid having a heart attack, anti-aging drugs fight multiple diseases at once instead of focusing on one ailment, Arrison added.

A person who goes by the name Reason, a technologist and author on FightAging.org, which highlights health- and longevity-enhancing medical technologies, told Healthline that the drugs are targeting a wide range of age-related ailments and diseases.

All age-related diseases are age-related because they are caused by the processes of aging, which is to say an accumulation of damage in cells and tissues, Reason wrote in an email.

The field of geroprotection involves understanding cell senescence, which is when cells stop dividing.

When senescent cells no longer divide, they stop functioning, so organ health deteriorates. Cell senescence also causes the release of proinflammatory cytokines, which damage tissues.

This arena involves the use of geroprotectors, which are compounds that can stop or reverse cellular aging, and senolytics, which are compounds that can pinpoint and destroy senescent cells.

This is why researchers are looking into geroprotectors and senolytics, which requires long spans of time to understand.

In essence, todays researchers may never see the outcomes of the research theyve started because it can take decades to study. Nevertheless, scientists continue to try to understand these compounds and how they can alter our cells.

Researchers are also looking into our cells telomeres, which are short segments of DNA in our chromosomes that protect cells from wear and tear that comes with aging. As cells divide, they can shorten and no longer protect the chromosome or cell.

Lengthening them is the focus of recent research. Last year, a BGRF study was able to lengthen human telomeres.

A recent report in the journal Cell detailed how peptides were able to boost the life span of mice. The study examined how cell therapy could reverse poor age-related kidney function, fur loss, and frailty in mice.

Scientists are looking into whether or not the approach can also prolong the life span of mice. Human safety studies are in the works.

Reason said there are two schools of thought when exploring extending life through genetic pathways.

One approach is to alter cellular metabolism and make cells age more slowly, but the work is difficult and expensive. The other is to fix old tissue because we understand how it compares with young tissue.

No one yet fully understands everything these [older] cells do to us, but the fastest way to find out is to get rid of them, and we know that doing that in mice extends life and reverts aspects of aging, Reason explained.

Either you slow down the damage, or you repair the damage. Aging is damage. It is in the how of achieving one of those goals that all the complexity starts up, Reason added.

The Human Aging Genomic Resources (HAGR) website recently released DrugAge, a database of lifespan-extending drugs and compounds.

It includes 418 compounds that were recorded from studies on 27 different model organisms.

HAGR already operates the GenAge database of age and longevity-related genes in humans and model organisms. They also operate AnAge, which has aging and longevity records of more than 4,000 species, the GenDR database of genes associated with the life-extending effects of dietary restriction, and LongevityMap, which includes more than 2,000 human gene and genetic variations linked to longevity.

DrugAge incorporates earlier efforts by Biogerontology Research Foundation (BGRF) scientists, who produced Geroprotectors.org. Right now, its the largest database of its kind.

According to the research teams from BGRF and the University of Liverpool, pharmaceuticals have not targeted most age-related pathways. The research is only focused on a small number of pathways that are currently known.

The goal behind the database is to pave the way for discovery of new life span-extending and health span-extending compounds.

I am confident that it [DrugAge] will gain widespread use in the aging research community, and represents a significant milestone along the way to the coming paradigm shift in modern healthcare away from single disease treatment and toward geroprotective multi-disease prevention, Dmitry Kaminskiy, managing trustee of BGRF, said in a statement.

Franco Cortese, deputy director and trustee of BGRF, said in a statement the database will be extremely valuable for biogerontologists. The BGRF did not respond to Healthlines request for comment.

Already, researchers are using the data to identify trends and develop a better understanding of the comparative effects of geroprotectors on organisms.

Arrison is excited when she sees people teaming up globally to battle human disease and decline something that the DrugAge team is hoping to do with their database.

The more knowledge the health community can get, the better. The wonderful thing about the internet is that knowledge gets distributed faster, making the quest for cures that much quicker, Arrison added.

Anti-aging peptide recovers fur growth, kidney health in mice – Medical News Today

An anti-aging therapy could be one step closer; in a new study, researchers reveal how a peptide led to the destruction of cells that play a role in aging, reversing fur loss, kidney damage, and frailty in mice.

