Self regulation of normal LRRK2 by DFG-flip
This video demonstrates molecular dynamics simulation of the Parkinson #39;s disease linked kinase called LRRK2. A typical kinase undergoes a duty cycle switching between "on" (DFG-in) and "off" (DFG-out) states regulating its own activity. The G2019S mutant however does not and stays in a perpetual "on" state that eventually eventually leads to Parkinson #39;s disease.