The research describes how the peptide stops levels of a protein called FOXO4 from increasing in senescent cells, which are cells that lose the ability to replicate and destroy themselves, but which remain metabolically active.

Senescent cells accumulate with age, and studies have shown that they can contribute to the aging process by causing damage to neighboring cells and impairing tissue function.

Previous research has shown that in senescent cells, levels of FOXO4 rise to prevent another protein called p53 from prompting the cells' self-destruction.

By blocking FOXO4 with the peptide, the research team has been able to restore programmed cell death, or apoptosis, in senescent cells.

"Only in senescent cells does this peptide cause cell death," says senior author Peter de Keizer, a researcher of aging at Erasmus University Medical Center in the Netherlands.

"FOXO4 is barely expressed in non-senescent cells, so that makes the peptide interesting, as the FOXO4-p53 interaction is especially relevant to those cells, but not normal cells."

On administering the peptide to fast-aging mice in regular doses, the researchers were able to reverse age-related conditions, such as fur loss and poor kidney health.

The findings were recently published in the journal Cell.

For their study, the researchers tested the peptide on older mice that had aged naturally and mice that had been genetically modified to age rapidly.

Both groups of mice developed characteristics and health problems commonly associated with aging, such as loss of fur, a decline in kidney health, and frailty.

Some of the rodents in each group were given infusions of the peptide three times a week for 10 months, while the remaining mice were monitored as controls.

Both the fast-aging and naturally aged mice saw improvements with peptide treatment, with no apparent side effects.

Within 10 days, the fast-aging mice began to experience fur regrowth. After 3 weeks, the naturally aged mice began to see improvements in fitness, compared with mice that did not receive the peptide.

Additionally, both the fast-aging and naturally aged mice started to demonstrate improvements in kidney function from 1 month after peptide treatment.

The team notes that the effects of peptide treatment were was so strong in fast-aging mice that doses needed to be reduced over the study period.

The researchers say that their findings support previous research showing that targeting senescent cells can help to reverse aging and increase lifespan, though much more research is warranted.

"The common thread I see for the future of anti-aging research is that there are three fronts in which we can improve: the prevention of cellular damage and senescence, safe therapeutic removal of senescent cells, to stimulate stem cells - no matter the strategy - to improve tissue regeneration once senescence is removed," says de Keizer.

He and his colleagues now plan to conduct a clinical trial to assess the safety of the peptide in humans.

Scientists unveil a giant leap for anti-aging – Science Daily

UNSW researchers have made a discovery that could lead to a revolutionary drug that actually reverses ageing, improves DNA repair and could even help NASA get its astronauts to Mars.

In a paper published in Science today, the team identifies a critical step in the molecular process that allows cells to repair damaged DNA.

Their experiments in mice suggest a treatment is possible for DNA damage from ageing and radiation. It is so promising it has attracted the attention of NASA, which believes the treatment can help its Mars mission.

While our cells have an innate capability to repair DNA damage -- which happens every time we go out into the sun, for example -- their ability to do this declines as we age.

The scientists identified that the metabolite NAD+, which is naturally present in every cell of our body, has a key role as a regulator in protein-to-protein interactions that control DNA repair.

Treating mice with a NAD+ precursor, or "booster," called NMN improved their cells' ability to repair DNA damage caused by radiation exposure or old age.

"The cells of the old mice were indistinguishable from the young mice, after just one week of treatment," said lead author Professor David Sinclair of UNSW School of Medical Sciences and Harvard Medical School Boston.

Human trials of NMN therapy will begin within six months.

"This is the closest we are to a safe and effective anti-ageing drug that's perhaps only three to five years away from being on the market if the trials go well," says Sinclair, who maintains a lab at UNSW in Sydney.

What it means for astronauts, childhood cancer survivors, and the rest of us

The work has excited NASA, which is considering the challenge of keeping its astronauts healthy during a four-year mission to Mars.

Even on short missions, astronauts experience accelerated ageing from cosmic radiation, suffering from muscle weakness, memory loss and other symptoms when they return. On a trip to Mars, the situation would be far worse: five per cent of the astronauts' cells would die and their chances of cancer would approach 100 per cent.

Professor Sinclair and his UNSW colleague Dr Lindsay Wu were winners in NASA's iTech competition in December last year.

"We came in with a solution for a biological problem and it won the competition out of 300 entries," Dr Wu says.

Cosmic radiation is not only an issue for astronauts. We're all exposed to it aboard aircraft, with a London-Singapore-Melbourne flight roughly equivalent in radiation to a chest x-ray.

In theory, the same treatment could mitigate any effects of DNA damage for frequent flyers. The other group that could benefit from this work is survivors of childhood cancers.

Dr Wu says 96 per cent of childhood cancer survivors suffer a chronic illness by age 45, including cardiovascular disease, Type 2 diabetes, Alzheimer's disease, and cancers unrelated to the original cancer.

"All of this adds up to the fact they have accelerated ageing, which is devastating," he says.

"It would be great to do something about that, and we believe we can with this molecule."

An anti-ageing pill could be on the horizon

For the past four years, Professor Sinclair and Dr Wu have been working on making NMN into a drug substance with their companies MetroBiotech NSW and MetroBiotech International.

The human trials will begin this year at Brigham and Women's Hospital, in Boston.

The findings on NAD+ and NMN add momentum to the exciting work the UNSW Laboratory for Ageing Research has done over the past four years.

They've been looking at the interplay of a number of proteins and molecules and their roles in the ageing process.

They had already established that NAD+ could be useful for treating various diseases of ageing, female infertility and also treating side effects of chemotherapy.

In 2003, Professor Sinclair made a link between the anti-ageing enzyme SIRT1 and resveratrol, a naturally occurring molecule found in tiny quantities in red wine.

"While resveratrol activates SIRT1 alone, NAD+ boosters activate all seven sirtuins, SIRT1-7, and should have an even greater impact on health and longevity," he says.

How Old Is Too Old to Work? – The New Yorker

In her book Elderhood: Redefining Aging, Transforming Medicine, Reimagining Life, Louise Aronson, a geriatrician and a professor of medicine at the University of California, San Francisco, examines Americans fraught relationship to the aging process, from our enthusiasm for supposed anti-aging cures to our troubled health-care system and the persistence of ageism among employers. Too often, Aronson argues, people over the age of sixty-five are viewed as a single groupone whose needs and desires and humanity are given short shrift, particularly within a medical system that is ostensibly centered around the needs of the elderly.

Questions about the place of the elderly in American society have become especially pertinent in our current political moment. On Inauguration Day, 2021, a septuagenarianDonald Trump, Joe Biden, or Bernie Sanderswill almost certainly be sworn into the office of the Presidency.

To discuss the perceived and real effects of aging, I recently spoke by phone with Aronson. During our conversation, which has been edited for length and clarity, we discussed why so many Americans over sixty-five are entering the workforce, whether the Presidency is the wrong job for someone over seventy, and why we tend to view older Americans as a single, distinct group.

Your book is very sympathetic to the realities of aging, as it should be. But what have you made of the fact that our next President is likely going to be seventy-four, seventy-eight, or seventy-nine when he is sworn into office? And how should we think about that as citizens?

What a lovely question. I guess I would see it as the most visible part of a national trend, which is that, for the last several years, by far the fastest-growing segments of the American workforce, and this comes from the Bureau of Labor Statistics, have been people age sixty-five and up. That includes two groups, and, actually, they parse it a bit, even though I complain in my book that we often dont parse it. One is ages sixty-five to seventy-four, and one is ages seventy-five and up. The relative increases are four to five times higher than the increases in workforce gains in people in their thirties, forties, and fifties. So its really this new phenomenon, which has come after a slight lag.

When we set Social Security and retirement in our sixties, the average age of death was about sixty-two. And now the average age at death is seventy-nine, with huge variation according to gender and race. The leading [Presidential] candidates are all white, which gives them a mortality advantage, as does the fact that theyre all educated. So people their age very routinely live into their eighties. And, actually, a man at age seventy-five has on average eleven to eleven and a half years of additional life, most likely, the vast majority of which are healthy.

To take a step back from the Presidency, how do you view this workplace trend?

I think its a good trend for a variety of reasons. One is that society simultaneously is quite ageist about, say, Isnt it time to retire?, or That persons over the hill, or not hiring, or all those hiring pull-down menus where they might not include a variety of birthdays of people who could be perfectly competent for the job. I speak to a lot of people in companies where theyre, like, But maybe they wont last very long. And they dont consider the youth equivalent, which is that most younger people now switch jobs really often.

Im digressing, but we cant have a society that simultaneously discriminates on the basis of age and doesnt hire or phases people out on the basis of age, and then complains about older people as a burden. You cant have it both ways. You cant say, Oh, youre not fiscally contributing, and then, Were not going to let you fiscally contribute. That is short of productiveeconomic and societal hypocrisy, basically.

One of the reasons I called the book Elderhood was because I really wanted to emphasize the fact that modern old agethe old age that evolved over the twentieth century, when we moved from an average lifespan of forty-nine to seventy-nineis colossal, that elderhood has become like childhood and adulthood. It is a phase of life that lasts decades and includes a huge diversity of people and functional abilities. So we easily accept that a neonate, infant, and toddler are children, just as teen-agers are children. And those are pretty different-looking animals. Similarly, we have a twenty-year-old in our household, and were in our mid-to-later fifties, and those are pretty different, too. You could argue a little less so, but, the further along I get, the more Im aware of the differences.

So how old are you? I know its a rude question, but, in this case

No, no. Fifty-six.

Im thirty-seven, just so were all honest and on the record.

I did search you, to get the sense of who I was talking to. I find I actually sometimes do get different questions depending on how old the person is.

So, while we all talk about our twenties or thirties as being so different from each other, youre saying the difference between sixties and seventies, or seventies and eighties, and eighties and nineties, are actually huge?

Exactly. A fascinating thing has happened in geriatrics just in the last decade, which is that we now sometimes take care of two generations. Ive had patients in their hundreds where their kids are in their seventies or eighties. And I routinely have people in their eighties and nineties who have kids in their sixties or seventies. So its really becoming multi-generational, because of the vast numbers of people living so many more years, and we need to acknowledge that those people are different. Different decades, no matter how old or young you are, are all very different, and thats part of lifes excitement. We keep moving forward.

Do you have some broad way of thinking about different ages or stages of elderhood?

I think one of the problems with old age is that we all begin young. So it all seems very old and foreign to us. There have been some subcategories. A psychologist, Bernice Neugarten, named the stages Young Old, Old, and Old Old, and that was roughly sixty-five to seventy-four, seventy-five to eighty-four, and then eighty-five up. We do have increasing numbers of centenarians, so that is a broad range. But one could argue that the sixty-five-to-seventy-four range, except for the most disadvantagedand there are all these structural things that contribute to that, like stress and povertyis a blurred line in medicine. Are you more like an adult? Are you more like an elder? And either can be true. And, as you get into seventies-to-eighties, there still is a range, but there are more people who are going to resemble the older people.

We got sidetracked, but can you say more about why you think it is good for older people to stay in the workforce?

One is that the economic burden goes down. The longer you work, the less likely you are to be the so-called burden. But perhaps more important is that health and quality of life go up when people keep working. I should also qualify that to use the word work broadly. We tend not to count traditionally female things, like caregiving, although were seeing increasing grandparents participating. But there are many ways in which you can contribute to your family or your community. People who have lots of wealth may be doing volunteer jobsthats still work. But we know unequivocally that people who keep working maintain higher levels of physical function and cognitive function. They just tend to be healthier. It probably relates to this growing database on purpose and health, and optimism and health. If youre doing something that feels like it has purpose or meaningalthough these are things we actually often intentionally strip old people ofthen you have a reason to get up.

Flower Pesticides Linked to Neurological Problems – Anti Aging News

Researchers from the University of California in San Diego and the University of Minnesota have discovered a link between neurological behaviors in Ecuadorian children and pesticide use during flower growing seasons. Their findings were published in the medical journal NeuroToxicology in May of 2017. Dr. Jose R. Suarez-Lopez Ph.D., an assistant professor at the University of San Diego in the Department of Family Medicine and Public Health, served as first author of the study.

Testing of Children in Agricultural Areas

Ecuador produces a large portion of the worlds cut flowers. The industry relies on pesticides to ensure successful harvests. Previous research using animals as test subjects suggested a link between temporary neurological disorders and pesticide exposure.

The research team tested 308 Ecuadorian children between the ages of 4 and 9 years of age who live near, but do not work in, flower producing areas. Behavioral and blood tests were conducted before peak production for the Mothers Day holiday, and within 100 days after harvest.

Results show that children tested shortly after the harvest ended displayed lower attention spans, less self-control, inferior hand-eye coordination, and problems perceiving and interacting with visual stimuli when compared to children tested later.

The authors of the study conclude that exposure to pesticides causes a short-term decline in neurological functions. According to Dr. Suarez-Lopez, this is of concern due to timing. The growing season corresponds with end-of-term testing in schools. Children suffering from temporary impairment due to pesticide exposure may receive lower scores on important academic exams. These lower scores may then affect their ability to qualify for certain opportunities in higher education or the job market.

The Effects of Agricultural Poisons

Newborns who are exposed to agricultural chemicals are known to have higher risks of certain neurological conditions, such as attention-deficit hyperactivity disorder (ADHD), and abnormalities in reflex, psychomotor, and mental function development. The effects are more prevalent in boys than girls. The short-term disorders detailed in the published study are in addition to these known, long-term effects.

The chemicals in question contain a substance called organophosphate, which is used to destroy pests on flowers before export. In the human body, organophosphates inhibit the actions of the enzyme acetylcholinesterase (AChE). AChE regulates acetylcholine, a neurotransmitter that allows nerves in the body and brain cells to communicate. Organophosphates are also known to destroy neurons and supporting brain cells. The study published by Dr. Suarez-Lopezs team is part of this larger effort to understand how environmental pollutants affect the long-term development and welfare of young people growing up in agricultural areas.

Researchers believe their findings prove that exposure to certain pesticides alters cognition, and the ability to learn and interact socially in children. Dr. Suarez-Lopez suggests that a better understanding can be achieved by repeating the tests with a larger sample group. In addition to conducting blood and behavioral tests before and after harvests, an additional test would be conducted during the harvest period, when exposure to agricultural pesticides peaks.

Why Anti-Aging Products Are the New Startup Frontier – Entrepreneur

The movement to look -- and feel -- younger continues to be big business, with an endless supply of customers. According to Transparency Market Research, the global anti-aging industry will be worth nearly $200 billion by 2019. The reasons for that growth are clear: All adults have insecurities surrounding aging and thus are potential customers.

Anti-aging technology changes so often that continuously furthering your education is a necessity.

While members of the younger generations will have some interest in your anti-aging product, baby boomers are your best investment source. People over 50 control 70 percent of the disposable income in the United States and are estimated to spend $52 billion globally each year.

For instance, anti-aging skin care is a robust market expected to eclipse $11 billion by 2018 in the United States alone. Like anything that touches on medical needs, the anti-aging industry comes with a slew of regulations. Because many consumers are still skeptical about anti-aging products, it's in everyone's best interest that his market not be flooded with ineffective or unsafe products.

Each year, the Food and Drug Administration sends warning letters to companies marketing as cosmetics anti-aging products that are really drugs. Intense Exercise Reduces Cellular Aging by Nine Years – Anti Aging News

Highly active adults have telomeres with a biological aging advantage of nine years over those who are sedentary.

Findings from a recent study show significant exercise is tied to nearly a decade of slower cell aging. Highly active individuals enjoy the benefits of a younger-feeling body. The study details were recently published in the popular medical journal Preventive Medicine.

The Aging Problem

Though scientists far and wide have been looking for a way to haltthe human aging process, none have succeeded. Sure, there are plenty of anti-aging creams on the market yet they merely improve the appearance and feel of skin.

Brigham Young University researchers may have pinpointed the best way to mitigate the aging process. Their research shows it might be possible to slow aging at the cellular level. However, slowing this type of aging will require plenty of hard work. Those who are willing to engage in demanding exercisecan slow their cellular aging level.

Though one's true age might be 60, it does not mean that hisbody is 60 years old in a biological sense. Consider individuals who seem much younger than their true age. These individuals are likely highly physically active. BYU researchers have determined such heightened levels of physical activity minimize the biological aging process within the body.

Study Details

The BYU research team determined individuals who engaged in demanding physical activity reaped the many benefits of elongated telomeres. Those who live comparably sedentary lifestyles have significantly shorter telomeres. It is interesting to note those who are moderately active have shorter telomeres than those who engage in high levels of exercise on a regular basis.

Telomeres are best described as theprotein endcaps on chromosomes within the human body. Telomeres are basically akin to the body's biological clock. They are strongly tied to age. Every time a cell within the human body replicates, a small portion of these endpoints is lost. As a result, the aging process gradually shrinks telomeres.

Data from nearly 6,000 adults who engaged in the CDC's National Health and Nutrition Examination Survey was analyzed for the study. This survey is revered as it is one of the few indexes that provides study subjects' telomere length. The index even includes information for 62 activities subjects may have participated in across 30 days. This data was also analyzed to accurately gauge subjects'levels of exercise.

Results Details

Adults who consistently engage in demanding exercises have telomeres with a biological aging boost of nine years versus those who live comparably idle lifestyles. Those who engage in high levels of physical activity enjoy a seven-year advantage over those who engage in a moderate level of physical activity. In order to be considered highly active, a woman must jog for half an hour five days per week. A man is considered to be highly active if he engages in 40 minutes of jogging five days per week. This means biological aging cannot be significantly slowed if one engages in minimal or even a moderate amount of exercise.

Sedentary individuals had the shortest telomeres, with 140 fewer base pairs at the endpoints of their telomeres than those who wereconsistently active. Yet there was no meaningful difference in telomere length between individuals with a low/moderate level of physical activity and those who were completely sedentary.

The Mystery of Lengthening Telomeres Through Exercise

The specific mechanism responsible for the preservation of telomeres resulting from exercise is unknown. Researchers believe the mechanism might be connected to a combination of inflammation and oxidative stress. Prior studies have determined telomere length is tied to these two factors. Furthermore, it is known that physical activity can suppress oxidative stress and inflammation as time progresses.

Peptide targeting senescent cells restores stamina, fur, and kidney function in old mice – Medical Xpress

Regular infusions of a peptide that can selectively seek out and destroy broken-down cells that hamper proper tissue renewal, called senescent cells, showed evidence of improving healthspan in naturally-aged mice and mice genetically engineered to rapidly age. The proof-of-concept study, published March 23 in Cell, found that an anti-senescent cell therapy could reverse age-related loss of fur, poor kidney function, and frailty. It is currently being tested whether the approach also extends lifespan, and human safety studies are being planned.

The peptide took over four years of trial and error to develop and builds on nearly a decade of research investigating vulnerabilities in senescent cells as a therapeutic option to combat some aspects of aging (Trends in Molecular Medicine, 10.1016/j.molmed.2016.11.006). It works by blocking the ability of a protein implicated in senescence, FOXO4, to tell another protein, p53, not to cause the cell to self-destruct. By interfering with the FOXO4-p53 crosstalk, the peptide causes senescent cells to go through apoptosis, or cell suicide.

"Only in senescent cells does this peptide cause cell death," says senior author Peter de Keizer, a researcher of aging at Erasmus University Medical Center in the Netherlands. "We treated mice for over 10 months, giving them infusions of the peptide three times a week, and we didn't see any obvious side effects. FOXO4 is barely expressed in non-senescent cells, so that makes the peptide interesting as the FOXO4-p53 interaction is especially relevant to senescent cells, but not normal cells."

Results appeared at different times over the course of treatment. Fast-aging mice with patches of missing fur began to recover their coats after 10 days. After about three weeks, fitness benefits began to show, with older mice running double the distance of their counterparts who did not receive the peptide. A month after treatment, aged mice showed an increase in markers indicating healthy kidney function.

Senescent cell therapy is one of several strategies being tested in mice aimed at reversing aging or lengthening healthspan. In 2015, the Valter Longo laboratory at the University of Southern California reported that mice on a calorie-restricted diet that mimics fasting benefited from a longer life, a reduction in inflammatory disease, and improved memory (Cell Metabolism, 10.1016/j.cmet.2015.05.012). And last December, Juan Carlos Izpisua Belmonte at the Salk Institute of Biological Science and colleagues made headlines with their discovery that cellular reprogramming of epigenetic marks could extend lifespan and improve health in fast-aging mice (Cell, 10.1016/j.cell.2016.11.052).

"This wave of research on how we can fight aging is complementary, and not in competition," says de Keizer. "The common thread I see for the future of anti-aging research is that there are three fronts in which we can improve: The prevention of cellular damage and senescence, safe therapeutic removal of senescent cells, to stimulate stem cellsno matter the strategyto improve tissue regeneration once senescence is removed."

de Keizer aims to start a company based on these findings, but in the short term, he and his group want to show that their peptide is non-toxic in humans with no unforeseen side effects. They plan to offer a safety clinical trial in people with Glioblastoma multiforme, an aggressive brain tumor, which also shows high levels of the biomarkers needed for this FOXO4 peptide to be effective.

AUSTRALIAN scientists have discovered a vitamin, nicotinamide mononucleotide (NMN), that helps cells repair DNA damage and its so good they are taking the pill themselves.

The cells of the old mice were indistinguishable from the young mice, after just one week of treatment, said Professor David Sinclair of UNSW School of Medical Sciences and Harvard Medical School Boston.

The study showed middle-aged mice given the vitamin lived 20 per cent longer and were able to run faster.

This is the closest we are to a safe and effective anti-ageing drug thats perhaps only three to five years away from being on the market if the trials go well, says joint researcher Dr Lindsay Wu from the University NSW.

Sydney businesswoman Kate Johnson, 34, runs a natural lip gloss company and said she is keen to try the anti-ageing product when it hits the market.

Im only in my early 30s myself but I would look at something like that in the future, she said.

I know certain facets of the market would think its not natural and say you should age gracefully and Im a huge believer in ageing gracefully, but at the same time you shouldnt apologise for doing something that makes you feel happy or healthy.

This sounds like a drug that will be revolutionary for women who are fickle about ageing and want to load up on drugs to fight it. And (it will be used by) other women who just want another product, just like cleansing their face in the morning.

Dr Wu, aged 33, is not old, but says he has been taking the vitamin pill for a year.

I am using it, Im not supposed to take it but I feel just fine, there are no side-effects, he said.

I take it out of intellectual curiosity.

In a paper published in Science today, the researchers identify a critical step in the molecular process that allows cells to repair damaged DNA.

Research by University NSW scientists Prof Sinclair and Dr Wu into the substance won NASAs iTech competition in December last year because of its potential use in the planned 2025 mission to Mars.

Accelerated ageing caused by cosmic radiation, mental impairment and increased risk of cancer are pitfalls of space travel.

On a trip to Mars five per cent of the astronauts cells will die affecting their mental and physical capacity.

Its hoped this vitamin might be able to reverse that damage says Dr Wu.

Back on earth the medicine promises to help every human defy the ageing process and stay healthy and it has potential to overcome the terrible side effects of cancer radiotherapy and chemotherapy.

Its also been found to treat Type 2 diabetes and restore vision following eye damage in animals.

A separate Japanese study showed mice given NMN gained less weight with ageing even when they consumed more food perhaps because their boosted metabolism used more energy.

However, he said the general population should not use it until it has been property tested in clinical trials.

And when clinical trials are complete it wont be on the market to treat ageing, instead it is likely to be used as a therapy to treat the side effects of cancer radiotherapy.

The big problem is that regulatory authorities dont recognise ageing as a disease even though you lose memory, you lose movement and it gives you cancer, he says.

To get approved by the US Food and Drug Administration and Australias Therapeutic Goods Administration NMN will have to be registered to treat a recognised disease like the side effects of radiation.

Things could be about to change though. A US trial is underway to test whether the diabetes drug metformin can extend human life and prevent cancer because of evidence it expands lifespan in animals.

The makers of Metformin will be the first to push the FDA to recognise ageing, change is in the air, says Dr Wu.

The discovery of the vitamin developed out of research into the famed life extending qualities of the molecule resveratrol found in red wine.

HOW IT WORKS

NMN works on the same biological pathway as resveratrol.

Every time we go out into the sun our DNA is damaged but our body repairs the damage, however as we age our bodys ability to do the repairs declines, Dr Wu explains.

A vitamin Nicotinamide adenine dinucleotide (NAD+) present in every cell boosts the bodys ability to repair cells but it declines by 60 per cent as we age or when we are exposed to radiation.

If you can boost NAD+ you can boost DNA repair, he says.

The research found that if you treated mice with a NAD+ precursor, or booster, called NMN it improved their cells ability to repair DNA damage caused by radiation exposure or old age.

Medical researchers are on a quest to develop a comprehensive understanding of the mechanism that causes the process in which cells become cancerous. This process is known as oncogenesis. A recent study keys in on the molecule generated amidst intense exercise known as lactate. The purpose of this research is to explain lactate's role in the creation of cancer cells. Lactate is the byproduct of glycolysis, a chemical process that breaks apart sugar into smaller molecular components with the ultimate aim of generating energy. Lactate builds up in the blood and tissues when one engages in intense physical activity. If enough lactate accumulates, it can cause diminished physical performance as well as muscle tightness.

A famous German scientist named Otto Warburg first noticed that cancer cells consume an abundance of glucose compared to regular cells. Known as the Warburg effect, this phenomenon is a reference to the fact that cancer cells proceed through more glycolysis and generate more lactate than regular cells. The Latest Research

New research on this subject matter has been spearheaded by the director of the Sports Performance Department and physiology laboratory at the University of Colorado-Boulder's Sports Medicine and Performance Center. The director, Inigo San Millan, was determined to figure out why the Warburg effect occurs. Cancer research has deviated from cell metabolism study to genetics since Warburg's heyday in the early 20th century. However, the latest research might shift the spotlight back to lactate's role in the context of cancer development. Lactate and Oncogenesis

San Millan's research team suggests that lactate is the sole metabolic compound necessary and involved in the nearly half-dozen stages that stem from carcinogenesis. Their study analyzed the role of lactate in the process through which new blood vessels develop within tumors. This medical term for this process is angiogenesis. The study also examined the role of lactate in immune escape. This term refers to the cancer cells' eluding of the human body's natural immune responses. It also delves into the role of lactate in cell migration, self-sufficient metabolism, and metastasis.

San Millan's paper details how lactate assists in the creation of an acidic microenvironment outside of the cancer cell during metastasis. This phenomenon triggers the spread of additional cancer cells. The study also explores the connection between genetic components and lactate. The research team hypothesized that three transcription factors (p53, cMYC and HIF-1) common in the majority of cancers catalyze the deregulation of lactate. Can Halting Lactate Stop Cancer?

The important role of lactate in cancer cell creation helps explain why people who exercise on a regular basis have a low risk of cancer development. Individuals who exercise enjoy a body that is trained to transfer lactate to an energy source for the body, preventing an excessive accumulation of the metabolic compound. The findings allow for speculation that an idle lifestyle combined with an excessive consumption of sugar might lead to an abundance of lactate that leads to cancer. These findings show that lactate is not only present when cancer develops but required for each step of its development. What's Next?

San Millan hopes to team up with the University of Colorado Hospital to analyze the effects of custom tailored physical fitness programs created for cancer patients. At the moment, San Millan is studying the nuances of breast cancer cell lines. His hope is that additional research will ameliorate the quest to develop drugs that prevent the accumulation of lactate. These drugs will likely help key in on monocarboxylate transporters that transmit lactate between the body's cells.
